The date the article was first made available online will be carried over.The majority of adults in the UK and US are overweight or obese due to multiple factors including excess energy intake. Training people to inhibit simple motor responses (key presses) to high-energy density food pictures reduces intake in laboratory studies. We examined whether online response inhibition training reduced real-world food consumption and weight in a community sample of adults who were predominantly overweight or obese (N = 83). Participants were allocated in a randomised, double-blind design to receive four 10-minute sessions of either active or control go/no-go training in which either high-energy density snack foods (active) or non-food stimuli (control) were associated with no-go signals. Participants’ weight, energy intake (calculated from 24-hour food diaries), daily snacking frequency and subjective food evaluations were measured for one week pre- and post-intervention. Participants also provided self-reported weight and monthly snacking frequency at pre-intervention screening, and one month and six months after completing the study. Participants in the active relative to control condition showed significant weight loss, reductions in daily energy intake and a reduction in rated liking of high-energy density (no-go) foods from the pre- to post-intervention week. There were no changes in self-reported daily snacking frequency. At longer-term follow-up, the active group showed significant reductions in self-reported weight at six months, whilst both groups reported significantly less snacking at one- and six-months. Excellent rates of adherence (97%) and positive feedback about the training suggest that this intervention is acceptable and has the potential to improve public health by reducing energy intake and overweight.National Institute for Health and Research Exeter Clinical Research FacilityWellcome TrustEuropean Research Counci

    Understanding and optimising the transfection of lipopolyplexes formulated in saline: the effects of peptide and serum

    Lipopolyplexes (LPDs) are of considerable interest for use as gene delivery vehicles. Here LPDs have been prepared from cationic vesicles (composed of a 1 : 1 molar ratio of DOTMA with the neutral helper lipid, DOPE), singly branched cationic peptides and plasmid DNA. All peptides contained a linker sequence (cleaved by endosomal furin) attached to a targeting sequence selected to bind human airway epithelial cells and mediate gene delivery. The current study investigates the effects of novel Arg-containing cationic peptide sequences on the biophysical and transfection properties of LPDs. Mixed His/Arg cationic peptides were of particular interest, as these sequences have not been previously used in LPD formulations. Lengthening the number of cationic residues in a homopolymer from 6 to 12 in each branch reduced transfection using LPDs, most likely due to increased DNA compaction hindering the release of pDNA within the target cell. Furthermore, LPDs containing mixed Arg-containing peptides, particularly an alternating Arg/His sequence exhibited an increase in transfection, probably because of their optimal ability to complex and subsequently release pDNA. To confer stability in serum, LPDs were prepared in 0.12 M sodium chloride solution (as opposed to the more commonly used water) yielding multilamellar LPDs with very high levels of size reproducibility and DNA protection, especially when compared to the (unilamellar) LPDs formed in water. Significantly for the clinical applications of the LPDs, those prepared in the presence of sodium chloride retained high levels of transfection in the presence of media supplemented with fetal bovine serum.     Overview of the spectrometer optical fiber feed for the Habitable-zone Planet Finder

    The Habitable-zone Planet Finder (HPF) is a highly stabilized fiber fed precision radial velocity (RV) spectrograph working in the Near Infrared (NIR): 810 - 1280 nm . In this paper we present an overview of the preparation of the optical fibers for HPF. The entire fiber train from the telescope focus down to the cryostat is detailed. We also discuss the fiber polishing, splicing and its integration into the instrument using a fused silica puck. HPF was designed to be able to operate in two modes, High Resolution (HR- the only mode mode currently commissioned) and High Efficiency (HE).     The Texas Department of Transportation (TxDOT), conducted National Register of Historic Places (NRHP) eligibility testing of the Cornelio Alvarez Sr. site (41SR242) as part of the State Loop (SL) 195 project (Project) (CSJ: 3632-01-001) in Starr County, Texas. Subsequent to the field investigations, SWCA Environmental Consultants (SWCA) conducted artifact analysis, reporting, and curation preparation for the multi-component historic and prehistoric site. Investigations were conducted in compliance with Section 106 of the National Historic Preservation Act (54 United State Code 30601) and the Antiquities Code of Texas (9 Natural Resources Code). The investigations assessed the site’s eligibility for listing on the NRHP (36 Code of Federal Regulations 60.4) and for designation as a State Antiquities Landmark (SAL; 13 Texas Administrative Code 26.8, 26.12). Christopher W. Ringstaff served as Principal Investigator under Texas Antiquities Permit Number 7912. TxDOT conducted the field investigations were from February 20–24, 2017, and April 10–14, 2017. Site 41SR242 is primarily a Middle to Late Archaic site with lesser Late Prehistoric and perhaps earlier components. The open occupational site is located on an upland margin landform in a tributary valley a few miles from the Rio Grande. The investigations revealed material assemblages consisting of diffusely scattered burned rock, debitage, and lithic tools, which were predominantly recovered from a 30- to 50- cm-thick stratum of mixed artifacts. However, a few concentrations of artifacts were identified, and each location yielded isolated intact features. Formation and post-depositional processes are generally not conducive to preservation of intact archeological surfaces, patterns, or site structure. Although the overall site lacks integrity and potential data yield, isolated discrete behavioral loci are present. Therefore, site 41SR242 is recommended as eligible for the NRHP and as an SAL. This recommendation pertains to the portions of the site within the APE. The site extends beyond the APE, and the areas outside of the APE have not been evaluated

    Heavy Fermion Behavior, Crystalline Electric Field Effects, and Weak Ferromagnetism in SmOs_{4}Sb_{12}

    The filled skutterudite compound SmOs_{4}Sb_{12} was prepared in single crystal form and characterized. The SmOs_{4}Sb_{12} crystals have the LaFe_{4}P_{12}-type structure with lattice parameter a = 9.3085 Angstroms. Specific heat measurements indicate a large electronic specific heat coefficient of ~880 mJ/mol K^{2}, from which an enhanced effective mass m^{*} ~ 170 m_{e} is estimated. The specific heat data also suggest crystalline electric field (CEF) splitting of the Sm^{3+} J = 5/2 multiplet into a Gamma_{7} doublet ground state and a Gamma_{8} quartet excited state separated by 37 K. Electrical resistivity rho(T) measurements reveal a decrease in rho(T) below ~50 K that is consistent with CEF splitting of ~33 K between a Gamma_(7) doublet ground state and Gamma_{8} quartet excited state. Specific heat and magnetic susceptibility measurements display a possible weak ferromagnetic transition at ~2.6 K, which could be an intrinsic property of SmOs_4Sb_{12} or possibly due to an unknown impurity phase.Comment: 24 pages, 11 Postscript figures, to be published in Physical Review
