    Long-term effects of ayahuasca in patients with recurrent depression: a 5-year qualitative follow-up

    Background Ayahuasca is a botanical hallucinogenic preparation traditionally used by indigenous populations of Northwestern Amazonian countries for ritual and therapeutic purposes. It is rich in β-carboline alkaloids and N,N-dimethyltryptamine (DMT). Preclinical, observational, and experimental studies suggest that ayahuasca and its alkaloids have anxiolytic and antidepressive effects. We recently reported in an open-label trial that ayahuasca administration was associated with significant decreases in depression symptoms for 2-3 weeks after the experimental session in 17 patients with treatment-resistant major depressive disorder. Objectives To investigate if the experiment had any long-lasting effects on patients Methods Eight patients were interviewed 4 to 7 years after ayahuasca intake. Results Our results suggest that ayahuasca was well tolerated and that symptom reductions were limited to a few weeks. Importantly, most patients believed that the experience was among the most important of their lives, even 4-7 years later. Discussion To the best of our knowledge, this is the first long-term follow-up of a clinical sample that participated in an ayahuasca trial. Further studies with different and repeated dosing should be designed to further explore the antidepressive and anxiolytic effects of ayahuasca

    Alkane-grown <i>Beauveria bassiana</i> produce mycelial pellets displaying peroxisome proliferation, oxidative stress, and cell surface alterations

    The entomopathogenic fungus Beauveria bassiana is able to grow on insect cuticle hydrocarbons as the sole carbon source, inducing several enzymes involved in alkane assimilation and concomitantly increasing virulence against insect hosts. In this study, we describe some physiological and molecular processes implicated in growth, nutritional stress response, and cellular alterations found in alkane-grown fungi. The fungal cytology was investigated using light and transmission electron microscopy (TEM) while the surface topography was examined using atomic force microscopy (AFM). Fungal hydrophobicity was also measured on the cell surface. Additionally, the expression pattern of several genes associated with oxidative stress, peroxisome biogenesis, and hydrophobicity were analysed by qPCR. We found a novel type of growth in alkane-cultured B. bassiana similar to mycelial pellets described in other alkane-free fungi, which were able to germinate and produce viable conidia in media without a carbon source and to be pathogenic against larvae of the beetles Tenebrio molitor and Tribolium castaneum. Optical microscopy and TEM showed that pellets were formed by hyphae cumulates with high peroxidase activity, exhibiting peroxisome proliferation and an apparent surface thickening. Alkane-grown conidia appeared to be more hydrophobic and cell surfaces displayed different topography than glucose-grown cells, as it was observed by AFM. We also found a significant induction in several genes encoding for peroxins, catalases, superoxide dismutases, and hydrophobins.     Data-Centric Resource Management in Edge-Cloud Systems for the IoT

    A major challenge in emergent scenarios such as the Cloud-assisted Internet of Things is efficiently managing the resources involved in the system while meeting requirements of applications. From the acquisition of physical data to its transformation into valuable services or information, several steps must be performed, involving the various players in such a complex ecosystem. Support for decentralized data processing on IoT devices and other devices near the edge of the network, in combination with the benefits of cloud technologies has been identified as a promising approach to reduce communication overhead, thus reducing delay for time sensitive IoT applications. The interplay of IoT, edge and cloud to achieve the final goal of producing useful information and value-added services to end user gives rise to a management problem that needs to be wisely tackled. The goal of this work is to propose a novel resource management framework for edge-cloud systems that supports heterogeneity of both devices and application requirements. The framework aims to promote the efficient usage of the system resources while leveraging the Edge Computing features, to meet the low latency requirements of emergent IoT applications. The proposed framework encompasses (i) a lightweight and data-centric virtualization model for edge devices, (ii) a set of components responsible for the resource management and the provisioning of services from the virtualized edge-cloud resources

    Four new coordination polymers involving transition metals with 1,2,4,5-benzenetetracarboxylate and pyridyl-donor ligand di(4-pyridyl) sulfide

    AbstractFour new coordination polymers namely {[Mn2(BT)(DPS)2(H2O)6]·10H2O}n (MnBTDPS), {[Co2(BT)(DPS)2(H2O)6]·10H2O}n (CoBTDPS), {[Cu2(BT)(DPS)(H2O)4]·5H2O}n (CuBTDPS) and {[Zn2(BT)(DPS)2]·6H2O}n (ZnBTDPS), where BT=1,2,4,5-benzenetetracarboxylate and DPS=di(4-pyridyl) sulfide, were synthesized and characterized by thermal analysis, vibrational spectroscopy (Raman and infrared) and single crystal X-ray diffraction analysis. In all compounds, the DPS ligands are coordinated to metal sites in a bridging mode and the carboxylate moiety of BT ligands adopts a monodentate coordination mode, as indicated by the Raman spectra data through the Δν (νasym(COO)−ν sym(COO)) value. According to X-ray diffraction analysis, MnBTDPS and CoBTDPS are isostructural and in these cases, the metal centers exhibit a distorted octahedral geometry. In CuBTBPP, the Cu2+ centers geometries are best described as square-pyramids, according to the trigonality index τ=0.14 for Cu1 and τ=0.10 for Cu2. On the other hand, in ZnBTDPS, the Zn2+ sites adopt a tetrahedral geometry. Finally, the four compounds formed two-dimensional sheets that are connected to each other through hydrogen bonding giving rise to three-dimensional supramolecular arrays

    Peripheral antinociceptive action of mangiferin in mouse models of experimental pain: Role of endogenous opioids, KATP-channels and adenosine

    AbstractThis study aimed to assess the possible systemic antinociceptive activity of mangiferin and to clarify the underlying mechanism, using the acute models of chemical (acetic acid, formalin, and capsaicin) and thermal (hot-plate and tail-flick) nociception in mice. Mangiferin at oral doses of 10 to 100mg/kg evidenced significant antinociception against chemogenic pain in the test models of acetic acid-induced visceral pain and in formalin- and capsaicin-induced neuro-inflammatory pain, in a naloxone-sensitive manner, suggesting the participation of endogenous opiates in its mechanism. In capsaicin test, the antinociceptive effect of mangiferin (30mg/kg) was not modified by respective competitive and non-competitive transient receptor potential vanilloid 1 (TRPV1) antagonists, capsazepine and ruthenium red, or by pretreatment with l-NAME, a non-selective nitric oxide synthase inhibitor, or by ODQ, an inhibitor of soluble guanylyl cyclase. However, mangiferin effect was significantly reversed by glibenclamide, a blocker of KATP channels and in animals pretreated with 8-phenyltheophylline, an adenosine receptor antagonist. Mangiferin failed to modify the thermal nociception in hot-plate and tail-flick test models, suggesting that its analgesic effect is only peripheral but not central. The orally administered mangiferin (10–100mg/kg) was well tolerated and did not impair the ambulation or the motor coordination of mice in respective open-field and rota-rod tests, indicating that the observed antinociception was unrelated to sedation or motor abnormality. The findings of this study suggest that mangiferin has a peripheral antinociceptive action through mechanisms that involve endogenous opioids, KATP-channels and adenosine receptors

    Taurine: A Potential Ergogenic Aid for Preventing Muscle Damage and Protein Catabolism and Decreasing Oxidative Stress Produced by Endurance Exercise

    The aim of this study was to evaluate the effects of taurine and chocolate milk supplementation on oxidative stress and protein metabolism markers, and aerobic parameters in triathletes. Methods: A double-blind, crossover study was conducted with 10 male triathletes, aged 30.9 ± 1.3 year, height 1.79 ± 0.01 m and body weight 77.45 ± 2.4 kg. Three grams of taurine and 400 ml of chocolate milk (TAUchoc), or a placebo (chocolate milk) (CHOC) was ingested post exercise for 8 weeks. Oxidative stress marker levels, and 24 h urinary nitrogen, creatinine, and urea excretion were measured before and after 8 weeks of training and supplementation with TAUchoc or CHOC. A maximal incremental running test on a treadmill was performed in order to evaluate aerobic parameters: Vmax, heart rate (HR) and rate of perceived exertion (RPE). Results: TAUchoc treatment during the 8 weeks resulted in increased taurine plasma levels (PRE 201.32 ± 29.03 μmol/L and POST 234.36 ± 35.51 μmol/L, p = 0.01), decreased malondialdehyde levels (19.4%, p = 0.03) and urinary nitrogen excretion (−33%, p = 0.03), and promoted positive nitrogen balance (p = 0.01). There were no changes in reduced glutathione (TAUchoc PRE 0.72 ± 0.08 mmol/L and POST 0.83 ± 0.08 mmol/L; CHOC PRE 0.69 ± 0.08 mmol/L and POST 0.81 ± 0.06 mmol/L), vitamin E plasma levels (TAUchoc PRE 33.99 ± 2.52 μmol/L and 35.95 ± 2.80 μmol/L and CHOC PRE 31.48 ± 2.12 μmol/L and POST 33.77 ± 3.64 μmol/L), or aerobic parameters, which were obtained in the last phase of the maximal incremental running test (Vmax TAUchoc PRE 13 ± 1.4 km/h and POST 13.22 ± 1.34 km/h; CHOC PRE 13.11 ± 2.34 km/h and POST 13.11 ± 2.72 km/h), the heart rate values were TAUchoc PRE 181.89 ± 24.18 bpm and POST 168.89 ± 46.56 bpm; CHOC PRE 181.56 ± 2.14 bpm and POST 179.78 ± 3.4 bpm, and the RPE were TAUchoc PRE 8.33 ± 2.4 AU and POST 9.1 ± 2.1 AU; CHOC PRE 8.11 ± 4.94 AU and POST 8.78 ± 2.78 AU). Conclusion: Taurine supplementation did not improve aerobic parameters, but was effective in increasing taurine plasma levels and decreasing oxidative stress markers, which suggests that taurine may prevent oxidative stress in triathletes

    Interconnectedness during high water maintains similarity in fish assemblages of island floodplain lakes in the Amazonian Basin

    We conducted a study to test the hypothesis that interconnectedness among island floodplain lakes and the adjacent Solimões River during the flood stage of the hydrologic cycle is enough to maintain similarity in fish species assemblages. Gill net samples were collected during high and low water periods for three consecutive years (July 2004 to July 2006) in four lakes on Paciência Island. Two lakes, Piranha and Ressaca, are connected to the river all year, and the other two, Preto and Cacau, which are in the center of the island, are isolated during low water periods. The abundance, species richness and evenness of the fish assemblages in these lakes did not differ according to their relative positions or the season of the hydrological cycle, which confirmed our hypothesis. However, fish abundance during the dry season was greater than in the flood season. Apparently, the short period of full connection between the lakes is enough to allow the colonization of all fish species, but not to cause similar abundances. Our study indicates that persistence of the species composition of island floodplain lakes is primarily due to the annual replenishment of fish to the lakes during the flood season
