    Fatigue Risks in the Connections of Sign Support Structures

    This research effort develops a reliability-based approach for prescribing inspection intervals for mast-arm sign support structures corresponding to user-specified levels of fatigue-induced fracture risk. The resulting level of risk for a particular structure is dependent upon its geographical location, the type of connection it contains, the orientation of its mast-arm relative to north and the number of years it has been in service. The results of this research effort indicate that implementation of state-of-the-art reliability-based assessment procedures can contribute very valuable procedures for assigning inspection protocols (i.e. inspection intervals) that are based upon probabilities of finding fatigue-induced cracking in these structures. The engineering community can use the results of this research effort to design inspection intervals based upon risk and thereby better align inspection needs with limited fiscal and human resources

    On the Progenitor and Supernova of the SN 2002cx-like Supernova 2008ge

    We present observations of supernova (SN) 2008ge, which is spectroscopically similar to the peculiar SN 2002cx, and its pre-explosion site that indicate that its progenitor was probably a white dwarf. NGC 1527, the host galaxy of SN 2008ge, is an S0 galaxy with no evidence of star formation or massive stars. Astrometrically matching late-time imaging of SN 2008ge to pre-explosion HST imaging, we constrain the luminosity of the progenitor star. Since SN 2008ge has no indication of hydrogen or helium in its spectrum, its progenitor must have lost its outer layers before exploding, requiring that it be a white dwarf, a Wolf-Rayet star, or a lower-mass star in a binary system. Observations of the host galaxy show no signs of individual massive stars, star clusters, or H II regions at the SN position or anywhere else, making a Wolf-Rayet progenitor unlikely. Late-time spectroscopy of SN 2008ge show strong [Fe II] lines with large velocity widths compared to other members of this class at similar epochs. These previously unseen features indicate that a significant amount of the SN ejecta is Fe (presumably the result of radioactive decay of 56Ni generated in the SN), further supporting a thermonuclear explosion. Placing the observations of SN 2008ge in the context of observations of other objects in the class of SN, we suggest that the progenitor was most likely a white dwarf.Comment: 9 pages, 6 figures, accepted by A

    Description of Supported Employment Practices, Cross-System Partnerships, and Funding Models of Four Types of State Agencies and Community Rehabilitation Providers

    In 2005, the National Institute on Disability and Rehabilitation Research (NIDRR) awarded the VR Rehabilitation Research and Training Center (RRTC) to the Institute for Community Inclusion (ICI) at the University of Massachusetts Boston and its partners, InfoUse in Berkeley, California and the Center for the Advancement and Study of Disability Policy. In 2010, NIDRR provided supplemental funds to the ICI so that the VR-RRTC could include a focus on the provision of supported employment (SE) services. This SE research would focus on vocational rehabilitation (VR)agency partnerships with other state entities, and sources and models for long-term funding (extended services). The design called for embedding supported employment questions in ongoing surveys of multiple state agencies and case studies of SE coordination and funding models in several states to illuminate issues identified through these surveys. Accordingly, the ICI included an SE module into ongoing surveys of four state agencies known to deliver public employment services to people with disabilities. These surveys and the response rates achieved for each included: a) the state VR agency (89 percent); b) the state intellectual and developmental disability (IDD) agency (82 percent); c) the state mental health (MH)agency (55 percent); and d) the state welfare agency (47 percent).1 The SE supplement also included additional analysis of data obtained from an ongoing survey of community rehabilitation programs (CRPs) relevant to supported employment (37 percent response rate). The ICI then conducted case studies of SE partnerships in five states. These case studies were designed to help us better understand the range of practices that VR systems might use to ensure more successful transitions to long-term support through other resources. This report presents the findings from the SE supplement, the central focus of which was to identify the role and impact of VR agencies within the larger SE delivery system.     Global MYCN Transcription Factor Binding Analysis in Neuroblastoma Reveals Association with Distinct E-Box Motifs and Regions of DNA Hypermethylation

    BACKGROUND: Neuroblastoma, a cancer derived from precursor cells of the sympathetic nervous system, is a major cause of childhood cancer related deaths. The single most important prognostic indicator of poor clinical outcome in this disease is genomic amplification of MYCN, a member of a family of oncogenic transcription factors. METHODOLOGY: We applied MYCN chromatin immunoprecipitation to microarrays (ChIP-chip) using MYCN amplified/non-amplified cell lines as well as a conditional knockdown cell line to determine the distribution of MYCN binding sites within all annotated promoter regions. CONCLUSION: Assessment of E-box usage within consistently positive MYCN binding sites revealed a predominance for the CATGTG motif (p\u3c0.0016), with significant enrichment of additional motifs CATTTG, CATCTG, CAACTG in the MYCN amplified state. For cell lines over-expressing MYCN, gene ontology analysis revealed enrichment for the binding of MYCN at promoter regions of numerous molecular functional groups including DNA helicases and mRNA transcriptional regulation. In order to evaluate MYCN binding with respect to other genomic features, we determined the methylation status of all annotated CpG islands and promoter sequences using methylated DNA immunoprecipitation (MeDIP). The integration of MYCN ChIP-chip and MeDIP data revealed a highly significant positive correlation between MYCN binding and DNA hypermethylation. This association was also detected in regions of hemizygous loss, indicating that the observed association occurs on the same homologue. In summary, these findings suggest that MYCN binding occurs more commonly at CATGTG as opposed to the classic CACGTG E-box motif, and that disease associated over expression of MYCN leads to aberrant binding to additional weaker affinity E-box motifs in neuroblastoma. The co-localization of MYCN binding and DNA hypermethylation further supports the dual role of MYCN, namely that of a classical transcription factor affecting the activity of individual genes, and that of a mediator of global chromatin structure

    The Short-Term Effect of Prunes in Improving Bone in Men

    Osteoporosis is a major health concern in aging populations, where 54% of the U.S. population aged 50 and older have low bone mineral density (BMD). Increases in inflammation and oxidative stress play a major role in the development of osteoporosis. Men are at a greater risk of mortality due to osteoporosis-related fractures. Our earlier findings in rodent male and female models of osteoporosis, as well as postmenopausal women strongly suggest the efficacy of prunes (dried plum) in reducing inflammation and preventing/reversing bone loss. The objective of this study was to examine the effects of two doses of prunes, daily, on biomarkers of inflammation and bone metabolism in men with some degree of bone loss (BMD; t-score between −0.1 and −2.5 SD), for three months. Thirty-five men between the ages of 55 and 80 years were randomized into one of three groups: 100 g prunes, 50 g prunes, or control. Consumption of 100 g prunes led to a significant decrease in serum osteocalcin (p < 0.001). Consumption of 50 g prunes led to significant decreases in serum osteoprotegerin (OPG) (p = 0.003) and serum osteocalcin (p = 0.040), and an increase in the OPG:RANKL ratio (p = 0.041). Regular consumption of either 100 g or 50 g prunes for three months may positively affect bone turnover
