    Metatranscriptomics reveals temperature-driven functional changes in microbiome impacting cheese maturation rate

The definitive version was published in Scientific Reports 6 (2016): 21871, doi:10.1038/srep21871.Traditional cheeses harbour complex microbial consortia that play an important role in shaping typical sensorial properties. However, the microbial metabolism is considered difficult to control. Microbial community succession and the related gene expression were analysed during ripening of a traditional Italian cheese, identifying parameters that could be modified to accelerate ripening. Afterwards, we modulated ripening conditions and observed consistent changes in microbial community structure and function. We provide concrete evidence of the essential contribution of non-starter lactic acid bacteria in ripening-related activities. An increase in the ripening temperature promoted the expression of genes related to proteolysis, lipolysis and amino acid/lipid catabolism and significantly increases the cheese maturation rate. Moreover, temperature-promoted microbial metabolisms were consistent with the metabolomic profiles of proteins and volatile organic compounds in the cheese.     Mental health in Italy after two years of COVID-19 from the perspective of 1281 Italian physicians: looking back to plan forward

    Background: The COVID-19 pandemic has generated an unprecedented global crisis that is profoundly affecting mental health and mental health care. The aim of this study was to survey a relatively large group of Italian physicians about their perceived impact of COVID-19 on the mental health of the Italian population and about their suggestions on the best strategies to address the current and future challenges. Methods: One thousand two hundred eighty-one (1,281) physicians were surveyed between November 2021 and February 2022. Results: Eighty-one percent of respondents reported an increase in the number of people with mental illness presenting to their practice during the COVID-19 pandemic. Thirty-four percent reported a 26-50% increase in the number of people with mental illness in their community; approximately 33% reported a 1-25% increase; and 26.9% reported a 51-75% increase. The most commonly reported mental issues that increased because of COVID-19 were agitation, mood and anxiety disorders. Regarding the suggested strategies to address future challenges related to the COVID-19 pandemic, 34.6% of respondents recommended providing psychoeducation to the general population for early detection of mental illness and developing strategies to reduce the impact of COVID-19-related stress. In addition, 12.6% of respondents suggested improving telehealth services, while 12.3% suggested the need for increased funding for community-based care. When asked about physicians' opinion on the possibility of an increased prevalence of mental illness in the next 12 months, more than 30% of them predicted an increase in stress-related illnesses, while 25.2% were more concerned about a worsening of the ongoing clinical conditions of patients with previous psychiatric disorders. However, 21% of respondents believed that people's ability to cope with the pandemic would increase in the next 12 months. Conclusions: This study confirmed a strong and negative impact on the mental health of the past 2 years of COVID-19 pandemic in the Italian population. Providing psychoeducation to the general population and improving the availability of telemedicine services could reduce the impact of future challenges related to the pandemic

    Vulnerabilities in Mental Health due to Covid-19 Pandemic: The Response of the Italian Physicians

    COVID-19 pandemic has exacerbated the pre-existing vulnerabilities and inequalities in societies. In this paper we analyse the categories that have suffered more than others from the pandemic and the restrictions on social life in terms of mental health. We rely on the Serendipity project based on a survey administered between November 2021 and February 2022 to a sample of Italian physicians (n = 1281). The survey aimed to assess the perception of general practitioners, paediatricians, geriatricians, and mental health specialists (psychiatrists, neurologists, child neuropsychiatrists), about changes in the mental health of the population as an effect of the COVID-19 pandemic and the lockdown. The strategies implemented by the doctors interviewed in terms of the intensity of the prevention, emergence, and treatment of mental health interventions, and their association with physicians' characteristics and their opinions on patient vulnerability have been illustrated by means of a multiple correspondence analysis. An overall result of the survey is the consensus of doctors on the worsening of mental health in general population, especially among their patients, due to the pandemic and on the onset of new discomforts. The most exposed individuals to the risk of onset or worsening of mental disorders include women, young people, and patients with psychiatric comorbidity. The paper also illustrates the interventions put in place by the physicians and deemed necessary from a public heath response perspective, that include providing psychoeducation to the general population, improving telehealth services, and increasing financial and human resources for community-based care

    Hydrological Web Services for Operational Flood Risk Monitoring and Forecasting at Local Scale in Niger

    Emerging hydrological services provide stakeholders and political authorities with useful and reliable information to support the decision-making process and develop flood risk management strategies. Most of these services adopt the paradigm of open data and standard web services, paving the way to increase distributed hydrometeorological services’ interoperability. Moreover,sharing of data, models, information, and the use of open-source software, greatly contributes to expanding the knowledge on flood risk and to increasing flood preparedness. Nevertheless, services’ interoperability and open data are not common in local systems implemented in developing countries. This paper presents the web platform and related services developed for the Local Flood Early Warning System of the Sirba River in Niger (SLAPIS) to tailor hydroclimatic information to the user’s needs, both in content and format. Building upon open-source software components and interoperable web services, we created a software framework covering data capture and storage, data flow management procedures from several data providers, real-time web publication, and service-based information dissemination. The geospatial infrastructure and web services respond to the actual and local decision-making context to improve the usability and usefulness of information derived from hydrometeorological forecasts, hydraulic models, and real-time observations. This paper presents also the results of the three years of operational campaigns for flood early warning on the Sirba River in Niger. Semiautomatic flood warnings tailored and provided to end users bridge the gap between available technology and local users’ needs for adaptation, mitigation, and flood risk management, and make progress toward the sustainable development goals

    Hydrothermal Liquefaction of Waste Biomass Model Compounds: a Study to Unravel the Complexity of Interactions in Biocrude Production from Mixtures of Cellulose-Albumin-Lipids

    Hydrothermal liquefaction is a promising technology for liquid biofuel production from a wide range of organic wastes. Waste biogenic feedstocks with a high moisture content are particularly suitable for this purpose due to the possibility to feed wet materials and to obtain high liquid yields in hydrothermal liquefaction (HTL). Although, yields and quality of the obtained liquid products are usually strongly dependent on the composition of the feedstocks, and due to their variability, it is often difficult to have reliable predictions. However, biogenic waste can be easily schematize based on their content of organic macro-components, mainly polysaccharides, proteins, and lipids. This work tries to summarize the effect of the variation of feedstock’s composition on yields and the quality of HTL products, with a particular focus on binary interactions between the macro-components. Cellulose, egg albumin and sunflower oil are used as model compounds to represent polysaccharides, proteins, and lipids, respectively. HTL tests are carried out in micro autoclaves of 10 mL using these model compounds alone and in binary and ternary mixtures as feedstocks, at 330°C and 10 minutes of retention time. Results showed that the biocrude yields did not follow the behaviour predicted by the linear combination of the three compounds but an increase of biocrude production and a reduction of solid residue is obtained for the mixtures. GC-MS results showed the presence of compounds related to Maillard reactions and amides formation. Some general reaction pathways were summarized to explain these results. The comprehension of these interactions can guide the future research to obtain a prediction model for biofuel production through a HTL process

    Lurasidone in adolescents and adults with schizophrenia: from clinical trials to real-world clinical practice

    Introduction: Lurasidone is an atypical antipsychotic agent approved in the European Union for the treatment of schizophrenia in adults and adolescents (13-17 years). Clinical trials have shown a generally favorable balance between efficacy and tolerability. Areas covered: This paper provides a review and commentary regarding the use of lurasidone in adults and adolescents with schizophrenia. The available information about efficacy, tolerability, dosing, and switching is analyzed, highlighting the strategies that may be most useful in real-world clinical practice. Virtual case studies, designed based on the authors' clinical experience with real-world patients, are provided. Expert opinion: Lurasidone is efficacious in adolescents and adults in a wide range of symptoms of schizophrenia. Choosing the right dose for each patient and combining lurasidone with other medications is key to treatment success. Lurasidone has proven effective both in adolescents and adults in treating the acute phase of schizophrenia and reducing the risk of relapse. It has shown a relatively favorable tolerability profile, with minimal effects on metabolic parameters and prolactin levels

    Antimicrobial Applications of Green Synthesized Bimetallic Nanoparticles from Ocimum basilicum

    Antibiotic resistance is an important and emerging alarm for public health that requires development of new potential antibacterial strategies. In recent years, nanoscale materials have emerged as an alternative way to fight pathogens. Many researchers have shown great interest in nanoparticles (NPs) using noble metals, such as silver, gold, and platinum, even though numerous nanomaterials have shown toxicity. To overcome the problem of toxicity, nanotechnology merged with green chemistry to synthesize nature-friendly nanoparticles from plants. Here, we describe the synthesis of NPs using silver (AgNPs) and platinum (PtNPs) alone or in combination (AgPtNPs) in the presence of Ocimum basilicum (O. basilicum) leaf extract. O. basilicum is a well-known medicinal plant with antibacterial compounds. A preliminary chemical–physical characterization of the extract was conducted. The size, shape and elemental analysis were carried out using UV–Visible spectroscopy, dynamic light scattering (DLS), and zeta potential. Transmission electron microscopy (TEM) confirmed polydisperse NPs with spherical shape. The size of the particles was approximately 59 nm, confirmed by DLS analysis, and the polydisperse index was 0.159. Fourier transform infrared (FTIR) demonstrated an effective and selective capping of the phytoconstituents on the NPs. The cytotoxic activities of AgNPs, PtNPs and AgPtNPs were assessed on different epithelial cell models, using the 3-[4.5-dimethylthiazol-2-yl]-2.5-diphenyltetrazolium bromide (MTT) cell proliferation assay, and discovered low toxicity, with a cell viability of 80%. The antibacterial potential of the NPs was evaluated against Escherichia coli (E. coli), Enterococcus faecalis (E. faecalis), Klebsiella pneumonia (K. pneumoniae), and Staphylococcus aureus (S. aureus) strains. Minimum inhibitory concentration (MIC) assays showed AgPtNP activity till the least concentration of NPs (3.15–1.56 g/mL) against ATCC, MS, and MDR E. coli, E. faecalis, and S. aureus and the Kirby–Bauer method showed that AgPtNPs gave a zone of inhibition for Gram-positive and Gram-negative bacteria in a range of 9–25 mm. In addition, we obtained AgPtNP synergistic activity in combination with vancomycin or ampicillin antibiotics. Taken together, these results indicate that bimetallic nanoparticles, synthesized from O. basilicum leaf extract, could represent a natural, ecofriendly, cheap, and safe method to produce alternative antibacterial strategies with low cytotoxicity

    The IDEA detector concept for a future e+^{+}e−^{-} collider adopts an ultra-low mass drift chamber as a central tracking system. The He-based ultra-low mass drift chamber is designed to provide efficient tracking, a high-precision momentum measurement, and excellent particle identification by exploiting the cluster counting technique. This paper describes the expected tracking performance, obtained with full and fast simulation, for track reconstruction on detailed simulated physics events. Moreover, the details of the construction parameters of the drift chamber, including the inspection of new material for the wires, new techniques for soldering the wires, the development of an improved schema for the drift cell, and the choice of a gas mixture, will be described

    Bioaccumulation of dioxin-like substances and selected brominated flame retardant congeners in the fat and livers of black pigs farmed within the Nebrodi Regional Park of Sicily.

    An observational study was designed to assess the bioaccumulation of polychlorodibenzodioxins (PCDD) and polychlorodibenzofurans (PCDF), dioxin-like polychlorobiphenyls (DL-PCB), and 13 selected polybromodiphenylethers (PBDE) in autochthonous pigs reared in the Nebrodi Park of Sicily (Italy). Perirenal fat and liver samples were drawn from animals representative of three different outdoor farming systems and from wild pigs and then analyzed for the chemicals mentioned previously. The highest concentrations of PCDD + PCDF and DL-PCB were detected in the fat (0.45 and 0.35 pg World Health Organization toxicity equivalents [WHO-TE] per g of fat base [FB], respectively) and livers (12.7 and 3.28 pg WHO-TE per g FB) of the wild group, whereas the free-ranging group showed the lowest levels (0.05 and 0.03 pg WHO-TE per g FB in fat and 0.78 and 0.27 pg WHO-TE per g FB in livers). The sum of PBDE congeners was highest in wild pigs (0.52 ng/g FB in fat and 5.64 ng/g FB in livers) and lowest in the farmed group (0.14 ng/g FB in fat and 0.28 ng/g FB in livers). The contamination levels in fat and livers of outdoor pigs had mean concentration values lower than those levels reported for intensively indoor-farmed animals. In wild pigs, bioaccumulation was associated with their free grazing in areas characterized by bush fires. The results of this study aid to emphasize the quality of the environment as a factor to guarantee food safety in typical processed pig meat products, specifically from outdoor and extensive Nebrodi farming systems
