    Rhabdomyosarcoma of the Oral Tissues : two new cases and literature review

    Rhabdomyosarcoma (RMS) is a malignant soft tissue neoplasm consisting of cells derived from the primitive mesenchyme that exhibit a profound tendency to myogenesis. About 35% of RMS arises in the head and neck, being classified as parameningeal and non-orbital non-parameningeal forms. Parameningeal tumors carry the worst prognosis. The use of contemporary, multi-agent chemotherapy, radiotherapy, and surgery has made treatment of the disseminated disease possible, and has significantly improved overall survival from 25% in 1970 to 70% in 1991. Here, we present the management of two cases of orofacial RMS in adolescents: an 18-year-old, white female that had a 9-month history of a nodule in the left buccal mucosa, and a 19-year-old, white male who had been aware of a nodule in the left, posterior maxillary ridge with progressive growth for 4 months. Before final diagnosis, both cases were previously treated as inflammatory lesions. Their clinicopathological aspects, treatment, and poor survival as a consequence of delays in diagnosis are discussed

    Contact endoscopy (microstomatoscopy) in oral lesions: evaluation of the method

    OBJETIVO: Descrever as dificuldades da endoscopia de contato aplicada Ă  boca e ao lĂĄbio (microestomatoscopia de contato) e aplicar nas lesĂ”es do lĂĄbio os critĂ©rios usados na endoscopia de contato de outras topografias. MÉTODO: Estudo prospectivo utilizando o endoscĂłpio de contato Storz 8715A e o corante azul de toluidina. Vinte exames foram feitos para aprendizado da tĂ©cnica e depois mais 31 (21 lesĂ”es do lĂĄbio e 10 da boca), todos exames in vivo. Os critĂ©rios avaliados foram: Celularidade, Arquitetura, Estratificação e Morfologia. RESULTADOS: A falta de nitidez, os tremores finos e o deslizamento do aparelho dificultaram o procedimento em 100% dos casos, mas permitiram um exame adequado. Os 13 diagnĂłsticos de malignidade das lesĂ”es do lĂĄbio apresentaram todos os critĂ©rios alterados. CONCLUSÃO: as dificuldades encontradas foram o contato do aparelho com a superfĂ­cie anatĂŽmica, o deslizamento do aparelho, os tremores finos e a falta de nitidez da imagem, que nĂŁo inviabilizaram a realização e a interpretação do exame. Os critĂ©rios sĂŁo aplicĂĄveis Ă s lesĂ”es da boca e do lĂĄbio inferior.BACKGROUND: 1- To describe difficulties of contact endoscopy related to mouth and lip (contact microstomatoscopy) and 2- apply the criteria used in contact endoscopy of other topographies when lip lesions are diagnosed. METHODS: A prospective study was performed with the contact endoscope Storz 8715A and toluidine blue was used. Twenty examinations were accomplished in order to learn the technique; after that, other 31 were included (21 lip lesions and 10 oral lesions), all in vivo examinations. The criteria assessed were: cellularity, architecture, stratification and morphology. RESULTS: Lack of image clearness, slight tremors and sliding of the equipment made the procedure difficult in 100% of the cases; even though, accomplishment of proper examination was obtained. The 13 cases of lip malignancy lesions had the criteria altered as a whole. CoNCLUSION: Difficulties found were the contact of the equipment with anatomic surface, the sliding of the equipment, the slight tremors, and the lack of image clearness. Nevertheless, such elements did not interrupt the accomplishment and the examination analysis. The criteria are applied to oral and lower lip lesions

    In Vitro and In Vivo Biological Effects of Novel Arylimidamide Derivatives against Trypanosoma cruzi

    ABSTRACT Chagas disease (CD), a neglected tropical disease caused by Trypanosoma cruzi , remains a serious public health problem in several Latin American countries. The available chemotherapies for CD have limited efficacy and exhibit undesirable side effects. Aromatic diamidines and arylimidamides (AIAs) have shown broad-spectrum activity against intracellular parasites, including T. cruzi . Therefore, our aim was to evaluate the biological activity of eight novel AIAs (16DAP002, 16SAB079, 18SAB075, 23SMB022, 23SMB026, 23SMB054, 26SMB070, and 27SMB009) against experimental models of T. cruzi infection in vitro and in vivo . Our data show that none of the compounds induced a loss of cellular viability up to 32 ÎŒM. Two AIAs, 18SAB075 and 16DAP002, exhibited good in vitro activity against different parasite strains (Y and Tulahuen) and against the two relevant forms of the parasite for mammalian hosts. Due to the excellent selective indexes of 18SAB075, this AIA was moved to in vivo tests for acute toxicity and parasite efficacy; nontoxic doses (no-observed-adverse-effect level [NOAEL], 50 mg/kg) were employed in the tests for parasite efficacy. In experimental models of acute T. cruzi infection, 18SAB075 reduced parasitemia levels only up to 50% and led to 40% protection against mortality (at 5 mg/kg of body weight), being less effective than the reference drug, benznidazole

    In Vitro and In Vivo Investigation of the Efficacy of Arylimidamide DB1831 and Its Mesylated Salt Form - DB1965 - against Trypanosoma cruzi Infection

    Chagas disease is caused by infection with the intracellular protozoan parasite Trypanosoma cruzi. At present, nifurtimox and benznidazole, both compounds developed empirically over four decades ago, represent the chemotherapeutic arsenal for treating this highly neglected disease. However, both drugs present variable efficacy depending on the geographical area and the occurrence of natural resistance, and are poorly effective against the later chronic stage. As a part of a search for new therapeutic opportunities to treat chagasic patients, pre-clinical studies were performed to characterize the activity of a novel arylimidamide (AIA - DB1831 (hydrochloride salt) and DB1965 (mesylate salt)) against T.cruzi. These AIAs displayed a high trypanocidal effect in vitro against both relevant forms in mammalian hosts, exhibiting a high selectivity index and a very high efficacy (IC50 value/48 h of 5–40 nM) against intracellular parasites. DB1965 shows high activity in vivo in acute experimental models (mouse) of T.cruzi, showing a similar effect to benznidazole (Bz) when compared under a scheme of 10 daily consecutive doses with 12.5 mg/kg. Although no parasitological cure was observed after treating with 20 daily consecutive doses, a combined dosage of DB1965 (5 mg/kg) with Bz (50 mg/kg) resulted in parasitaemia clearance and 100% animal survival. In summary, our present data confirmed that aryimidamides represent promising new chemical entities against T.cruzi in therapeutic schemes using the AIA alone or in combination with other drugs, like benznidazole

    The biological in vitro effect and selectivity of aromatic dicationic compounds on Trypanosoma cruzi

    Trypanosoma cruzi is a parasite that causes Chagas disease, which affects millions of individuals in endemic areas of Latin America. One hundred years after the discovery of Chagas disease, it is still considered a neglected illness because the available drugs are unsatisfactory. Aromatic compounds represent an important class of DNA minor groove-binding ligands that exhibit potent antimicrobial activity. This study focused on the in vitro activity of 10 aromatic dicationic compounds against bloodstream trypomastigotes and intracellular forms of T. cruzi. Our data demonstrated that these compounds display trypanocidal effects against both forms of the parasite and that seven out of the 10 compounds presented higher anti-parasitic activity against intracellular parasites compared with the bloodstream forms. Additional assays to determine the potential toxicity to mammalian cells showed that the majority of the dicationic compounds did not considerably decrease cellular viability. Fluorescent microscopy analysis demonstrated that although all compounds were localised to a greater extent within the kinetoplast than the nucleus, no correlation could be found between compound activity and kDNA accumulation. The present results stimulate further investigations of this class of compounds for the rational design of new chemotherapeutic agents for Chagas disease

Based on a document produced according to the Millennium Goals, we sought to show the relevant points that infl uence the goal of “Achieving universal primary education for all”.Este artigo procura discutir a perda de vĂ­nculos familiares, escolares e sociais e suas implicaçÔes no processo educacional, principalmente no que diz respeito ao abandono e/ou nĂŁo continuidade da educação bĂĄsica. A escolha do tema decorreu de um estudo qualitativo atravĂ©s de um programa de informĂĄtica especĂ­fi co, o Atlas TI. A partir de um documento produzido com base nos Objetivos do MilĂȘnio, procuramos evidenciar os pontos relevantes que interferem na meta “Educação bĂĄsica de qualidade para todos”
