    Fatigue crack propagation prediction of a pressure vessel mild steel based on a strain energy density model

    Fatigue crack growth (FCG) rates have traditionally been formulated from fracture mechanics, whereas fatigue crack initiation has been empirically described using stress-life or strain-life methods. More recently, there has been efforts towards the use of the local stress-strain and similitude concepts to formulate fatigue crack growth rates. A new model has been developed which derives stress-life, strain-life and fatigue crack growth rates from strain energy density concepts. This new model has the advantage to predict an intrinsic stress ratio effect of the form ?ar=(?amp)?·(?max )(1-?), which is dependent on the cyclic stress-strain behaviour of the material. This new fatigue crack propagation model was proposed by Huffman based on Walkerlike strain-life relation. This model is applied to FCG data available for the P355NL1 pressure vessel steel. A comparison of the experimental results and the Huffman crack propagation model is made

    Over-expression of AhR (aryl hydrocarbon receptor) induces neural differentiation of Neuro2a cells: neurotoxicology study

    BACKGROUND: Dioxins and related compounds are suspected of causing neurological disruption in human and experimental animal offspring following perinatal exposure during development and growth. The molecular mechanism(s) of the actions in the brain, however, have not been fully investigated. A major participant in the process of the dioxin-toxicity is the dioxin receptor, namely the aryl hydrocarbon receptor (AhR). AhR regulates the transcription of diverse genes through binding to the xenobiotic-responsive element (XRE). Since the AhR has also been detected in various regions of the brain, the AhR may play a key role in the developmental neurotoxicity of dioxins. This study focused on the effect of AhR activation in the developing neuron. METHODS: The influence of the AhR on the developing neuron was assessed using the Neuro2a-AhR transfectant. The undifferentiated murine neuroblastoma Neuro2a cell line (ATCC) was stably transfected with AhR cDNA and the established cell line was named N2a-Rα. The activation of exogenous AhR in N2a-Rα cells was confirmed using RNAi, with si-AhR suppressing the expression of exogenous AhR. The neurological properties of N2a-Rα based on AhR activation were evaluated by immunohistochemical analysis of cytoskeletal molecules and by RT-PCR analysis of mRNA expression of neurotransmitter-production related molecules, such as tyrosine hydroxylase (TH). RESULTS: N2a-Rα cells exhibited constant activation of the exogenous AhR. CYP1A1, a typical XRE-regulated gene, mRNA was induced without the application of ligand to the culture medium. N2a-Rα cells exhibited two significant functional features. Morphologically, N2a-Rα cells bore spontaneous neurites exhibiting axon-like properties with the localization of NF-H. In addition, cdc42 expression was increased in comparison to the control cell line. The other is the catecholaminergic neuron-like property. N2a-Rα cells expressed tyrosine hydroxylase (TH) mRNA as a functional marker of catecholaminergic neurotransmitter production. Thus, exogenous AhR induced catecholaminergic differentiation in N2a-Rα cells. CONCLUSION: The excessive activation of AhR resulted in neural differentiation of Neuro2a cells. This result revealed that dioxins may affect the nervous system through the AhR-signaling pathway. Activated AhR may disrupt the strictly regulated brain formation with irregular differentiation occurring rather than cell death

    Dental Health and Mortality in People With End-Stage Kidney Disease Treated With Hemodialysis: A Multinational Cohort Study

    Background Dental disease is more extensive in adults with chronic kidney disease, but whether dental health and behaviors are associated with survival in the setting of hemodialysis is unknown. Study Design Prospective multinational cohort. Setting & Participants 4,205 adults treated with long-term hemodialysis, 2010 to 2012 (Oral Diseases in Hemodialysis [ORAL-D] Study). Predictors Dental health as assessed by a standardized dental examination using World Health Organization guidelines and personal oral care, including edentulousness; decayed, missing, and filled teeth index; teeth brushing and flossing; and dental health consultation. Outcomes All-cause and cardiovascular mortality at 12 months after dental assessment. Measurements Multivariable-adjusted Cox proportional hazards regression models fitted with shared frailty to account for clustering of mortality risk within countries. Results During a mean follow-up of 22.1 months, 942 deaths occurred, including 477 cardiovascular deaths. Edentulousness (adjusted HR, 1.29; 95% CI, 1.10-1.51) and decayed, missing, or filled teeth score ≥ 14 (adjusted HR, 1.70; 95% CI, 1.33-2.17) were associated with early all-cause mortality, while dental flossing, using mouthwash, brushing teeth daily, spending at least 2 minutes on oral hygiene daily, changing a toothbrush at least every 3 months, and visiting a dentist within the past 6 months (adjusted HRs of 0.52 [95% CI, 0.32-0.85], 0.79 [95% CI, 0.64-0.97], 0.76 [95% CI, 0.58-0.99], 0.84 [95% CI, 0.71-0.99], 0.79 [95% CI, 0.65-0.95], and 0.79 [95% CI, 0.65-0.96], respectively) were associated with better survival. Results for cardiovascular mortality were similar. Limitations Convenience sample of clinics. Conclusions In adults treated with hemodialysis, poorer dental health was associated with early death, whereas preventive dental health practices were associated with longer survival

    Analysis of Structural Glass Panels under Impact Loading using Operational Modal Analysis

    The latest technology and architectural trends have significantly improved the use of a large variety of glass products in construction which, in function of their own characteristics, allow to design and calculate structural glass elements under safety conditions. Both monolithic as well as laminated glass can be used for these proposals. In the case of laminated elements interlayers of polymers are introduced between the monolithic glass layers so that an improvement of some properties, such as damping or safety, is achieved. In this paper, glass panels are tested under soft impact loading (using the European standard UNE-EN 12600:2003 specifications) to analyse its dynamic response. With this proposal, operational modal analysis is used to identify the modal parameters of the panels and operational deflection shapes are obtained during the impact tests. Both data are used to assembly and validate a numerical model of the impact loading tests that allows the stresses estimation in the glass panels

    Unified two-stage fatigue methodology based on a probabilistic damage model applied to structural details

    Fatigue cracks are often a result of incremental stable crack growths that can be described by fracture mechanics. Fatigue crack initiation, however, has been most often described using empirical relationships between an alternating stress or strain and the number of cycles at those loads until either a specimen fails or a crack is observed. There are, however, modern views that consider fatigue crack growth and crack initiation directly related phenomena, the former being repeated instances of the latter, localized at an existing crack tip. Models of this newer class allow for an integrated initiation and crack growth calculation process which is advantageous when dealing with notched details in elastoplastic strain conditions. Recently, Huffman developed such a model which relates Walker-like stress-life relation and fatigue crack growth behaviour. This paper expands upon this new approach in order to account for the expected statistical variation using probabilistic S-N and ε-N fields as per works of Castillo and Fernandez-Canteli. A unified local approach proposed to treat fatigue crack initiation and growth is used on notched geometries with the predicted probabilistic variations being accounted. Notched details made of puddle iron from the Eiffel bridge and P355NL1 pressure vessel steel are considered in this study
