    Caracterização da imunogenicidade das proteínas recombinantes Virb9, Virb10 e fator termo instável de Elongação de Peptídeos de Anaplasma marginale em camundongos

    Considering the limitations of current methods of anaplasmosis control, the development of a more effective vaccine is required. Previous studies, using proteomic and genomic approaches, have identified new membrane proteinsin Anaplasma marginale that may be vaccine candidates. These include VirB9, VirB10 and elongation factor-Tu (EFTu).Although the role of these proteins in the membrane of A. marginale has not been properly characterized, production of the recombinant proteins rVirB9, rVirB10, and rEF-Tu has been achieved. However, these recombinant proteins have not yet been exploited in vaccine formulations. The present study describes the use of rVirB9, rVirB10and rEF-Tu, emulsificated in adjuvant Montanide, in mice. A strong humoral immune response was induced under these conditions, with both IgG1 and IgG2a production. The IgG2a/IgG1 ratios revealed a predominance of IgG1.However, splenocytes of the animals that received rVirB9 or rVirB10 produced high levels of gamma interferon after in vitro stimulation, indicating a specific cellular immune response to these proteins. Therefore, further studies are required to adjust the profile of the immune response in order to perform tests of protection against A. marginale in cattle.Considerando as limitações dos atuais métodos de controle contra a anaplasmose bovina, o desenvolvimento de uma vacina efetiva se faz necessário. A partir do advento da análise genômica e proteômica, novas proteínas de membrana de Anaplasma marginale foram identificadas como possíveis candidatas a componentes de uma vacina, tais como, VirB9,VirB10 e Fator Termo Instavél de Elongação de Peptídeos (EF-Tu). Embora estas proteínas ainda não estejam bem caracterizadas na membrana de A. marginale, a produção destas na forma recombinante (rVirB 9, rVirB10 e rEF-Tu)tem sido realizada, mas as mesmas ainda não foram exploradas em formulações vacinais. Neste trabalho, avaliou-se ouso de rVirB9, rVirB10 e rEF-Tu emulsionadas em adjuvante Montanide em camundongos. Nas condições testadas, verificou-se a indução de forte resposta imune humoral com a produção de IgG1 e IgG2a, sendo que as proporções dos níveis de produção destas subclasses indicam predomínio de IgG1. Entretanto, esplenócitos de animais, que foram injetados com rVirB9 ou rVirB10, produziram interferon-gama acima do limite de detecção do ensaio após estimulação in vitro, sinalizando assim resposta celular específica. Assim, novas avaliações serão realizadas com a finalidade de modular o perfil de resposta imune obtido em bovinos e avaliar a proteção contra A. marginale

    Mycobacterium bovis is a causal agent of bovine tuberculosis (bTB), one of the most important diseases currently facing the cattle industry worldwide. Tracing the source of M. bovis infections of livestock is an important tool for understanding the epidemiology of bTB and defining control/eradication strategies. In this study, whole genome sequencing (WGS) of 74 M. bovis isolates sourced from naturally infected cattle in the State of Rio Grande do Sul (RS), southern Brazil, was used to evaluate the population structure of M. bovis in the region, identify potential transmission events and date the introduction of clonal complex (CC) European 2 (Eu2). In silico spoligotyping identified 11 distinct patterns including four new profiles and two CCs, European 1 (Eu1) and Eu2. The analyses revealed a high level of genetic diversity in the majority of herds and identified putative transmission clusters that suggested that within- and between-herd transmission is occurring in RS. In addition, a comparison with other published M. bovis isolates from Argentina, Brazil, Paraguay and Uruguay demonstrated some evidence for a possible cross-border transmission of CC Eu1 into RS from Uruguay or Argentina. An estimated date for the introduction of CC Eu2 into RS in the middle of the 19th century correlated with the historical introduction of cattle into RS to improve existing local breeds. These findings contribute to the understanding of the population structure of M. bovis in southern Brazil and highlight the potential of WGS in surveillance and helping to identify bTB transmission

    SEROLOGICAL, PARASITOLOGICAL AND MOLECULAR ASSESSMENT OF Babesia bovis AND Babesia bigemina IN CATTLE FROM STATE OF MARANHÃO

    The aim of the present study was to determine the prevalence of Babesia bovis and Babesia bigemina in dairy cattle from São Luis Island in the state of Maranhão, Brazil. A total of 281 blood samples were collected. In total, 275 (97.9%) animals were B. bovis-reactive and B. bigemina reactive in the Enzyme-Linked Immunosorbent Assay (ELISA). The microscopy examination detected 22 (7.8%) animals that were positive for Babesia sp. and the Polymerase Chain Reaction (PCR) analysis showed that 91 animals (32.38%) and 23 animals (8.18%) were positive for B. bovis and B. bigemina, respectively, while 17 animals (6.04%) were co-infected. There is a high level of transmission of these protozoa in Maranhão, and the animals were naturally exposed. Therefore, it is possible to characterize the island as enzootic stability for babesiosis, indicat-ing a risk of financial losses when susceptible animals are introduced from areas of enzootic instability or free regions of B. bovis and B. bigemina

    Freqüência de anticorpos para Babesia bigemina, B. bovis, Anaplasma marginale, Trypanosoma vivax e Borrelia burgdorferi em bovinos do nordeste do Estado do Pará, Brasil

    Babesiosis, anaplasmosis, and trypanosomosis are relevant diseases, potentially causing morbidity in cattle, leading to economic losses. Borreliosis is import as a potential zoonosis. The objective of this study was to determine, by indirect enzyme-linked immunosorbent assay (ELISA), the frequency of seropositive cattle to Babesia bigemina, B. bovis, Anaplasma marginale, Trypanosoma vivax and Borrelia burgdorferi in cattle from the Northeastern region of Pará, Brazil. Sera samples from 246 female adult cattle from municipalities of Castanhal and São Miguel do Guamá were used. Crude antigens ELISAs were used to detect antibodies to all agents, except to A. marginale, to which an indirect ELISA with recombinant major surface 1a protein (MSP1a) antigen was used. Overall frequencies of seropositive animals were: B. bigemina - 99.2%; B. bovis - 98.8%; A. marginale - 68.3%; T. vivax - 93.1% and B. burgdorferi - 54.9%. The frequencies of seropositive cattle to B. bovis and B. bigemina suggest a high rate of transmission of these organisms by tick in the studied region, which can be classified as enzootically stable to these hemoprotozoans. The low frequency of seropositive cattle to A. marginale may be attributed to a lower sensitivity of the recombinant antigen ELISA utilized or a distinct rate of inoculation of this rickettsia by ticks, as compared with Babesia sp. transmission. The high frequency of seropositive cattle to T. vivax indicates that this hemoprotozoan is prevalent in herds from the Northeastern region of Pará. The rate of animal that showed homologues antibodies to B. burgdorferi indicates the presence of the tickborne spirochaetal agent in the cattle population in the studied region.A babesiose, a anaplasmose e a tripanossomose são enfermidades relevantes, potencialmente causadoras de morbidade em bovinos, levando a perdas econômicas. A borreliose assume importância como zoonose potencial. O objetivo desse estudo foi determinar, por meio do ensaio de imunoadsorção enzimática (ELISA), a freqüência de anticorpos para Babesia bigemina, B. bovis, Anaplasma marginale, Trypanosoma vivax e Borrelia burgdorferi em bovinos da região nordeste do Estado do Pará, Brasil. Amostras de soro de 246 vacas dos municípios de Castanhal e São Miguel do Guamá foram usadas. ELISAs com antígeno bruto foram utilizados para detector anticorpos contra todos os agentes, exceto para A. marginale, para o qual um ELISA indireto com proteína principal de superfície 1a (MSP1a) foi usado. As freqüências de bovinos soropositivos foram: B. bigemina - 99,2%; B. bovis - 98,8%; A. marginale - 68,3%; T. vivax - 93,1% and B. burgdorferi -54,9% As freqüências de bovinos soropositivos para B. bovis e B. bigemina sugerem uma alta taxa de transmissão desses organismos por carrapatos, na região estudada, a qual pode ser classificada com sendo de estabilidade enzoótica para os hemoparasitos. A baixa freqüência de bovinos soropositivos para A. marginale pode ser atribuída a uma menor sensibilidade do ELISA com antígeno recombinante, ou uma menor taxa de inoculação da riquétsia pelos carrapatos, quando comparada àquelas observadas para Babesia sp. A alta freqüência de bovinos soropositivos para T. vivax indica que esse hemoprotozoário é prevalente em rebanhos do nordeste do Estado do Pará. O percentual de animais com anticorpos homólogos para B. burgdorferi indica a presenças deste espiroquetídeo transmitido por carrapatos na população de bovinos da região estudada

    Morfologia e morfometria de três isolados de Plasmodium juxtanucleare (Apicomplexa: Plasmodiidae)

    In this work, three isolates of Plasmodium juxtanucleare have been analyzed based on morphological, morphometric and parasitic parameters. Each isolate was sampled from naturally infected adult chicken (Gallus gallus) from rural areas of three Brazilian municipalities: Seropédica (22º 48' S; 43º 41'W), in the state of Rio de Janeiro; Cruzeiro (22º 33' S; 44º 57'W), in the state of São Paulo; and Santa Bárbara do Tugúrio (21º 15' S; 43º 27' W), in the state of Minas Gerais. The blood samples taken from each infected chicken were inoculated in three groups of ten young chicken (21 days old). Blood smears of the experimentally infected chicken were sampled every two days until the 69th day in order to evaluate the parasitemia. For the morphological-descriptive and morphometric analyses, we measured 30 individuals from each of the intraerythocytic states, measures of the major (MD) and minor diameters (md), the estimation of morphometric index (Mi=md/MD) and size (T=pab, a= md/2; b=MD/2). The results indicated low and homogeneous parasitemia rates in the three strains, which showed differences among shape and size of the parasitic stadia displayed.Neste trabalho, três isolados de Plasmodium juxtanucleare foram analisados com base na morfologia, morfometria e parâmetros parasitológicos. Cada isolado foi coletado de aves (Gallus gallus) adultas infectadas naturalmente de áreas rurais de três municípios brasileiros: Seropédica (22º 48' S; 43º 41' W), no estado do Rio de Janeiro; Cruzeiro (22º 33' S; 44º 57' W), no estado de São Paulo; e Santa Bárbara do Tugúrio (21º 15' S; 43º 27' W), no estado de Minas Gerais. As amostras de sangue coletadas de cada ave infectada foram inoculadas em três grupos de dez aves (21 dias de idade). Esfregaços sangüíneos das aves infectadas experimentalmente foram realizados de dois em dois dias durante um período de 69 dias para avaliar a parasitemia. Para análises morfofisiológica e morfométrica, foram mensurados 30 indivíduos de cada estágio intraeritrocítico. Foram tomadas as medidas do diâmetro maior (DM) e diâmetro menor (dm), com os quais foi estimado o índice morfométrico (Mi=md/MD) e o tamanho (T=pab, a= md/ 2; b=MD/2). Os resultados indicaram uma parasitemia homogênea entre os três isolados, havendo diferenças apenas nas formas e tamanhos dos estádios parasitários

    Molecular identification of Hepatozoon canis in dogs from Campo Grande, Mato Grosso do Sul, Brazil

    The aim of this study was to investigate the occurrence of Hepatozoon species infecting dogs in the municipality of Campo Grande, Mato Grosso do Sul (MS), Brazil, using blood samples (n = 165) drawn from dogs. The species Hepatozoon caniswas identified in 3.63% of the tested animals using molecular tools. Further studies are needed to determine the clinical relevance of this infection and the main arthropod vectors involved in its transmission
