    Are all pastures eligible for conservation? A phytosociological survey of the Sardinian–Corsican Province as a basic tool for the Habitats Directive

    A consistent vegetation classification is an essential tool for conservation and monitoring purposes, also for semi-natural habitats such as pastures and hay meadows that are linked to traditional land use and generally considered crucial to maintain biodiversity within agricultural systems. Indeed, these habitats can be strongly affected by land abandonment or agriculture intensification. Despite their importance in the framework of the EU Habitats Directive (43/92/EEC), information on distribution, species composition, and conservation status is still lacking for many regions. To fill these gaps, we investigated from a phytosociological point of view the sheep pastures of the North-Western Sardinian trachy-basaltic sector. Three main communities were described as new: (i) perennial montane (meso-supratemperate) cattle and sheep pastures (Loto alpini- Festucetum morisianae); (ii) mixed (annual and perennial) Mediterranean (lower Thermomediterranean to lower supratemperate) sheep pastures (Ornithogalo corsici-Poetum bulbosae); and (iii) annual ploughed subnitrophilous Mediterranean and Temperate pastures (Cynosuro polybracteati-Vulpietum ligusticae). As the first two host numerous endemic taxa and show a peculiar floristic composition, we assign them a higher conservation value, testified also by their classification in two new syntaxa: Danthonio decumbentis-Caricenion insularis (Molinio-Arrhenatheretea) and Ornithogalo corsici-Trifolienion subterranei (Poetea bulbosae), respectively

    A phytosociological study of the deciduous oak woods of Sardinia (Italy)

    We present here the results from a phytosociological and synchorological study of the deciduous oak woods of Sardinia. This vegetation analysis has allowed the individuation of three new associations: Lonicero implexae-Quercetum virgilianae, Ornithogalo pyrenaici-Quercetum ichnusae and Glechomo sardoae-Quercetum congestae. The Lonicero implexae-Quercetum virgilianae association includes the woods on calcareous substrata spread throughout Northern Sardinia, which can occasionally be found in southern areas; the first of these are rich in mesophilous species and are attributed to the subassociation cyclaminetosum repandi, while the others are referred to quercetosum virgilianae. The Ornithogalo pyrenaici-Quercetum ichnusae association, more widely distributed in Central-Northern Sardinia, includes the climatophilous deciduous woods that are found on lithological substrata of a non-carbonatic nature, and in particular, on andesites, trachytes and metarenites. The typical aspect is referred to the subassociation cytisetosum villosi, while the subassociation ilicetosum aquifolii includes the more mesophilous woods on basaltic substrata. The Glechomo sardoae- Quercetum congestae association always includes the woods found on the non-carbonatic substrata in the higher altitude areas with an oceanic pluviseasonal temperate bioclimate. Of this, the subassociation quercetosum congestae, on the granite areas, and oenanthetosum pimpinelloidis, on those metamorphic and of basalt, are proposed. At the higher hierarchical levels, the association Lonicero implexae-Quercetum virgilianae is referred to the endemic Sardinian-Corsican suballiance Clematido cirrhosae-Quercenion ilicis, of the alliance Fraxino orni-Quercion ilicis, while for the other associations the new suballiance Paeonio morisii-Quercenion ichnusae, (holotypus: Glechoma sardoae-Quercetum congestae ass. nova) particular to the Sardinian biogeographic subprovince, is proposed, of the alliance Pino calabricae-Quercion congestae, the order Quercetalia pubescentipetraeae, and the class Querco roboris-Fagetea sylvaticae

    ETP calculation for three important irriguous areas of Southern (39°17' N. I), Central (39°53' N. I) and Northern (40°41' N. I) Sardinia through F.A.O. method shows too much high values with originai Blaney-Criddle equation, reduced differences with Penman (original and F.A.O.) equation, Radiation method and B.-C. F.A.O. equation respectively. The last method, believed the most suitable according to the first lisimetric measurements, estimates annual mean ETP equal to 1194 mm, 1077 mm and 1022 mm for Southern, Central and Northern Sardinia. Effective rainfall successive analysis shows they are 50% of total rainfall (in case of grasses) and they hold ETP for about 22%. Therefore seasonal irrigation requirements, calculated through F.A.O. crop coefficients, are among 841 mm, 791 mm and 706 mm concerning grasses, and 353 mm, 322 mm and 311 mm concerning olive tree, for Southern, Central and Northern Sardinia

    The diacylglycerol kinase α/Atypical PKC/β1 integrin pathway in SDF-1α mammary carcinoma invasiveness

    Diacylglycerol kinase α (DGKα), by phosphorylating diacylglycerol into phosphatidic acid, provides a key signal driving cell migration and matrix invasion. We previously demonstrated that in epithelial cells activation of DGKα activity promotes cytoskeletal remodeling and matrix invasion by recruiting atypical PKC at ruffling sites and by promoting RCP-mediated recycling of α5β1 integrin to the tip of pseudopods. In here we investigate the signaling pathway by which DGKα mediates SDF-1α-induced matrix invasion of MDA-MB-231 invasive breast carcinoma cells. Indeed we showed that, following SDF-1α stimulation, DGKα is activated and localized at cell protrusion, thus promoting their elongation and mediating SDF-1α induced MMP-9 metalloproteinase secretion and matrix invasion. Phosphatidic acid generated by DGKα promotes localization at cell protrusions of atypical PKCs which play an essential role downstream of DGKα by promoting Rac-mediated protrusion elongation and localized recruitment of β1 integrin and MMP-9. We finally demonstrate that activation of DGKα, atypical PKCs signaling and β1 integrin are all essential for MDA-MB-231 invasiveness. These data indicates the existence of a SDF-1α induced DGKα - atypical PKC - β1 integrin signaling pathway, which is essential for matrix invasion of carcinoma cells

    Both ghrelin deletion and unacylated ghrelin overexpression preserve muscles in aging mice

    Sarcopenia, the decline in muscle mass and functionality during aging, might arise from age-associated endocrine dysfunction. Ghrelin is a hormone circulating in both acylated (AG) and unacylated (UnAG) forms with antiatrophic activity on skeletal muscle. Here, we show that not only lifelong overexpression of UnAG (Tg) in mice, but also the deletion of ghrelin gene (Ghrl KO) attenuated the age-associated muscle atrophy and functionality decline, as well as systemic inflammation. Yet, the aging of Tg and Ghrl KO mice occurs with different dynamics: while old Tg mice seem to preserve the characteristics of young animals, Ghrl KO mice features deteriorate with aging. However, young Ghrl KO mice show more favorable traits compared to WT animals that result, on the whole, in better performances in aged Ghrl KO animals. Treatment with pharmacological doses of UnAG improved muscle performance in old mice without modifying the feeding behavior, body weight, and adipose tissue mass. The antiatrophic effect on muscle mass did not correlate with modifications of protein catabolism. However, UnAG treatment induced a strong shift towards oxidative metabolism in muscle. Altogether, these data confirmed and expanded some of the previously reported findings and advocate for the design of UnAG analogs to treat sarcopenia

    The Wikiplantbase project: the role of amateur botanists in building up large online floristic databases

    The Wikiplantbase project, started in 2013, provides a framework where the full set of georeferenced floristic records of Tuscany and Sardinia can be entered, stored, updated and freely accessed through the Internet. Mainly thanks to the collaboration of amateur botanists, data have accumulated quickly. All records entered by collaborators are submitted to the project coordinators, who are enabled to accept, modify, or reject them. As of 22 November 2016, Wikiplantbase #Toscana holds 116,402 verified floristic records (90% based on published literature, 5% on unpublished herbarium specimens, 5% on field observations), and Wikiplantbase #Sardegna 40,043 (77% published literature, 18% unpublished herbarium specimens, 5% on field observations ). The records include over 90% of the specific and subspecific taxa known for Tuscany and about 70% – but rapidly growing – of those known for Sardinia. The most recorded species are Quercus ilex L. (Fagaceae) for Tuscany and Pistacia lentiscus L. (Anacardiaceae) for Sardinia. With minor software tweaking, the online platform Wikiplantbase might be adopted in other contexts, resulting in a well connected network of regional floristic databases suited to exploit the involvement – still largely untapped – of nonacademic collaborators, as advocated by citizen science

    Évaluation préliminaire des essais de provenances de chêne-liège et perspectives de recherche

    Le réseau international des essais de provenance et de descendance de chêne-liège (établie dans le cadre du projet Action Concertée FAIR CT 95-202 “European network for the evaluation of genetic resources of cork oak for appropriate use in breeding and gene conservation strategies” financé par l’Union Européenne) constitue un matériel vivant unique pour la recherche, qu’elle soit appliquée, ou fondamental, sur l’espèce. Les essais génétiques ont été réalisés sur une base méthodologique harmonisée au niveau des plants utilisés (tous les plants ont été élevées dans une même pépinière au Portugal) et sur terrain un même dispositif expérimental a été suivi. En 1997/98, 13 essais de provenances ont été établis. Ceux ayant réussi sont situés en Espagne (2), France (1), Italie (3), Portugal (3) et Tunisie (1). Concernent les essais de descendances il existe 4 des 5 initiales - Espagne (1), Portugal (2) et Tunisie (1). Dans ce travail nous présentons quelques résultats du comportement des provenances à travers des caractéristiques adaptatifs tels que la survie, l’efficacité d’utilisation de l’eau et des études de génomique. Nous avons procédé à la comparaison de chaque provenance dans les essais - niveau horizontal- et à la comparaison de chaque provenance entre les essais - niveau vertical afin de discuter la plasticité phénotypique et l’interaction génotype X environnement. Les données disponibles pour hauteur en 2006 montre des différences de comportement entre provenances et entre les essais. Par exemple, les provenances du Maroc montre une croissance supérieure dans l’essai plus au Sud au Portugal et aussi dans un essai en Espagne. Pour l’essai de Sardaigne les meilleures provenances sont de Tunisie et d’Espagne. A la fin on discute sur les avantages des essais comme matériel vivant dénominateur commun pour une recherche avancée, multidisciplinaire, synergétique, transnational et concertée. Les perspectives pour la recherche sur les grands thèmes tels que la gestion durable du chêne-liège, notamment la sélection, amélioration génétique, adaptation aux changements globaux, fluxes génétiques, problèmes phytosanitaires, qualité du liège en liaison avec la respective génomique sont adressées.PTDC/AGR-AAM/104364/200

    A Concerted Kinase Interplay Identifies PPARγ as a Molecular Target of Ghrelin Signaling in Macrophages

    The peroxisome proliferator-activator receptor PPARγ plays an essential role in vascular biology, modulating macrophage function and atherosclerosis progression. Recently, we have described the beneficial effect of combined activation of the ghrelin/GHS-R1a receptor and the scavenger receptor CD36 to induce macrophage cholesterol release through transcriptional activation of PPARγ. Although the interplay between CD36 and PPARγ in atherogenesis is well recognized, the contribution of the ghrelin receptor to regulate PPARγ remains unknown. Here, we demonstrate that ghrelin triggers PPARγ activation through a concerted signaling cascade involving Erk1/2 and Akt kinases, resulting in enhanced expression of downstream effectors LXRα and ABC sterol transporters in human macrophages. These effects were associated with enhanced PPARγ phosphorylation independently of the inhibitory conserved serine-84. Src tyrosine kinase Fyn was identified as being recruited to GHS-R1a in response to ghrelin, but failure of activated Fyn to enhance PPARγ Ser-84 specific phosphorylation relied on the concomitant recruitment of docking protein Dok-1, which prevented optimal activation of the Erk1/2 pathway. Also, substitution of Ser-84 preserved the ghrelin-induced PPARγ activity and responsiveness to Src inhibition, supporting a mechanism independent of Ser-84 in PPARγ response to ghrelin. Consistent with this, we found that ghrelin promoted the PI3-K/Akt pathway in a Gαq-dependent manner, resulting in Akt recruitment to PPARγ, enhanced PPARγ phosphorylation and activation independently of Ser-84, and increased expression of LXRα and ABCA1/G1. Collectively, these results illustrate a complex interplay involving Fyn/Dok-1/Erk and Gαq/PI3-K/Akt pathways to transduce in a concerted manner responsiveness of PPARγ to ghrelin in macrophages
