Background: Healthcare workers (HCW) are at increased risk of SARS-CoV-2 infection. Here, we describe the SARS-CoV-2 seroprevalence in HCW who work daily at a COVID-19 front-line hospital in Portugal. Methods: To this end, the seroprevalence of 1027 HCW, assessed after the peak of the first pandemic wave, was determined using the following immunoassays: Euroimmun Anti-SARS-CoV-2 ELISA IgG (Euroimmun, Luebeck, Germany), Abbott SARS-CoV-2 IgG (Abbott Laboratories, Chicago), and Elecsys Anti-SARS-CoV–2 Total (Roche Diagnostics, Basel, Switzerland). Results: We found a 2.7% seroprevalence, very close to the one determined in the community (2.9%) for the same period. Conclusions: This low SARS-CoV–2 seroprevalence highlights the effectiveness of infection prevention and control measures implemented very early in the pandemic, namely the use of appropriate personal protective equipment. This research was supported by a grant from FCT Research4-COVID–19 (Project n° 186_596855206 – CertCOV)

AbstractReinforcing high performance ceramic particulates is an effective approach to improve solderability of lead-free Sn-0.7Cu solder. Various weight percentage compositions (0.5, 1.0, and 1.5) of Silicon Nitride (Si3N4) reinforced in Sn-0.7Cu solder were developed using powder metallurgy (PM) routes to investigate their solderability properties on copper (Cu) substrate. The solderability performances of the new composite solder will be determined and analyzed based on their contact angles on Cu substrate, including interface intermetallic compound (IMC) layer thickness and IMC phases formed for different Si3N4 ratios. Results also show an improvement in solderability of the Sn-0.7Cu/Si3N4 composite lead-free solder with optimum wettability achieved by 1.0wt.% Si3N4. The minimal average decrease in IMC layer thickness and the formation of the different shaped of scallops figuring the IMC layer were observed. X-ray diffraction (XRD) also revealed the decreasing peak intensity of Cu6Sn5 phases with Si3N4. Overall, the entire range of composition of Si3N4 into Sn-0.7Cu monolithic solder use in this study indicated an enhancement of solderability performances on Cu-substrates

Learning physical education has a different approach from other subjects. This is due to the emphasis on the development of psychomotor aspects rather than cognitive and affective aspects. In fact, the process of learning physical education emphasizes not only psychomotor aspects but also cognitive and affective aspects simultaneously, as internationally standardized by ICHPER-SD and UNESCO that physical education and sports in schools must describe the discipline of knowledge, skills and behavior covering the domains psychomotor, cognitive, and affective. This demand results in the need for a learning approach that can involve and develop all aspects of education. The Tactics Learning Approach (TGfU) pays very broad attention to the cognitive and psychomotor domains. For example, the cognitive domain is obtained from exams in high and secondary schools. Behavioral measurement is carried out to measure the effectiveness and involvement of the game that was developed to contribute to the measurement of actions and skills that are measured in relation to the cognitive domain. Very little attention has been paid to researchers and educators in the cognitive and affective domains. The influence of work and pleasure in sports will give greater influence through game learning. For example, children, coaches and parents all need to know that games and game situations are more enjoyable than continuous, technique-oriented practice

Background: Polycystic ovary syndrome (PCOS) is a common endocrine disorder associated with insulin resistance, even in the absence of overweight/obesity. The aim of the present study is to examine the global serum proteomic profile of adolescent, normal‐weight females with PCOS in order to gain novel insight in the association of this endocrine disorder with insulin physiology and to identify novel circulating markers that can guide intervention protocols. Methods: Non‐depleted serum from normal‐weight (BMI: 18–23 kg m^(−2)), adolescent females (13–21 years old) with PCOS (n = 20) is compared to BMI‐ and age‐matched healthy controls (n = 20) using our 3D quantitative proteomics methodology. Serum samples from study participants are randomly pooled to form four biological replicates of females with PCOS and four of healthy controls (n = 5 per sample pool). Results: One‐hundred and twenty‐six proteins are differentially expressed in females with PCOS compared to controls. Gene ontology analysis shows significant enrichment for terms related to inflammatory immune response, metabolism and insulin‐like growth factor receptor signaling pathway. Circulating levels of IGF‐1 and ‐2 and IGFBP‐2, ‐3, and ‐4 are found to be lower in females with PCOS compared to healthy controls. Conclusions: The present serum proteomics study provides insight into the pro‐inflammatory status and insulin dysregulation in young females with PCOS and identifies potential serological markers that can guide early intervention protocols

Assessment of Molecular Measures in Non-FXTAS Male Premutation Carriers

Approximately 30–40% of male and 8–16% of female carriers of the Fragile X premutation will develop a neurodegenerative movement disorder characterized by intentional tremor, gait ataxia, autonomic dysfunction, cognitive decline, and Parkinsonism during their lifetime. At the molecular level, premutation carriers have increased expression levels of the FMR1 and the antisense FMR1 (ASFMR1) mRNAs. Both genes undergo alternative splicing giving rise to a number of different transcripts. Alteration in the alternative splicing process might be associated with FXTAS. In this study, we have investigated the correlation between objective measures of movement (balance and tremor using the CATSYS battery) and the expression of both the FMR1 and the ASFMR1 genes. In addition, we investigated whether their expression level and that of the ASFMR1 131 bp splice isoform could distinguish between premutation carriers with FXTAS and non-FXTAS premutation carriers. Confirming previous findings, the expression levels of transcripts at the FMR1 locus positively correlated with the CGG repeat number and significantly differentiated the premutation carriers from the control groups. Furthermore, premutation carriers with and without FXTAS, showed a significant difference in the expression level of the ASFMR1 131 bp splice isoform when compared to age and gender matched controls. However, there was no significant difference in the ASFMR1 131 bp splice isoform expression level when comparing premutation carriers with and without FXTAS. Finally, our results indicate significant group differences in CATSYS dominant hand reaction time and postural sway with eyes closed in premutation carriers without FXTAS compared to controls. In addition, a significant inverse association between the tremor intensity and the expression level of ASFMR1 131 bp splice isoform in premutation carriers compared to controls, was observed, suggesting a potential role in the pathogenesis of FXTAS
