    Intrinsic Domain and Loop Dynamics Commensurate with Catalytic Turnover in an Induced-Fit Enzyme

    SummaryArginine kinase catalyzes reversible phosphoryl transfer between ATP and arginine, buffering cellular ATP concentrations. Structures of substrate-free and -bound enzyme have highlighted a range of conformational changes thought to occur during the catalytic cycle. Here, NMR is used to characterize the intrinsic backbone dynamics over multiple timescales. Relaxation dispersion indicates rigid-body motion of the N-terminal domain and flexible dynamics in the I182–G209 loop, both at millisecond rates commensurate with kcat, implying that either might be rate limiting upon catalysis. Lipari-Szabo analysis indicates backbone flexibility on the nanosecond timescale in the V308–V322 loop, while the rest of the enzyme is more rigid in this timescale. Thus, intrinsic dynamics are most prominent in regions that have been independently implicated in conformational changes. Substrate-free enzyme may sample an ensemble of different conformations, of which a subset is selected upon substrate binding, with critical active site residues appropriately configured for binding and catalysis

    We report on a comparative study that examines four conic luminance probes in their ability to measure small‐spot display contrast. We performed linear scans of a slit using a computer‐controlled stage.     Patient-focused Websites Related to Stress Urinary Incontinence and Pelvic Organ Prolapse: a DISCERN Quality Analysis

    Objective: To evaluate the quality of information presented on English-language websites regarding treatment options for POP and SUI using a validated instrument. Methods: Utilizing the International Urogynecology Association list of continence societies worldwide, faculty nomination and a complementary Google search using the terms “professional organizations stress urinary incontinence / pelvic organ prolapse” English-language, patient-focused websites related to POP and SUI were identified. The websites were evaluated by faculty at an academic medical center, including 4 faculty members of the urogynecology division, 2 urogynecology fellows, 3 urologists, and 1 obstetrician gynecologist. The websites were independently evaluated using the validated DISCERN instrument assessing the reliability and quality of consumer health information.Results: From the IUGA continence societies worldwide list, 47 websites were identified. Five provided patient information in English. Eight additional websites were identified from the Google search or provider nomination. One website provided information only for POP (rectocele), 5 for SUI, and 7 for both. The lowest mean total score for any website was 38.7 and the highest mean total score was 61.5 across all the websites. There were no statistically significant differences in the website mean total scores (p value \u3c0.40 for POP and 0.14 for SUI). For websites covering both topics, POP scores were generally higher than SUI scores, but not statistically significantly different (p value\u3c3.75). The overall quality item scores were also not significantly different (SUI: p\u3c0.923; POP: p \u3c0.813). Missing information most commonly included lack of clear objectives, sources, and information related to the expected outcome of no intervention.Conclusions: Available English-language professional websites written to inform patients about management choices for SUI and POP miss key components of quality patient information

    Removing subordinate species in a biodiversity experiment to mimic observational field studies

    Background: Positive effects of plant species richness on community biomass in biodiversity experiments are often stronger than those from observational field studies. This may be because experiments are initiated with randomly assembled species compositions whereas field communities have experienced filtering. Methods: We compared aboveground biomass production of randomly assembled communities of 2–16 species (controls) with experimentally filtered communities from which subordinate species were removed, resulting in removal communities of 1–8 species. Results: Removal communities had (1) 12.6% higher biomass than control communities from which they were derived, that is, with double species richness and (2) 32.0% higher biomass than control communities of equal richness. These differences were maintained along the richness gradient. The increased productivity of removal communities was paralleled by increased species evenness and complementarity. Conclusions: Result (1) indicates that subordinate species can reduce community biomass production, suggesting a possible explanation for why the most diverse field communities sometimes do not have the highest productivity. Result (2) suggests that if a community of S species has been derived by filtering from a pool of 2S randomly chosen species it is more productive than a community derived from a pool of S randomly chosen species without filtering

    Acclimation, Adaptation, Traits and Trade-Offs in Plankton Functional Type Models: Reconciling Terminology for Biology and Modelling

    We propose definitions in terminology to enhance ongoing collaborations between biologists and modellers on plankton ecology. Organism functional type should refer to commonality in ecology not biogeochemistry; the latter is largely an emergent property of the former, while alignment with ecology is also consistent with usage in terrestrial science. Adaptation should be confined, as in genetics, to consideration of species inter-generational change; most so-called adaptive plankton models are thus acclimative, modifying vital rates in response to stimuli. Trait trade-off approaches should ideally only be considered for describing intra-generational interactions; in applications between generations, and certainly between unrelated species, such concepts should be avoided. We suggest that systems biology approaches, through to complex adaptive/acclimative systems modelling, with explicit modelling of feedback processes (which we suggest should define mechanistic models), would provide realistic and flexible bases upon which to develop descriptions of functional type models

    A fast radio burst with a low dispersion measure

    Fast radio bursts (FRBs) are millisecond pulses of radio emission of seemingly extragalactic origin. More than 50 FRBs have now been detected, with only one seen to repeat. Here we present a new FRB discovery, FRB 110214, which was detected in the high latitude portion of the High Time Resolution Universe South survey at the Parkes telescope. FRB 110214 has one of the lowest dispersion measures of any known FRB (DM = 168.9±\pm0.5 pc cm3^{-3}), and was detected in two beams of the Parkes multi-beam receiver. A triangulation of the burst origin on the sky identified three possible regions in the beam pattern where it may have originated, all in sidelobes of the primary detection beam. Depending on the true location of the burst the intrinsic fluence is estimated to fall in the range of 50 -- 2000 Jy ms, making FRB 110214 one of the highest-fluence FRBs detected with the Parkes telescope. No repeating pulses were seen in almost 100 hours of follow-up observations with the Parkes telescope down to a limiting fluence of 0.3 Jy ms for a 2-ms pulse. Similar low-DM, ultra-bright FRBs may be detected in telescope sidelobes in the future, making careful modeling of multi-beam instrument beam patterns of utmost importance for upcoming FRB surveys.Comment: 8 pages, 3 figures, accepted for publication in MNRA

    A comprehensive comparison of the Sun to other stars: searching for self-selection effects

    If the origin of life and the evolution of observers on a planet is favoured by atypical properties of a planet's host star, we would expect our Sun to be atypical with respect to such properties. The Sun has been described by previous studies as both typical and atypical. In an effort to reduce this ambiguity and quantify how typical the Sun is, we identify eleven maximally-independent properties that have plausible correlations with habitability, and that have been observed by, or can be derived from, sufficiently large, currently available and representative stellar surveys. By comparing solar values for the eleven properties, to the resultant stellar distributions, we make the most comprehensive comparison of the Sun to other stars. The two most atypical properties of the Sun are its mass and orbit. The Sun is more massive than 95 -/+ 2% of nearby stars and its orbit around the Galaxy is less eccentric than 93 +/- 1% of FGK stars within 40 parsecs. Despite these apparently atypical properties, a chi^2 -analysis of the Sun's values for eleven properties, taken together, yields a solar chi^2 = 8.39 +/- 0.96. If a star is chosen at random, the probability that it will have a lower value (be more typical) than the Sun, with respect to the eleven properties analysed here, is only 29 +/- 11%. These values quantify, and are consistent with, the idea that the Sun is a typical star. If we have sampled all reasonable properties associated with habitability, our result suggests that there are no special requirements for a star to host a planet with life.Comment: Published in the Astrophysical Journal, 684:691-706, 2008 September 1.     Dietary fat intakes in Irish adults in 2011: how much has changed in 10 years?

    Imbalances in dietary fat intakes are linked to several chronic diseases. This study describes dietary intakes and food sources of fat and fatty acids in 1051 Irish adults (aged 18–90 years), using data from the 2011 national food consumption survey, the National Adult Nutrition Survey. It also compares current intakes for 18–64-year-olds with those reported in the last such survey in 2001, the North/South Ireland Food Consumption Survey. Dietary fat intakes were estimated using data from 4-d semi-weighed (2011) and 7-d estimated (2001) food diaries. In 2011, intakes for 18–64-year-olds were as follows: total fat, 34·1 (sd 6·1) % total energy (%TE); SFA, 13·3 (sd 3·3) %TE; MUFA, 12·5 (sd 2·6) %TE; PUFA, 6·1 (sd 2·2) %TE; and trans-fat, 0·511 (sd 0·282) %TE. Apart from MUFA, intakes decreased (P65 years had the highest intakes of SFA; however, intakes were typically higher than UK-recommended values for all groups. In contrast, intakes of long-chain n-3 fatty acids were lowest in younger age groups. Intakes of trans-fat were well within UK-recommended levels. Although there have been some improvements in the profile of intakes since 2001, imbalances persist in the quantity and quality of dietary fat consumed by Irish adults, most notably for total and SFA and for younger age groups for long-chain n-3 fatty acids
