    Agreement Between the Douleur Neuropathique in 4 Questions and Leeds Assessment of Neuropathic Symptoms and Signs Questionnaires to Classify Neuropathic Pain Among Patients with Leprosy.

    Neuropathic pain (NP) often occurs during the course of leprosy, and screening tools to differentiate NP from non-NP are often used. However, their performance varies in different settings. The most frequently used scales are the Douleur Neuropathique in 4 questions (DN4) and the Leeds assessment of neuropathic symptoms and signs (LANSS) questionnaires. Thus, we conducted a study to evaluate the agreement between DN4 and LANSS questionnaires to classify NP in 195 leprosy patients attending two reference centers in Sergipe, Brazil. The DN4 and LANSS classified 166 and 110 patients, respectively, as having NP. One hundred and seven (54.8%) were classified as NP by both questionnaires; 59 (30.2%) solely by the DN4 questionnaire and three (1.5%) solely by the LANSS. The agreement of the questionnaires was 66.2% (weak agreement, Kappa = 0.30). Although both questionnaires identified a high proportion of NP, the development of more robust instruments is necessary to ensure the accuracy of diagnosis of leprosy patients classified as having NP

    Synthesis of novel psoralen analogues and in vitro antitumor activity

    Psoralens are natural products present in several plant families that are extremely toxic to a wide variety of prokaryotic and eukaryotic organisms. They are potentially active in diseases such as vitiligo, psoriasis, and several types of cancer. Following our interest on this type of compounds 1 four new psoralen analogues were prepared, 1a-1c and 1e. To synthesize 1a (R = H) the method of Harayama and Ishii was used where the cinnamate was obtained by the Wittig reaction followed by ring closure. Condensation of 1-formyl-2-hydroxycarbazole with diethyl malonate gave 1b which by basic hydrolysis yielded compound 1c. Compound 1d was prepared before.2 Condensation of the 2-hydroxycarbazole with ethyl acetoacetate gave 1e. The products were characterized by elemental analysis, 1H and 13C NMR. Moreover, the anti-proliferative effect of compounds 1a-1e on human cancer cell lines (MDA-MB 231, HeLa and TCCSUP) was evaluated. Results suggest that these psoralen analogues possess a potent cytotoxic effect against the cell lines studied.     Novel benzopsoralen analogues : synthesis, biological activity and molecular docking studies

    New benzopsoralen analogues were synthesized and their inhibitory effect on the growth of tumourtumour cell lines (MDA MB231 and TCC-SUP) was evaluated. The in vitro antitumour activity of the new benzopsoralen analogues was discussed in terms of structure–activity relationship. Molecular docking studies with human-CYP2A6 enzymes were also carried out with the synthesized compounds to evaluate the potential of these molecules to interact with the haem group of the enzymes.     The orthoptera of Castro Verde special protection area (Southern Portugal): new data and conservation value

    With the increasing awareness of the need for Orthoptera conservation, greater efforts must be gathered to implement specific monitoring schemes. Despite recent surveys, little is known about Portuguese Orthoptera populations. This study was performed in 2014 and 2015 mainly in Castro Verde Special Protection Area (SPA), southern Portugal, and is the first Orthoptera inventory conducted in the area. A total of 35 Orthoptera species was recorded, with two new species reported for Portugal. We provide species’ habitat occurrences within the protected area and use information on the conservation status and the Iberian distribution of each documented species to discuss the importance of Castro Verde SPA for Orthoptera conservation.     A Fog and Blockchain Software Architecture for a Global Scale Vaccination Strategy

    Nowadays, there are many fragmented records of patient’s health data in different locations like hospitals, clinics, and organizations all around the world. With the arrival of the COVID-19 pandemic, several governments and institutions struggled to have satisfactory, fast, and accurate decision-making in a wide, dispersed, and global environment. In the current literature, we found that the most common related challenges include delay (network latency), software scalability, health data privacy, and global patient identification. We propose to design, implement and evaluate a healthcare software architecture focused on a global vaccination strategy, considering healthcare privacy issues, latency mitigation, support of scalability, and the use of a global identification. We have designed and implemented a prototype of a healthcare software called Fog-Care, evaluating performance metrics like latency, throughput and send rate of a hypothetical scenario where a global integrated vaccination campaign is adopted in wide dispensed locations (Brazil, USA, and United Kingdom), with an approach based on blockchain, unique identity, and fog computing technologies. The evaluation results demonstrate that the minimum latency spends less than 1 second to run, and the average of this metric grows in a linear progression, showing that a decentralized infrastructure integrating blockchain, global unique identification, and fog computing are feasible to make a scalable solution for a global vaccination campaign within other hospitals, clinics, and research institutions around the world and its data-sharing issues of privacy, and identification.N/

    New tetracyclic benzofurocoumarin (benzopsoralen) analogues were synthesized and their inhibitory effect on the growth of tumor cell lines was evaluated. The human tumor cell lines used were MDA MB231 (breast adenocarcinoma), HeLa (cervix adenocarcinoma) and TCC-SUP (bladder transitional cell carcinoma). The in vitro antitumor activity of the new benzopsoralens was discussed in terms of structure–activity relationship. Molecular docking studies with human-CYP2A6 enzymes were also carried out with the synthesized compounds in order to evaluate the potential of these compounds to interact with the heme group of the enzymes. The results have demonstrated that the linear compounds have the most pronounced activity against tumor cell lines and this might be related to the better accessibility that these compounds have to the active site in relation to the angular ones that have shown in the majority of the cases multiple binding poses in the active site of CYP2A6.To the Foundation for the Science and Technology (FCT, Portugal) for financial support to the NMR portuguese network (PTNMR, Bruker Avance III 400-Univ.     Activation of the Wnt/β-Catenin Signaling Pathway by Mechanical Ventilation Is Associated with Ventilator-Induced Pulmonary Fibrosis in Healthy Lungs

    BACKGROUND: Mechanical ventilation (MV) with high tidal volumes (V(T)) can cause or aggravate lung damage, so-called ventilator induced lung injury (VILI). The relationship between specific mechanical events in the lung and the cellular responses that result in VILI remains incomplete. Since activation of Wnt/β-catenin signaling has been suggested to be central to mechanisms of lung healing and fibrosis, we hypothesized that the Wnt/β-catenin signaling plays a role during VILI. METHODOLOGY/PRINCIPAL FINDINGS: Prospective, randomized, controlled animal study using adult, healthy, male Sprague-Dawley rats. Animals (n = 6/group) were randomized to spontaneous breathing or two strategies of MV for 4 hours: low tidal volume (V(T)) (6 mL/kg) or high V(T) (20 mL/kg). Histological evaluation of lung tissue, measurements of WNT5A, total β-catenin, non-phospho (Ser33/37/Thr41) β-catenin, matrix metalloproteinase-7 (MMP-7), cyclin D1, vascular endothelial growth factor (VEGF), and axis inhibition protein 2 (AXIN2) protein levels by Western blot, and WNT5A, non-phospho (Ser33/37/Thr41) β-catenin, MMP-7, and AXIN2 immunohistochemical localization in the lungs were analyzed. High-V(T) MV caused lung inflammation and perivascular edema with cellular infiltrates and collagen deposition. Protein levels of WNT5A, non-phospho (Ser33/37/Thr41) β-catenin, MMP-7, cyclin D1, VEGF, and AXIN2 in the lungs were increased in all ventilated animals although high-V(T) MV was associated with significantly higher levels of WNT5A, non-phospho (Ser33/37/Thr41) β-catenin, MMP-7, cyclin D1, VEGF, and AXIN2 levels. CONCLUSIONS/SIGNIFICANCE: Our findings demonstrate that the Wnt/β-catenin signaling pathway is modulated very early by MV in lungs without preexistent lung disease, suggesting that activation of this pathway could play an important role in both VILI and lung repair. Modulation of this pathway might represent a therapeutic option for prevention and/or management of VILI

    AbstractHydrothiolation of symmetrical and unsymmetrical buta-1,3-diynes with sodium organylthiolate anions in reflux, generated in situ by reacting C4H9SH with NaOH, afforded (Z)-organylthioenynes in low to good yields (25–80%). By using tetrabutylammonium hydroxide (TBAOH) as base instead of NaOH, the hydrothiolation of buta-1,3-diynes was more rapid and efficient, providing (Z)-organylthioenynes in good to excellent yields (70–95%)

    O uso de recursos naturais pelos artesãos da Aldeia Hippie de Arembepe, Bahia, Brasil

    The hippie village of Arembepe is one of the few still existing in the world. Currently this local community is composed by not more than 50 people (children and adults). They look for a new style of life isolated from the great urban centers and encircled by the nature. The art craft is the main economic activity. For the confection of the art craft the community uses the local and external natural local resources. In this study it was possible to identify the intense use of seeds (86.5%). Of the 23 plant resources used by craftsmen of the hippie village, six are locally found: Cocos nucifera L., Crotalaria retusa L., Canavalia rosea (Sw.) DC., Leucaena leucocephala (Lam.) de Wit, Coix lacrima-jobi L., and Canna indica L. The majority of the used seeds (75%) come from other regions of the country, gotten through purchase in specific stores and/or exchange with craftsmen fron other localities. The habit to collecting the seeds and of planting species that supply the necessary seeds for the subsistence of local art craft was observed among the craftsmen of Arembepe village.A aldeia hippie de Arembepe é uma das poucas ainda existentes no mundo. Atualmente, esta comunidade local é composta por cerca de 50 pessoas (crianças e adultos) que buscam outro estilo de vida, isolada dos grandes centros urbanos e rodeada pela natureza. O artesanato é a principal atividade econômica. Para a confecção do artesanato a comunidade usa os recursos naturais locais e externos. Neste estudo foi possível observar o uso intenso de sementes (86,5%). Dos 23 recursos vegetais utilizados pelos artesãos da aldeia hippie, seis estão presentes em Arembepe: Cocos nucifera L., Crotalaria retusa L., Canavalia rosea (Sw.) DC., Leucaena leucocephala (Lam.) de Wit, Coix lacrima-jobi L. e Canna indica L. Cerca de 75% das sementes usadas vem de outras regiões do país, obtidas por meio da compra em lojas específicas e/ou troca com artesãos de outras localidades. Foi constatado entre os artesãos residentes na aldeia hippie de Arembepe o hábito de coletar as sementes em áreas naturais e de plantar espécies que forneçam aquelas necessárias para a subsistência do artesanato local
