    A Framework to Evaluate Software Developer’s Productivity The VALORTIA Project

    Currently, there is a lack in companies developing software in relation to assessing their staff’s productivity before executing software projects, with the aim of improving effectiveness and efficiency. QuEF (Quality Evaluation Framework) is a framework that allows defining quality management tasks based on a model. The main purpose of this framework is twofold: improve an entity’s continuous quality, and given a context, decide between a set of entity’s instances on the most appropriate one. Thus, the aim of this paper is to make this framework available to evaluate productivity of professionals along software development and select the most appropriate experts to implement the suggested project. For this goal, Valortia platform, capable of carrying out this task by following the QuEF framework guidelines, is designed. Valortia is a platform to certify users' knowledge on a specific area and centralize all certification management in its model by means of providing protocols and methods for a suitable management, improving efficiency and effectiveness, reducing cost and ensuring continuous quality.Ministerio de Ciencia e Innovación TIN2013-46928-C3-3-

    Magneto-thermal evolution in the cores of adolescent neutron stars: The Grad-Shafranov equilibrium is never reached in the 'strong-coupling' regime

    At the high temperatures present inside recently formed neutron stars (T5×108KT\gtrsim 5\times 10^{8}\, \text{K}), the particles in their cores are in the "strong-coupling" regime, in which collisional forces make them behave as a single, stably stratified, and thus non-barotropic fluid. In this regime, axially symmetric hydromagnetic quasi-equilibrium states are possible, which are only constrained to have a vanishing azimuthal Lorentz force. In such equilibria, the particle species are not in chemical (β\beta) equilibrium, so β\beta decays (Urca reactions) tend to restore the chemical equilibrium, inducing fluid motions that change the magnetic field configuration. If the stars remained hot for a sufficiently long time, this evolution would eventually lead to a chemical equilibrium state, in which the fluid is barotropic and the magnetic field, if axially-symmetric, satisfies the non-linear Grad-Shafranov equation. In this work, we present a numerical scheme that decouples the magnetic and thermal evolution, enabling to efficiently perform, for the first time, long-term magneto-thermal simulations in this regime for different magnetic field strengths and geometries. Our results demonstrate that, even for magnetar-strength fields 1016G\gtrsim 10^{16} \, \mathrm{G}, the feedback from the magnetic evolution on the thermal evolution is negligible. Thus, as the core passively cools, the Urca reactions quickly become inefficient at restoring chemical equilibrium, so the magnetic field evolves very little, and the Grad-Shafranov equilibrium is not attained in this regime. Therefore, any substantial evolution of the core magnetic field must occur later, in the cooler "weak-coupling" regime (T5×108KT\lesssim 5\times 10^8 \, \mathrm{K}), in which Urca reactions are effectively frozen and ambipolar diffusion becomes relevant.Comment: 19 pages, 12 figures, submitted to MNRA

    Oligodendrogenesis in iron-deficient rats: Effect of apotransferrin

    In rats, iron deficiency produces an alteration in myelinformation. However, there is limited information on theeffects of this condition on oligodendroglial cell (OLGc)proliferation and maturation. In the present study, wefurther analyzed the hypomyelination associated withiron deficiency by studying the dynamics of oligodendrogenesis.Rats were fed control (40 mg Fe/kg) oriron-deficient (4 mg Fe/kg) diets from gestation day 5until postnatal day 3 (P3) or 11 (P11). OLGc proliferation,migration and differentiation were investigatedbefore and after an intracranial injection of apotransferrinat 3 days of age (P3). The proliferating cell populationwas evaluated at P3. Iron-deficient (ID) animalsshowed an increase in the oligodendrocyte precursorscell (OPC) population in comparison with controls. Theoverall pattern of migration of cells labeled with BrdUwas investigated at P11. Iron deficiency increased theamount of BrdU1 cells in the corpus callosum (CC) anddecreased OLGc maturation and myelin formation.Changes in nerve conduction were analyzed by measuringvisual evoked potentials. Latency and amplitudewere significantly disturbed in ID rats compared withcontrols. Both parameters were substantially normalizedwhen animals were treated with a single intracranialinjection of 350 ng apotransferrin (aTf). The currentresults give support to the idea that iron deficiencyincreases the number of proliferating and undifferentiatedcells in the CC compared with the control. Treatmentwith aTf almost completely reverted the effects ofiron deficiency, both changing the migration patternand increasing the number of mature cells in the CCand myelin formation.Fil: Rosato Siri, María Victoria.     Aplicación de estrategias de riego deficitario controlado en albaricoquero

    Se estudiaron los efectos de estrategias de riego deficitario controlado (RDC) sobre albaricoqueros de 9 años de edad (Prunus armeniaca L. cv. “Búlida”) injertados sobre patrón franco de ‘Real Fino’. Para ello, se establecieron 2 tratamientos de riego, uno de riego control, que se regó satisfaciendo los requerimientos hídricos del cultivo (100% ETc) y un tratamiento de RDC, que consistió en reducir los aportes de agua con respecto a la ETc en los períodos no críticos en este cultivo: a) 40% de la ETc desde floración hasta el final de la primera fase de crecimiento del fruto; b) 60% de la ETc durante la segunda fase de crecimiento del fruto y c) 50% y 25% durante la post‐cosecha final (60 días después de recolección), diferenciando 2 períodos de 30 días, el primero al 50% de la ETc, y otro hasta la caída de hojas al 25% de la ETc. Los resultados obtenidos indican que el albaricoquero es una especie adecuada para realizar estrategias de RDC, debido principalmente a la clara separación entre el crecimiento vegetativo y reproductivo y, también al efecto de crecimientos compensatorios que se producen en los frutos que han estado bajo RDC, lo cual hace que finalmente no se produzcan mermas significativas de calibre. Además, ciertas características cualitativas, como el nivel en sólidos solubles, sabor y coloración de la fruta, se ven incrementadas. Estas dos razones, junto con ahorros de agua vía riego, del 39%, conducen a que estrategias de RDC puedan suponer una clara alternativa en zonas con infradotación hídrica, como es el sureste español.Este estudio fue cofinanciado por tres proyectos: IRRIQUAL (EU‐FP6‐FOOD‐CT‐2006‐023120), CONSOLIDER INGENIO 2010 (MEC CSD2006‐0067) y SÉNECA (05665/PI/07) concedidos a los autores

    Characterization of an immunologically conserved epitope from hepatitis C virus E2 glycoprotein recognized by HLA-A2 restricted cytotoxic T lymphocytes

    BACKGROUND/AIMS: Identification of epitopes recognized by cytotoxic T lymphocytes (CTL) in hepatitis C virus (HCV) proteins is of importance because they can be used for vaccination, treatment of infection or monitoring of immune responses. Our purpose was to characterize new CTL epitopes in HCV structural proteins. METHODS: Peptides were synthesized and tested in HLA-A2 binding assays. Binder peptides were used to stimulate peripheral blood mononuclear cells from HCV+ patients and controls, and activity measured in chromium release and ELISPOT assays. RESULTS: Twenty binder peptides were found, and stimulation of HCV+ patient cells with nine peptides showing high binding ability led to the growth of CD8+ CTL recognizing peptide E2(614-622) in association with HLA-A2. Peptide E2(614-622) was recognized by 30% of HLA-A2+ patients with chronic HCV infection, but no responses were observed in control groups. Five peptides derived from region E2(614-622) from 26 different viral isolates bound to HLA-A2 molecules, and all of them but one, containing Phe at position 622, were recognized by E2(614-622) specific CTL. CONCLUSIONS: These results show that peptide E2(614-622) belongs to a highly conserved region of HCV E2, and might be a good candidate to induce anti-HCV CTL responses in HLA-A2+ subjects

    Medium-resolution Isaac Newton Telescope library of empirical spectra - II. The stellar atmospheric parameters

    We present a homogeneous set of stellar atmospheric parameters (T-eff, log g, [Fe/H]) for MILES, a new spectral stellar library covering the range lambda lambda 3525-7500 angstrom at 2.3 angstrom (FWHM) spectral resolution. The library consists of 985 stars spanning a large range in atmospheric parameters, from super-metal-rich, cool stars to hot, metal-poor stars. The spectral resolution, spectral type coverage and number of stars represent a substantial improvement over previous libraries used in population synthesis models. The atmospheric parameters that we present here are the result of a previous, extensive compilation from the literature. In order to construct a homogeneous data set of atmospheric parameters we have taken the sample of stars of Soubiran, Katz & Cayrel, which has very well determined fundamental parameters, as the standard reference system for our field stars, and have calibrated and bootstrapped the data from other papers against it. The atmospheric parameters for our cluster stars have also been revised and updated according to recent metallicity scales, colour-temperature relations and improved set of isochrones

    A Single Argonaute Gene Participates in Exogenous and Endogenous RNAi and Controls Cellular Functions in the Basal Fungus Mucor circinelloides

    The mechanism of RNAi is well described in metazoans where it plays a role in diverse cellular functions. However, although different classes of endogenous small RNAs (esRNAs) have been identified in fungi, their biological roles are poorly described due, in part, to the lack of phenotype of mutants affected in the biogenesis of these esRNAs. Argonaute proteins are one of the key components of the RNAi pathways, in which different members of this protein family participate in the biogenesis of a wide repertoire of esRNAs molecules. Here we identified three argonaute genes of the fungus Mucor circinelloides and investigated their participation in exogenous and endogenous RNAi. We found that only one of the ago genes, ago-1, is involved in RNAi during vegetative growth and is required for both transgene-induced RNA silencing and the accumulation of distinct classes of esRNAs derived from exons (ex-siRNAs). Classes I and II ex-siRNAs bind to Ago-1 to control mRNA accumulation of the target protein coding genes. Class III ex-siRNAs do not specifically bind to Ago-1, but requires this protein for their production, revealing the complexity of the biogenesis pathways of ex-siRNAs. We also show that ago-1 is involved in the response to environmental signals, since vegetative development and autolysis induced by nutritional stress are affected in ago-1(-) M. circinelloides mutants. Our results demonstrate that a single Ago protein participates in the production of different classes of esRNAs that are generated through different pathways. They also highlight the role of ex-siRNAs in the regulation of endogenous genes in fungi and expand the range of biological functions modulated by RNAi

    Multi-scale stamps for real-time classification of alert streams

    In recent years, automatic classifiers of image cutouts (also called "stamps") have shown to be key for fast supernova discovery. The upcoming Vera C. Rubin Observatory will distribute about ten million alerts with their respective stamps each night, which it is expected to enable the discovery of approximately one million supernovae each year. A growing source of confusion for these classifiers is the presence of satellite glints, sequences of point-like-sources produced by rotating satellites or debris. The currently planned Rubin stamps will have a size smaller than the typical separation between these point sources. Thus, a larger field of view image stamp could enable the automatic identification of these sources. However, the distribution of larger field of view stamps would be limited by network bandwidth restrictions. We evaluate the impact of using image stamps of different angular sizes and resolutions for the fast classification of events (AGNs, asteroids, bogus, satellites, SNe, and variable stars), using available data from the Zwicky Transient Facility survey. We compare four scenarios: three with the same number of pixels (small field of view with high resolution, large field of view with low resolution, and a proposed multi-scale strategy) and a scenario with the full ZTF stamp that has a larger field of view and higher resolution. Our multi-scale proposal outperforms all the scenarios, with a macro f1-score of 87.39. We encourage Rubin and its Science Collaborations to consider the benefits of implementing multi-scale stamps as a possible update to the alert specification.Comment: Submitted to ApJ

    The RNAi machinery controls distinct responses to environmental signals in the basal fungus Mucor circinelloides

    BACKGROUND: RNA interference (RNAi) is a conserved mechanism of genome defence that can also have a role in the regulation of endogenous functions through endogenous small RNAs (esRNAs). In fungi, knowledge of the functions regulated by esRNAs has been hampered by lack of clear phenotypes in most mutants affected in the RNAi machinery. Mutants of Mucor circinelloides affected in RNAi genes show defects in physiological and developmental processes, thus making Mucor an outstanding fungal model for studying endogenous functions regulated by RNAi. Some classes of Mucor esRNAs map to exons (ex-siRNAs) and regulate expression of the genes from which they derive. To have a broad picture of genes regulated by the silencing machinery during vegetative growth, we have sequenced and compared the mRNA profiles of mutants in the main RNAi genes by using RNA-seq. In addition, we have achieved a more complete phenotypic characterization of silencing mutants.  RESULTS: Deletion of any main RNAi gene provoked a deep impact in mRNA accumulation at exponential and stationary growth. Genes showing increased mRNA levels, as expected for direct ex-siRNAs targets, but also genes with decreased expression were detected, suggesting that, most probably, the initial ex-siRNA targets regulate the expression of other genes, which can be up- or down-regulated. Expression of 50% of the genes was dependent on more than one RNAi gene in agreement with the existence of several classes of ex-siRNAs produced by different combinations of RNAi proteins. These combinations of proteins have also been involved in the regulation of different cellular processes. Besides genes regulated by the canonical RNAi pathway, this analysis identified processes, such as growth at low pH and sexual interaction that are regulated by a dicer-independent non-canonical RNAi pathway.  CONCLUSION: This work shows that the RNAi pathways play a relevant role in the regulation of a significant number of endogenous genes in M. circinelloides during exponential and stationary growth phases and opens up an important avenue for in-depth study of genes involved in the regulation of physiological and developmental processes in this fungal model
