Between status and spiritual salvation: New data on the Portinari triptych and the circumstances of its commission
Die Monumentalität des Portinari-Triptychons in den Uffizien scheint in gewisser Hinsicht die Lebensweise seines Auftraggebers, Tommaso Portinari, zu spiegeln: Die Porträts des Florentiner Kaufmanns, seiner Frau und dreier seiner Kinder in den Seitenflügeln, die die Familie in burgundischer Hofmode zeigen, erscheinen in ihrer repräsentativen Funktion als besonders eindringlicher Ausdruck von Portinaris gesellschaftlicher Stellung, die er als Leiter der Medici-Filiale in Brügge inne hatte. Seit Aby Warburg die kulturhistorische Vorstellung vom italienischen Kaufmann in Brügge prägte, der nur auf Zeit in der Stadt blieb und der dadurch, dass er flandrische Kunst für seine Heimat in Auftrag gab, den Austausch der Künste zwischen Nord und Süd förderte, gilt die Persönlichkeit Portinaris als die ideale Verkörperung dieser Vorstellung und das Portinari-Retabel bis heute als ihr einprägsamstes Abbild. Quellen in den Brügger Archiven zeichnen jedoch eine ganz andere Sachlage: Tommaso Portinari versuchte, sich mit umfangreichen Stiftungsaktivitäten in Brügge einen eigenen Status in der dortigen Gesellschaft aufzubauen. Er förderte die Brügger Observanten, wie jüngst auch Diane Wolfthal in ihrem Aufsatz zeigte („Florentine Bankers, Flemish Friars, and the Patronage of the Portinari Altarpiece“, in: Cultural Exchange between the Low Countries and Italy (1400-1600), hg. v. Ingrid Alexander-Skipnes, Turnhout 2007, S. 1-17), und stiftete eine Familienkapelle in St Jacob, vor deren Altar er und seine Frau begraben werden sollten. Die Stifterporträts der religiösen Bildwerke, die Portinari bei Hans Memling und Hugo van der Goes in Auftrag gab, waren demnach nicht für die Florentiner Gesellschaft bestimmt, ihnen kam vielmehr eine zentrale Rolle bei Portinaris Bemühungen zu, seinen Status innerhalb der Brügger Gesellschaft zu festigen. Cell autonomous requirement of neurofibromin (Nf1) for postnatal muscle hypertrophic growth and metabolic homeostasis
Background
Neurofibromatosis type 1 (NF1) is a multi-organ disease caused by mutations in neurofibromin 1 (NF1). Amongst other features, NF1 patients frequently show reduced muscle mass and strength, impairing patients' mobility and increasing the risk of fall. The role of Nf1 in muscle and the cause for the NF1-associated myopathy are mostly unknown.
Methods
To dissect the function of Nf1in muscle, we created muscle-specific knockout mouse models for NF1, inactivatingNf1in the prenatal myogenic lineage either under the Lbx1 promoter or under the Myf5 promoter. Mice were analysed during prenatal and postnatal myogenesis and muscle growth.
Results
Nf1(Lbx1)and Nf1(Myf5)animals showed only mild defects in prenatal myogenesis. Nf1(Lbx1)animals were perinatally lethal, while Nf1(Myf5)animals survived only up to approximately 25 weeks. A comprehensive phenotypic characterization of Nf1(Myf5)animals showed decreased postnatal growth, reduced muscle size, and fast fibre atrophy. Proteome and transcriptome analyses of muscle tissue indicated decreased protein synthesis and increased proteasomal degradation, and decreased glycolytic and increased oxidative activity in muscle tissue. High-resolution respirometry confirmed enhanced oxidative metabolism in Nf1(Myf5)muscles, which was concomitant to a fibre type shift from type 2B to type 2A and type 1. Moreover, Nf1(Myf5)muscles showed hallmarks of decreased activation of mTORC1 and increased expression of atrogenes. Remarkably, loss of Nf1 promoted a robust activation of AMPK with a gene expression profile indicative of increased fatty acid catabolism. Additionally, we observed a strong induction of genes encoding catabolic cytokines in muscle Nf1(Myf5)animals, in line with a drastic reduction of white, but not brown adipose tissue.
Conclusions
Our results demonstrate a cell autonomous role for Nf1 in myogenic cells during postnatal muscle growth required for metabolic and proteostatic homeostasis. Furthermore, Nf1 deficiency in muscle drives cross-tissue communication and mobilization of lipid reserves
Case Report: Allogeneic Stem Cell Transplantation Following Induction With CPX-351 in Patients With Acute Myeloid Leukemia Is Feasible
Acute myeloid leukemia with myelodysplasia-related changes (AML-MRC) and
treatment-related acutemyeloid leukemia (tAML) after chemotherapy or radiation therapy
for other neoplasms are associated with poor outcomes. CPX-351, a dual-drug liposomal
encapsulation of daunorubicin and cytarabine, has been shown to improve outcomes
in AML-MRC and tAML compared with standard 7+3 regimens. Here we report the
cases of four consecutive patients with AML-MRC or tAML who received CPX-351
as outpatient induction therapy immediately followed by allogeneic hematopoietic stem
cell transplantation (allo-HSCT). Two patients received allo-HSCT in remission (one in
complete remission and one in partial remission) and two patients received allo-HSCT
in aplasia (one at 11 days and one at 52 days after the start of induction therapy with
CPX-351). With a median follow-up of 188 days after allo-HSCT, all but one patient are
alive and two are in remission. Further studies will help define and expand the role of
CPX-351 in the treatment of AML-MRC and tAML, especially in patients expected to
undergo allo-HSCT
Randomized Controlled Trial of Individualized Arousal-Biofeedback for children and adolescents with Disruptive Behavior Disorders (DBD)
Background: Disruptive behavior disorders (including conduct disorder (CD) and oppositional defiant disorder (ODD)) are common childhood and adolescent psychiatric conditions often linked to altered arousal. The recommended first-line treatment is multi-modal therapy and includes psychosocial and behavioral interventions. Their modest effect sizes along with clinically and biologically heterogeneous phenotypes, emphasize the need for innovative personalized treatment targeting impaired functions such as arousal dysregulation.
Methods: A total of 37 children aged 8-14 years diagnosed with ODD/CD were randomized to 20 sessions of individualized arousal biofeedback using skin conductance levels (SCL-BF) or active treatment as usual (TAU) including psychoeducation and cognitive-behavioral elements. The primary outcome was the change in parents´ ratings of aggressive behavior measured by the Modified Overt Aggression Scale. Secondary outcome measures were subscales from the Child Behavior Checklist, the Inventory of Callous-Unemotional traits and the Reactive-Proactive Aggression Questionnaire.
Results: The SCL-BF treatment was neither superior nor inferior to the active TAU. Both groups showed reduced aggression after treatment with small effects for the primary outcome and large effects for some secondary outcomes. Importantly, successful learning of SCL self-regulation was related to reduced aggression at post-assessment.
Conclusions: Individualized SCL-BF was not inferior to active TAU for any treatment outcome with improvements in aggression. Further, participants were on average able to self-regulate their SCL, and those who best learned self-regulation showed the highest clinical improvement, pointing to specificity of SCL-BF regulation for improving aggression. Further studies with larger samples and improved methods, for example by developing BF for mobile use in ecologically more valid settings are warranted
