    Diagnostics of the tropical tropopause layer from in-situ observations and CCM data

    A suite of diagnostics is applied to in-situ aircraft measurements and one Chemistry-Climate Model (CCM) data to characterize the vertical structure of the Tropical Tropopause Layer (TTL). The diagnostics are based on vertical tracer profiles and relative vertical tracer gradients, using tropopause-referenced coordinates, and tracer-tracer relationships in the tropical Upper Troposphere/Lower Stratosphere (UT/LS). Observations were obtained during four tropical campaigns performed from 1999 to 2006 with the research aircraft Geophysica and have been compared to the output of the ECHAM5/MESSy CCM. The model vertical resolution in the TTL (~500 m) allows for appropriate comparison with high-resolution aircraft observations and the diagnostics used highlight common TTL features between the model and the observational data. The analysis of the vertical profiles of water vapour, ozone, and nitrous oxide, in both the observations and the model, shows that concentration mixing ratios exhibit a strong gradient change across the tropical tropopause, due to the role of this latter as a transport barrier and that transition between the tropospheric and stratospheric regimes occurs within a finite layer. The use of relative vertical ozone and carbon monoxide gradients, in addition to the vertical profiles, helps to highlight the region where this transition occurs and allows to give an estimate of its thickness. The analysis of the CO-O3 and H2O-O3 scatter plots and of the Probability Distribution Function (PDF) of the H2O-O3 pair completes this picture as it allows to better distinguish tropospheric and stratospheric regimes that can be identified by their different chemical composition. The joint analysis and comparison of observed and modelled data allows to state that the model can represent the background TTL structure and its seasonal variability rather accurately. The model estimate of the thickness of the interface region between tropospheric and stratospheric regimes agrees well with average values inferred from observations. On the other hand, the measurements can be influenced by regional scale variability, local transport processes as well as deep convection, that can not be captured by the model

    Comet Machholz (C/2004 Q2): morphological structures in the inner coma and rotation parameters

    Extensive observations of comet C/2004 Q2 (Machholz) were carried out between August 2004 and May 2005. The images obtained were used to investigate the comet's inner coma features at resolutions between 350 and 1500 km/pixel. A photometric analysis of the dust outflowing from the comet's nucleus and the study of the motion of the morphological structures in the inner coma indicated that the rotation period of the nucleus was most likely around 0.74 days. A thorough investigation of the inner coma morphology allowed us to observe two main active sources on the comet's nucleus, at a latitude of +85{\deg} \pm 5{\deg} and +45{\deg} \pm 5{\deg}, respectively. Further sources have been observed, but their activity ran out quite rapidly over time; the most relevant was at latcom. = 25{\deg} \pm 5{\deg}. Graphic simulations of the geometrical conditions of observation of the inner coma were compared with the images and used to determine a pole orientation at RA=95{\deg} \pm 5{\deg}, Dec=+35{\deg} \pm 5{\deg}.     Procalcitonin Reveals Early Dehiscence in Colorectal Surgery: The PREDICS Study

    Objectives: We designed a multicentric, observational study to test if Procalcitonin (PCT) might be an early and reliable marker of anastomotic leak (AL) after colorectal surgery (ClinicalTrials.govIdentifier:NCT01817647). Background: Procalcitonin is a biomarker used to monitor bacterial infections and guide antibiotic therapy. Anastomotic leak after colorectal surgery is a severe complication associated with relevant short and long-term sequelae. Methods: Between January 2013 and September 2014, 504 patients underwent colorectal surgery, for malignant colorectal diseases, in elective setting. White blood count (WBC), C-reactive protein (CRP) and PCT levels were measured in 3rd and 5th postoperative day (POD). AL and all postoperative complications were recorded. Results: We registered 28 (5.6%) anastomotic leaks. Specificity and negative predictive value for AL with PCT less than 2.7 and 2.3 ng/mL were, respectively, 91.7% and 96.9% in 3rd POD and 93% and 98.3% in 5th POD. Receiver operating characteristic curve for biomarkers shows that in 3rd POD, PCTand CRP have similar area under the curve (AUC) (0.775 vs 0.772), both better than WBC (0.601); in 5th POD, PCT has a better AUC than CRP and WBC (0.862 vs 0.806 vs 0.611). Measuring together PCT and CRP significantly improves AL diagnosis in 5th POD (AUC: 0.901). Conclusions: PCTand CRP demonstrated to have a good negative predictive value for AL, both in 3rd and in 5th POD. Low levels of PCT, together with low CRP values, seem to be early and reliable markers of AL after colorectal surgery. These biomarkers might be safely added as additional criteria of discharge protocols after colorectal surgery

    Guía de bienes patrimoniales de Chacras de Coria: Un paisaje con identidad

    La presente Guía es el resultado del trabajo conjunto entre un grupo de vecinos/as y distintos/as especialistas formados en patrimonio, por lo que recoge diversas voces y miradas cuyo tema en común es el compromiso y sensibilización en relación con los bienes tangibles e intangibles del distrito de Chacras de Coria en Luján de Cuyo, Mendoza. Los/as vecinos/as autoconvocados se reúnen periódicamente para intentar hacer frente, por medio de distintas acciones, a las transformaciones urbanas que consideran atentan contra su forma de vida y contra la experiencia de los/ as visitantes quienes gozan del entorno, la infraestructura y la historia de este lugar. Entre estas transformaciones se destacan la densificación acelerada mediante emprendimientos que no respetan las determinaciones del Código de Edificación, como son el F.O.S. (Factor de Ocupación del Suelo ) y el F.O.T. (Factor de Ocupación Total); la demolición de construcciones antiguas por falta de su reconocimiento y protección en el centro histórico; el proyecto impetuoso de ensanche de la Ruta Provincial n°82 (que afortunadamente se modificó debido a los reclamos); las excepciones municipales que permiten diversos proyectos que son implementados sin criterio y que atentan a la conformación urbana histórica del centro de Chacras de Coria y sus alrededores; la depredación forestal constante, entre otros. En los últimos meses, el descontento frente a estos cambios, ha llevado a realizar notas a distintos organismos, reclamos, marchas, reuniones, etc. Una suerte de movilización vecinal que sigue de cerca los nuevos proyectos y las transformaciones urbanas que no están acordes a lo que el grupo vecinal considera el espíritu del lugar.Fil: Esteves, Matias Jose.     Stable S/MAR-based episomal vectors are regulated at the chromatin level

    Episomal vectors assembled from defined genetic components are a promising alternative to traditional gene therapy vectors that integrate in the host genome and may cause insertional mutations. The vector pEPI-eGFP is stably retained in the episomal state in cultured mammalian cells at low copy number for many generations without integration into the host genome. Although pEPI-eGFP is a fully engineered vector, little is known about how it interacts with the host genome and about the molecular mechanisms that are responsible for its transcriptional activity. We have analyzed the expression of the episomal reporter gene eGFP under conditions that affect the chromatin state of the genome. We have also constructed pEPI derivatives carrying a tandem array of lac operator sequences, which allows in vivo visualization and manipulation of the chromatin state of the episome. We show that changes in chromatin state of both the host and pEPI-eGFP induces changes in episomal gene activity and influences the episome’s nuclear distributions. We conclude that episomal genes are subject to control systems of the host, similarly to their counterparts in the host genome
