    Conservation biologists face many challenges in assessing health, immune status and infectious diseases in protected species. These challenges include unpredictable sample populations, diverse genetic and environmental backgrounds of the animals, as well as the practical, legal and ethical issues involved in experimentation. The use of whole genome scale transcriptomics with animal samples obtained in a minimally invasive manner is an approach that shows promise for health assessment. In this study we assessed the utility of a microarray to identify changes in gene expression predictive of health status by interrogating blood samples from California sea lions (Zalophus californianus) in rehabilitation. A custom microarray was developed from the commercially available dog microarray (Canis familiaris) by selecting probes that demonstrated reliable cross-hybridization with RNA in sea lion blood. This custom microarray was used for the analysis of RNA from 73 sea lion blood samples, from animals with a broad spectrum of health changes. Both traditional classifying techniques and newer artificial neural network approaches correctly classified sea lions with respect to health status, primarily distinguishing between leptospirosis infection and domoic acid exposure. Real time PCR validation for a small set of genes, followed by sequencing, showed good correlation with array results and high identity (96–98%) between the dog and sea lion sequences.     Hematology and serum chemistry in stranded and wildcaught harbor seals in central California:Reference intervals, predictors of survival, and parameters affecting blood variables

    Blood was collected from stranded harbor seal (Phoca vitulina) pups at admission (n=64) and release (n=45) from rehabilitation in 2007 and 2008 and from wild-caught harbor seal pups, subadults, and adults (n=110) in 2004, 2007, and 2008. Blood values measured at the time of admission were not predictive of survival during rehabilitation. Mass was associated with survival until release, and all pups that died weighed less than 10 kg at the time of admission. Dohle bodies were observed in leukocytes from 15% to 22% of the pups in rehabilitation, but not in the wild pups. Thresholds (95% confidence intervals) among wild pups were less than those in the released pups for leukocytes, neutrophils, total cholesterol, alanine aminotransferase (ALT), glucose, phosphorus, sodium, potassium, total protein, albumin, and globulin; thresholds were greater in wild pups than in released pups for hemoglobin (HGB), hematocrit (HCT), and glucose. Thresholds among released pups were less than those in wild pups for HGB, HCT, mean cell volume, chloride, and creatine kinase; thresholds among released pups were greater than those in wild pups for neutrophils, platelets, total cholesterol, triglycerides, ALT, aspartate aminotransferase, sorbitol dehydrogenase, bilirubin, phosphorus, potassium, total protein, and albumin. Age, girth, and geographic location affected the blood variables from wild-caught pups; age class, geographic location, sex, and body condition affected the blood variables of wild-caught, subadult and adult harbor seals.</p

    Predictors of reproductive cost in female Soay sheep

    1. We investigate factors influencing the trade-off between survival and reproduction in female Soay sheep (Ovis aries). Multistate capture-recapture models are used to incorporate the state-specific recapture probability and to investigate the influence of age and ecological conditions on the cost of reproduction, defined as the difference between survival of breeder and non-breeder ewes on a logistic scale. 2. The cost is identified as a quadratic function of age, being greatest for females breeding at 1 year of age and when more than 7 years old. Costs, however, were only present during severe environmental conditions (wet and stormy winters occurring when population density was high). 3. Winter severity and population size explain most of the variation in the probability of breeding for the first time at 1 year of life, but did not affect the subsequent breeding probability. 4. The presence of a cost of reproduction was confirmed by an experiment where a subset of females was prevented from breeding in their first year of life. 5. Our results suggest that breeding decisions are quality or condition dependent. We show that the interaction between age and time has a significant effect on variation around the phenotypic trade-off function: selection against weaker individuals born into cohorts that experience severe environmental conditions early in life can progressively eliminate low-quality phenotypes from these cohorts, generating population-level effects. © 2005 British Ecological Society.G.T. was supported by a BBRSC grant (ref. 96/E 14253)Peer Reviewe

    Parapoxviruses of seals and sea lions make up a distinct subclade within the genus Parapoxvirus

    AbstractPoxviruses of seals and sea lions have been tentatively identified as both orthopoxviruses and parapoxviruses, but their exact identity remained unconfirmed. Here, poxviral DNA sequences were generated from 39 clinical cases and compared to sequences from earlier poxvirus isolates from seals (Phocidae) and sea lions (Otariidae). Six genetically distinct poxvirus strains were detected, of which three were previously unrecognized. All detected strains were closely related to the parapoxviruses, confirming their classification as members of the genus Parapoxvirus. A phylogenetic analysis showed that pinniped parapoxviruses form a monophyletic group within the genus Parapoxvirus. Parapoxviruses from Atlantic pinnipeds were phylogenetically distant from those of Pacific pinnipeds. Parapoxviruses from phocids and otariids that inhabit the same geographical region were also phylogenetically distant, suggesting that parapoxviruses are not commonly transmitted between free-ranging phocids and otariids. However, one strain was detected in two otariid species, suggesting that pinniped parapoxviruses are capable of infecting multiple species within a phylogenetic family

    Effects of satellitelinked telemetry tags on humpback whales in the Gulf of Maine:Photographic assessment of tag sites

    Hundreds of large whales have been tracked using consolidated (Type-C) satellite tags, yet there have been few studies on their impacts on whale health. In 2011, we initiated the first study designed to evaluate the effects of these tags on a baleen whale. Between 2011 and 2018, we tagged 79 North Atlantic humpback whales (Megaptera novaeangliae) in the Gulf of Maine. We initially deployed commonly-used tags with an articulation between the anchor and transmitter (n = 35, 2011–12), before evidence of breakage prompted the development and use of more robust, integrated tags (n = 45). Tagged individuals were photographed immediately before, during and up to 11 years after tagging. They were re-encountered on an average of 41.3 days (SD = 44.3), yielding 2,971 photographed sightings through 2022. An objective scoring system was developed to characterise tag-site tissue responses based on photographs and to identify risk factors for prolonged healing. The initial tissue response to tagging was minimal, followed by skin loss around the tag, sometimes a degree of subcutaneous swelling, occasional extrusion of blubber, changes in skin colour, local depression formation around the implant site, tag loss and skin healing over the tag site, sometimes with a depression remaining. At last sighting, most non-integrated and integrated tag sites exhibited small, shallow skin depressions (58.8% and 66.7%, respectively). Some exhibited deeper depressions with differing adjacent skin coloration (26.5% and 15.6%, respectively) or barely detectable marks (11.8% and 15.6%, respectively). Mild subcutaneous swellings occasionally persisted at the tag site, but this was uncommon for both tag designs (2.9% and 2.2%, respectively). More severe tissue responses were associated with non-integrated tags and placements lower on the body. This study highlights the importance of using robust tag designs to minimise negative effects from Type-C tags. Furthermore, because tag placement was shown to affect outcome, precision equipment, experienced taggers and vessel operators are critical for optimal deployment.</p

    Evaluation of circulating eosinophil count and adrenal gland function in California sea lions naturally exposed to domoic acid

    Objective-To determine the effect of natural exposure to domoic acid (DA) on eosinophil counts and adrenal gland function in California sea lions (Zalophus californianus). Design-Cross-sectional prospective study. Animals-39 California sea lions. Procedures-Adult female sea lions admitted to a rehabilitation hospital during 2009 were classified into 1 of 3 groups (acute DA toxicosis, chronic DA toxicosis, or no DA exposure) on the basis of clinical signs, DA concentration in urine or feces, and hippocampal morphology. Endoparasite burden, eosinophil count, and serum cortisol and plasma ACTH concentrations were determined for each sea lion. For a subset of 8 sea lions, fecal glucocorticoid concentration after IM administration of cosyntropin was determined. Results-Sea lions exposed to DA (acute DA toxicosis, n = 11; chronic DA toxicosis, 19) had higher eosinophil counts and lower serum cortisol concentrations, compared with values for sea lions with no DA exposure (9). Eosinophil count was not associated with endoparasite burden. Serum cortisol concentration was associated with plasma ACTH concentrations in sea lions from the no DA exposure group but not in sea lions in the acute or chronic DA toxicosis groups. Following cosyntropin injection, fecal glucocorticoid concentrations increased in all sea lions evaluated except 1. Conclusions and Clinical Relevance-In adult sea lions, eosinophilia may be a cost-effective biomarker for DA exposure and may reflect alterations in hypothalamic, pituitary gland, or adrenal gland function. Domoic acid exposure may have subtle health effects on marine animals in addition to induction of neurologic signs.</p

    The symptomatology and diagnosis of domoic acid toxicosis in stranded California sea lions (Zalophus californianus) : a review and evaluation of 20 years of cases to guide prognosis

    Abstract: Introduction Domoic acid (DA) is a glutaminergic excitatory neurotoxin that causes the morbidity and mortality of California sea lions (Zalophus californianus; CSL) and other marine mammals due to a suite of effects mostly on the nervous and cardiac systems. Between 1998 and 2019, 11,737 live-stranded CSL were admitted to The Marine Mammal Center (TMMC; Sausalito, CA, USA), over 2,000 of which were intoxicated by DA. A plethora of clinical research has been performed over the past 20 years to characterize the range of toxic effects of DA exposure on CSLs, generating the largest dataset on the effects of natural exposure to this toxin in wildlife.Materials and methods In this study, we review published methods for diagnosing DA intoxication, clinical presentation, and treatment of DA-intoxicated CSL and present a practical, reproducible scoring system called the neuroscore (NS) to help assess whether a DA-affected CSL is fit for release to the wild following rehabilitation. Logistic regression models were used to assess the relationships between outcome (released vs. euthanized or died) and multiple variables to predict the outcome for a subset of 92 stranded CSLs.Results The largest proportion of DA-intoxicated CSLs was adult females (58.6%). The proportions of acute and chronic cases were 63.5 and 36.5% respectively, with 44% of affected CSL released and 56% either dying naturally or euthanized. The average time in rehabilitation was 15.9 days (range 0-169) for all outcomes. The best-performing model (85% accuracy; area under the curve = 0.90) assessing the relationship between outcome and predictor variables consisted of four variables: final NS, change in NS over time, whether the animal began eating in rehabilitation, and the state of nutrition on admission.Discussion Our results provide longitudinal information on the symptomatology of CSL intoxicated by domoic acid and suggest that a behavioral scoring system is a useful tool to assess the fitness for the release of DA-intoxicated CSL
