Dual Band Electrodes in Generator-Collector Mode: Simultaneous Measurement of Two Species
A computational model for the simulation of a double band collector-generator
experiment is applied to the situation where two electrochemical reactions
occur concurrently. It is shown that chronoamperometric measurements can be
used to take advantage of differences in diffusion coefficients to measure the
concentrations of both electroactive species simultaneously, by measuring the
time at which the collection efficiency reaches a specific value. The
separation of the electrodes is shown to not affect the sensitivity of the
method (in terms of percentage changes in the measured time to reach the
specified collection efficiency), but wider gaps can provide a greater range of
(larger) absolute values of this characteristic time. It is also shown that
measuring the time taken to reach smaller collection efficiencies can allow for
the detection of smaller amounts of whichever species diffuses faster. The case
of a system containing both ascorbic acid and opamine in water is used to
exemplify the method, and it is shown that mole fractions of ascorbic acid
between 0.055 and 0.96 can, in principle, be accurately measured.Comment: 34 pages, 8 figure
Hypoglycemic Effect of Gongronema latifolia Extracts in Rats
Phytochemistry and hypoglycemic effect of the water and Ethanolic Extracts of the leaves of Gongronema latifolium were screened for. The extracts were Soxhlet extracted and phytochemically screened. Three tolerated doses were estimated from acute toxicity studies and orally administered to three groups of 10 rats per group, daily for 21 days. After treatments, rats of groups 1 to 3 were sacrificed and blood samples collected for glucose analysis and was compared to control group 4. Phytochemistry reveals that, both extracts of the plant contain predominant amount of Saponins and Polyphenols among other phytochemicals. Glucose analysis shows dose related reduction in the blood glucose levels compared to control. The polyphenols content of the plant extracts could cause the hypoglycemic effect observed, suggesting insulin-like activity which justifies the use of the plant leaf in the management of diabetic Mellitus in Nigeria herbal medicine. Flexible Lipid Bilayers in Implicit Solvent
A minimalist simulation model for lipid bilayers is presented. Each lipid is
represented by a flexible chain of beads in implicit solvent. The hydrophobic
effect is mimicked through an intermolecular pair potential localized at the
``water''/hydrocarbon tail interface. This potential guarantees realistic
interfacial tensions for lipids in a bilayer geometry. Lipids self assemble
into bilayer structures that display fluidity and elastic properties consistent
with experimental model membrane systems. Varying molecular flexibility allows
for tuning of elastic moduli and area/molecule over a range of values seen in

Influence of Crohnâs disease related polymorphisms in innate immune function on ileal microbiome
We have previously identified NOD2 genotype and inflammatory bowel diseases (IBD) phenotype, as associated with shifts in the ileal microbiome (âdysbiosisâ) in a patient cohort. Here we report an integrative analysis of an expanded number of Crohn's disease (CD) related genetic defects in innate immune function (NOD2, ATG16L1, IRGM, CARD9, XBP1, ORMDL3) and composition of the ileal microbiome by combining the initial patient cohort (Batch 1, 2005â2010, n = 165) with a second consecutive patient cohort (Batch 2, 2010â2012, n = 118). These combined patient cohorts were composed of three non-overlapping phenotypes: 1.) 106 ileal CD subjects undergoing initial ileocolic resection for diseased ileum, 2.) 88 IBD colitis subjects without ileal disease (predominantly ulcerative colitis but also Crohnâs colitis and indeterminate colitis, and 3.) 89 non-IBD subjects. Significant differences (FDR C. difficile infection, and NOD2 genotype on ileal dysbiosis in the expanded analysis. The relative abundance of the Proteobacteria phylum was positively associated with ileal CD and colitis phenotypes, but negatively associated with NOD2R genotype. Additional associations with ORMDL3 and XBP1 were detected at the phylum/subphylum level. IBD medications, such as immunomodulators and anti-TNFα agents, may have a beneficial effect on reversing dysbiosis associated with the IBD phenotype. Exploratory analysis comparing microbial composition of the disease unaffected region of the resected ileum between 27 ileal CD patients who subsequently developed endoscopic recurrence within 6â12 months versus 34 patients who did not, suggested that microbial biomarkers in the resected specimen helped stratify patients with respect to risk of post-surgical recurrence.</div
Interdigitated ring electrodes: Theory and experiment
The oxidation of potassium ferrocyanide, K_4Fe(CN)_6, in aqueous solution
under fully supported conditions is carried out at interdigitated band and ring
electrode arrays, and compared to theoretical models developed to simulate the
processes. Simulated data is found to fit well with experimental results using
literature values of diffusion coefficients for Fe(CN)_6^(4-) and
Fe(CN)_6^(3-). The theoretical models are used to compare responses from
interdigitated band and ring arrays, and the size of ring array required to
approximate the response to a linear band array is investigated. An equation is
developed for the radius of ring required for a pair of electrodes in a ring
array to give a result with 5% of a pair of electrodes in a band array. This
equation is found to be independent of the scan rate used over six orders of
magnitude
Parent-identified barriers to accessing exposure therapy: A qualitative study using process mapping
BackgroundYouth with anxiety and obsessiveâcompulsive disorder (OCD) rarely access exposure therapy, an evidence-based treatment. Known barriers include transportation, waitlists, and provider availability. Efforts to improve access to exposure require an understanding of the process that families take to find therapists, yet no prior studies have examined parentsâ perspectives of the steps involved.MethodsParents of children who have received exposure therapy for anxiety and/or OCD (Nâ=â23) were recruited from a hospital-based specialty anxiety clinic where the majority of their children previously received exposure. Recruitment was ongoing until thematic saturation was reached. Parents completed questionnaires and attended an online focus group during which they were asked to describe each step they tookâfrom recognizing their child needed treatment to beginning exposure. A process map was created and shown in real-time, edited for clarity, and emailed to parents for member checking. Authors analyzed process maps to identify common themes.ResultsSeveral themes emerged, as visually represented in a final process map. Participants identified a âsearch-outreachâ loop, in which they repeated the cycle of looking for therapists, contacting them, and being unable to schedule an appointment due to factors such as cost, waitlists, and travel time. Parents often did not know about exposure and reported feeling guilty about their lack of knowledge and inability to find a suitable provider. Parents reported frustration that medical providers did not often know about exposure and sometimes dismissed parentsâ concerns. Participants emphasized the difficulty of navigating the mental health system; many reported that it took years to find an exposure therapist, and that the search was sometimes stalled due to fluctuating symptoms.ConclusionA common thread among identified barriers was the amount of burden placed on parents to find treatment with limited support, and the resultant feelings of isolation and guilt. Findings point to several directions for future research, such as the development of parent support groups for navigating the mental health system; enhancing coordination of care between medical and mental health providers; and streamlining referral processes
Exercise and Omega-3 Polyunsaturated Fatty Acid Supplementation for the Treatment of Hepatic Steatosis in Hyperphagic OLETF Rats
Background and Aims. This study examined if exercise and omega-3 fatty acid (n3PUFA) supplementation is an effective treatment for hepatic steatosis in obese, hyperphagic Otsuka Long-Evans Tokushima Fatty (OLETF) rats. Methods. Male OLETF rats were divided into 4 groups (n=8/group): (1) remained sedentary (SED), (2) access to running wheels; (EX) (3) a diet supplemented with 3% of energy from fish oil (n3PUFA-SED); and (4) n3PUFA supplementation plus EX (n3PUFA+EX). The 8 week treatments began at 13 weeks, when hepatic steatosis is present in OLETF-SED rats. Results. EX alone lowered hepatic triglyceride (TAG) while, in contrast, n3PUFAs failed to lower hepatic TAG and blunted the ability of EX to decrease hepatic TAG levels in n3PUFAs+EX. Insulin sensitivity was improved in EX animals, to a lesser extent in n3PUFA+EX rats, and did not differ between n3PUFA-SED and SED rats. Only the EX group displayed higher complete hepatic fatty acid oxidation (FAO) to CO2 and carnitine palmitoyl transferase-1 activity. EX also lowered hepatic fatty acid synthase protein while both EX and n3PUFA+EX decreased stearoyl CoA desaturase-1 protein. Conclusions. Exercise lowers hepatic steatosis through increased complete hepatic FAO, insulin sensitivity, and reduced expression of de novo fatty acid synthesis proteins while n3PUFAs had no effect
Feedback-amplified electrochemical dual-plate boron-doped diamond microtrench detector for flow injection analysis
An electrochemical flow cell with a boronâdoped diamond dualâplate microtrench electrode has been developed and demonstrated for hydroquinone flow injection electroanalysis in phosphate buffer pH 7. Using the electrochemical generatorâcollector feedback detector improves the sensitivity by one order of magnitude (when compared to a single working electrode detector). The diffusion process is switched from an analyte consuming âexternalâ process to an analyte regenerating âinternalâ process with benefits in selectivity and sensitivity
Environmental drivers of variation in bleaching severity of Acropora species during an extreme thermal anomaly
High sea surface temperatures caused global coral bleaching during 2015â2016. During this thermal stress event, we quantified within- and among-species variability in bleaching severity for critical habitat-forming Acropora corals. The objective of this study was to understand the drivers of spatial and species-specific variation in the bleaching susceptibility of these corals, and to evaluate whether bleaching susceptibility under extreme thermal stress was consistent with that observed during less severe bleaching events. We surveyed and mapped Acropora corals at 10 sites (N = 596) around the Lizard Island group on the northern Great Barrier Reef. For each colony, bleaching severity was quantified using a new image analysis technique, and we assessed whether small-scale environmental variables (depth, microhabitat, competition intensity) and species traits (colony morphology, colony size, known symbiont clade association) explained variation in bleaching. Results showed that during severe thermal stress, bleaching of branching corals was linked to microhabitat features, and was more severe at reef edge compared with lagoonal sites. Bleaching severity worsened over a very short time-frame (âŒ1 week), but did not differ systematically with water depth, competition intensity, or colony size. At our study location, within- and among-species variation in bleaching severity was relatively low compared to the level of variation reported in the literature. More broadly, our results indicate that variability in bleaching susceptibility during extreme thermal stress is not consistent with that observed during previous bleaching events that have ranged in severity among globally dispersed sites, with fewer species escaping bleaching during severe thermal stress. In addition, shaded microhabitats can provide a refuge from bleaching which provides further evidence of the importance of topographic complexity for maintaining the biodiversity and ecosystem functioning of coral reefs
