    Cynomolgus monkey's choroid reference database derived from hybrid deep learning optical coherence tomography segmentation.

    Cynomolgus monkeys exhibit human-like features, such as a fovea, so they are often used in non-clinical research. Nevertheless, little is known about the natural variation of the choroidal thickness in relation to origin and sex. A combination of deep learning and a deterministic computer vision algorithm was applied for automatic segmentation of foveolar optical coherence tomography images in cynomolgus monkeys. The main evaluation parameters were choroidal thickness and surface area directed from the deepest point on OCT images within the fovea, marked as the nulla with regard to sex and origin. Reference choroid landmarks were set underneath the nulla and at 500 ”m intervals laterally up to a distance of 2000 ”m nasally and temporally, complemented by a sub-analysis of the central bouquet of cones. 203 animals contributed 374 eyes for a reference choroid database. The overall average central choroidal thickness was 193 ”m with a coefficient of variation of 7.8%, and the overall mean surface area of the central bouquet temporally was 19,335 ”m2 and nasally was 19,283 ”m2. The choroidal thickness of the fovea appears relatively homogeneous between the sexes and the studied origins.     Die interorganisatorische Zusammenarbeit in Produktionsnetzwerken kann Herausforderungen durch eine hohe Marktdynamik, immer anspruchsvollere KundenbedĂŒrfnisse und steigenden Kostendruck entgegenwirken. Neben der klassischen vertikalen Verschiebung von KapazitĂ€ten in Richtung geeigneter Zulieferer, lassen sich FertigungskapazitĂ€ten auch durch eine horizontale Zusammenarbeit zwischen produzierenden Unternehmen handeln. Im Sinne der Sharing Economy bieten digitale Plattformen eine geeignete Infrastruktur zur VerknĂŒpfung und Koordination der Marktakteure eines Produktionsnetzwerks. So können Fertigungsunternehmen flexibel ProduktionsausfĂ€llen entgegenwirken und freie MaschinenkapazitĂ€ten auslasten. Eine wesentliche Voraussetzung fĂŒr den Erfolg solcher digitalen Plattformen fĂŒr Produktionsnetzwerke ist die Definition von Zielen, welche bisher in der Literatur nur unzureichend und nicht bezogen auf diese spezifische Plattformart untersucht wurden. In dieser Arbeit wird ein umfĂ€ngliches konzeptionelles Zielmodell fĂŒr diese spezifische Plattformart erstellt. Zu spezifischen Zielen digitaler Plattformen fĂŒr Produktionsnetzwerke zĂ€hlen neben wirtschaftlichen oder technischen Zielen beispielsweise auch produktionsbezogene Marktleistungsziele wie die GewĂ€hrleistung von ProduktionsflexibilitĂ€t. Aufbauend darauf wird gezeigt, wie das Design der beschriebenen Plattformen einen Einfluss auf die Erreichung bestimmter Ziele hat und wie spezielle Mechanismen zur Zielerreichung beitragen.Interorganizational collaboration in production networks can counteract challenges posed by high market dynamics, increasingly demanding customer requirements and rising cost pressure. In addition to the vertical shift of capacities towards suitable suppliers, manufacturing capacities can also be traded through horizontal cooperation between manufacturing companies. Following the concept of the sharing economy, digital platforms offer a suitable infrastructure for connecting and coordinating the market actors in a production network. This enables manufacturing companies to flexibly counteract production downtimes and to utilize unused machine capacity. An essential requirement for the success of such digital platforms for production networks is the specification of goals, which have so far been insufficiently investigated in literature and are not specific to this type of platform. In this paper, we develop a comprehensive conceptual goal model for this specific type of platform. Specific goals of digital platforms for production networks include, in addition to economic or technical goals, production-related market service goals such as ensuring production flexibility. Based on this, we demonstrate how the design of the described platforms has an impact on the achievement of certain goals and how special mechanisms contribute to the achievement of goals

    Macular thickness measurements of healthy, naĂŻve cynomolgus monkeys assessed with spectral-domain optical coherence tomography (SD-OCT).

    The purpose of this study was to measure central macular thickness in an unprecedented number of cynomolgus monkeys. Macular thickness was measured with Heidelberg spectral-domain OCT in 320 eyes of healthy and treatment-naĂŻve cynomolgus monkeys (80 males and 80 females). The macula was successfully measured in all 320 eyes. Macular thickness was not significantly different between the sexes. The mean central macular thickness was 244 ÎŒm (+/- 21 ÎŒm). Macular thicknesses in the quadrants were 327 +/-17 ÎŒm (temporal inner), 339 +/- 17 ÎŒm (inferior inner), 341 +/- 14 ÎŒm (superior inner), 341 +/-18 ÎŒm (nasal inner), and 299 +/- 20 ÎŒm (temporal outer), 320 +/- 16 ÎŒm (superior outer), 332 +/-23 ÎŒm (inferior outer), and 337 +/-18 ÎŒm (nasal outer). Highly significant differences between the nasal and temporal quadrants were detected. This study successfully demonstrated the feasibility of retinal thickness measurements in healthy cynomolgus monkeys. The present findings indicate that the macula is thicker in cynomolgus monkeys than in humans and provide important normative data for future studies

    Histamine-3 Receptor Inverse Agonists for the Treatment of Obesity: Validation of the Target and Identification of Novel Series

    Obesity is a major risk factor for the development of conditions such as hypertension, hyperglycemia, dyslipidemia, coronary artery disease and cancer. Several pieces of evidence, including data in primates, have demonstrated the beneficial effects of histamine-3 receptor (H 3 R) inverse agonists in the regulation of food intake and body weight. A pharmacophore model based on selected published H 3 R ligands and validated by previ- ous investigations, was used to identify the 5-oxy-2-carboxamide-indole core as a novel series of H 3 R inverse agonists. Extensive structure-activity relationship (SAR) investigations were rewarded by the identification of several compounds reversing ( R )- α -methyl-histamine-induced water intake increase and reducing food intake/ body weight in rodent models of obesity. Among those compounds, (4,4-difluoro-piperidin-1-yl)-[1-isopropyl-5- (1-isopropyl-piperidin-4-yloxy)-1 H -indol-2-yl]-methanone, selected as a lead compound, was exhibiting a prom- ising profile, including excellent pharmacokinetic properties, good in vitro safety profile and high efficacy in a chronic rodent model of obesity
