Based on these reasons, many researchers try to develop an anti-diabetic drugs derived from natural resources.One of the plants which potential to decrease glucose level in diabetic patients is Cinnamon (Cinnamomun zeylanicum). Cinnamon extract is very potent to decreases the glucose level in diabetic conditions. Based on this, the cinnamon extract will formulate in softgel forms. The manufacturing process occurs by encapsulate the extract with gelatin film as a shell material. Gelatin is a chewy shell material. The addition of plasticizer in gelatin is very important because its can affected to flexibility of capsule. The encapsulation process with rotary die method occurs using a special machine. The encapsulation process occurs by create a contract manufacture with third person. To ensure theproduct quality, a various test like organoleptic, weight-content uniformity, and rupture test was conducted.Penyakit serius yang terjadi ketika pankreas tidak dapat menghasilkan hormon insulin yang cukup ialah diabetes. Berbagai macam komplikasi yang berdampak pada mata, jantung dan sistem saraf disebabkan oleh diabetes. Penggunaan obat sintetik baik oral maupun injeksi dapat menyebabkan efek samping serius berupa hipoglikemi. Berdasarkan hal ini, banyak peneliti mencoba untuk mengembangkan obat anti-diabet dari bahan alam. Salah satu jenis tanaman yang berpotensi dalam menurunkan kadar glukosa pada pasien diabetes ialah kayu manis (Cinnamomum zeylanicum). Ekstrak dari kayu manis sangat berpotensi menurunkan kadar glukosa pada kondisi diabetes. Berdasarkan hal ini, ekstrak kayu manis akan diformulasikan dalam bentuk kapsul lunak.Proses pembuatan dilakukan dengan cara enkapsulasi ekstrak terstandar pada lembaran polimer gelatin sebagai cangkang. Gelatin merupakan polimer cangkang yang memiliki sifat kenyal.Penambahan bahan plasticizer pada gelatin sangat penting karena dapat mempengaruhi fleksibilitas kapsul. Proses enkaspulasi menggunakan metode rotary die dengan menggunakan mesin khusus.Proses enkapsulasi dilakukan dengan mengadakan kontrak pembuatan kepada pihak ketiga.     Proposing a Solution for a Self-Managed Data-Ecosystem in Production: Use-Case-Driven IT-OT-Integration with an Event-Driven IT-Architecture

    With the development of publicly accessible broker systems within the last decade, the complexity of data-driven ecosystems is expected to become manageable for self-managed digitalisation. Having identified event-driven IT-architectures as a suitable solution for the architectural requirements of Industry 4.0, the producing industry is now offered a relevant alternative to prominent third-party ecosystems. Although the technical components are readily available, the realisation of an event-driven IT-architecture in production is often hindered by a lack of reference projects, and hence uncertainty about its success and risks. The research institute FIR and IT-expert synyx are thus developing an event-driven IT-architecture in the Center Smart Logistics' producing factory, which is designed to be a multi-agent testbed for members of the cluster. With the experience gained in industrial projects, a target IT-architecture was conceptualised that proposes a solution for a self-managed data-ecosystem based on open-source technologies. With the iterative integration of factory-relevant Industry 4.0 use cases, the target is continuously realised and validated. The paper presents the developed solution for a self-managed event-driven IT-architecture and presents the implications of the decisions made. Furthermore, the progress of two use cases, namely an IT-OT-integration and a smart product demonstrator for the research project BlueSAM, are presented to highlight the iterative technical implementability and merits, enabled by the architecture

    Die deutsche Wirtschaft wird im Prognosezeitraum deutlich schwächer wachsen als in den vergangenen zwei Jahren. Nach drei Prozent 2011 dürfte die Produktion in diesem Jahr nur noch um ein Prozent zunehmen, 2013 werden es knapp zwei Prozent sein. Die Expansion in Deutschland ist in erster Linie durch die binnenwirtschaftliche Dynamik getrieben. Erst ab der Jahreswende dürften auch die Exporte wieder stärker zulegen. Die zunächst schwächere Konjunktur wird vorübergehend wohl auch zu einem Rückgang der Beschäftigung führen; die Arbeitslosenquote bleibt mit knapp sieben Prozent in diesem und im nächsten Jahr in jahresdurchschnittlicher Betrachtung aber stabil

    Structural insights into Cullin4-RING ubiquitin ligase remodelling by Vpr from simian immunodeficiency viruses

    Viruses have evolved means to manipulate the host's ubiquitin-proteasome system, in order to down-regulate antiviral host factors. The Vpx/Vpr family of lentiviral accessory proteins usurp the substrate receptor DCAF1 of host Cullin4-RING ligases (CRL4), a family of modular ubiquitin ligases involved in DNA replication, DNA repair and cell cycle regulation. CRL4DCAF1 specificity modulation by Vpx and Vpr from certain simian immunodeficiency viruses (SIV) leads to recruitment, poly-ubiquitylation and subsequent proteasomal degradation of the host restriction factor SAMHD1, resulting in enhanced virus replication in differentiated cells. To unravel the mechanism of SIV Vpr-induced SAMHD1 ubiquitylation, we conducted integrative biochemical and structural analyses of the Vpr protein from SIVs infecting Cercopithecus cephus (SIVmus). X-ray crystallography reveals commonalities between SIVmus Vpr and other members of the Vpx/Vpr family with regard to DCAF1 interaction, while cryo-electron microscopy and cross-linking mass spectrometry highlight a divergent molecular mechanism of SAMHD1 recruitment. In addition, these studies demonstrate how SIVmus Vpr exploits the dynamic architecture of the multi-subunit CRL4DCAF1 assembly to optimise SAMHD1 ubiquitylation. Together, the present work provides detailed molecular insight into variability and species-specificity of the evolutionary arms race between host SAMHD1 restriction and lentiviral counteraction through Vpx/Vpr proteins

    Thymosin beta 4 Improves Differentiation and Vascularization of EHTs

    Induced pluripotent stem cells (iPSC) constitute a powerful tool to study cardiac physiology and represents a promising treatment strategy to tackle cardiac disease. However, iPSCs remain relatively immature after differentiation. Additionally, engineered heart tissue (EHT) has been investigated as a therapy option in preclinical disease models with promising results, although their vascularization and functionality leave room for improvement. Thymosin beta 4 (T beta 4) has been shown to promote the differentiation of progenitor cell lines to cardiomyocytes while it also induces angiogenic sprouting and vascular maturation. We examined the potential impact of T beta 4 to enhance maturation of cardiomyocytes from iPSCs. Assessing the expression of transcription factors associated with cardiac differentiation, we were able to demonstrate the increased generation of cells displaying cardiomyocyte characteristics in vitro. Furthermore, we demonstrated, in a zebrafish model of embryonic vascular development, that T beta 4 is crucial for the proper execution of lymphatic and angiogenic vessel sprouting. Finally, utilizing T beta 4-transduced EHTs generated from mice genetically engineered to label endothelial cells in vitro, we show that treatment with T beta 4 promotes vascularization and contractility in EHTs, highlighting T beta 4 as a growth factor improving the formation of cardiomyocytes from iPSC and enhancing the performance of EHTs generated from neonatal cardiomyocytes
