    CCL13/MCP-4 belongs to the CC chemokine family, which induces chemotaxis in many immune cells. Despite extensive research into its function in numerous disorders, a thorough analysis of CCL13 is not yet accessible. The role of CCL13 in human disorders and existing CCL13-focused therapies are outlined in this study. The function of CCL13 in rheumatic diseases, skin conditions, and cancer is comparatively well-established, and some studies also suggest that it may be involved in ocular disorders, orthopedic conditions, nasal polyps, and obesity. We also give an overview of research that found very little evidence of CCL13 in HIV, nephritis, and multiple sclerosis. Even though CCL13-mediated inflammation is frequently linked to disease pathogenesis, it’s fascinating to note that in some conditions, like primary biliary cholangitis (PBC) and suicide, it might even act as a preventative measure

    Comparison of bleeding risk and hypofibrinogenemia-associated risk factors between tigecycline with cefoperazone/sulbactam therapy and other tigecycline-based combination therapies

    Background: Tigecycline and cefoperazone/sulbactam can cause coagulation disorders; tigecycline may also lead to hypofibrinogenemia, raising safety concerns. This study aimed to investigate whether tigecycline plus cefoperazone/sulbactam increases the risk of bleeding compared with other tigecycline-based combination therapies and identify risk factors for tigecycline-associated hypofibrinogenemia.Methods: In this multi-method, multicenter, retrospective study, coagulation and other baseline variables were compared using a cohort study, and risk factors for hypofibrinogenemia using a case-control study.Results: The 451 enrolled participants were divided into three group: tigecycline plus cefoperazone/sulbactam (Group A, 193 patients), tigecycline plus carbapenems (Group B, 200 patients) and tigecycline plus β-lactams without N-methylthio-tetrazole (NMTT) side chains (Group C, 58 patients). Activated partial thromboplastin time and prothrombin time were prolonged, and fibrinogen declined for all patients after tigecycline-based medication (all p < 0.05). Prothrombin time in Group B was significantly longer than in other groups (p < 0.05), but there were no significant differences in bleeding events between the three groups (p = 0.845). Age greater than 80 years (OR: 2.85, 95% CI: 1.07–7.60), treatment duration (OR: 1.29, 95% CI: 1.19–1.41), daily dose (OR: 2.6, 95% CI: 1.29–5.25), total bilirubin (OR: 1.01, 95% CI: 1.01–1.02) and basal fibrinogen (OR: 1.32, 95% CI: 1.14–1.63) were independent risk factors of hypofibrinogenemia. The optimal cut-off for treatment course was 6 days for high-dose and 11 days for low-dose.Conclusion: Tigecycline plus cefoperazone/sulbactam did not increase the risk of bleeding compared with tigecycline plus carbapenem, or tigecycline plus β-lactam antibiotics without NMTT-side-chains. Coagulation function should be closely monitored in patients receiving tigecycline treatment

    Metric Nearness Made Practical

    Given a square matrix with noisy dissimilarity measures between pairs of data samples, the metric nearness model computes the best approximation of the matrix from a set of valid distance metrics. Despite its wide applications in machine learning and data processing tasks, the model faces non-trivial computational requirements in seeking the solution due to the large number of metric constraints associated with the feasible region. Our work designed a practical approach in two stages to tackle the challenge and improve the model's scalability and applicability. The first stage computes a fast yet high-quality approximate solution from a set of isometrically embeddable metrics, further improved by an effective heuristic. The second stage refines the approximate solution with the Halpern-Lions-Wittmann-Bauschke projection algorithm, which converges quickly to the optimal solution. In empirical evaluations, the proposed approach runs at least an order of magnitude faster than the state-of-the-art solutions, with significantly improved scalability, complete conformity to constraints, less memory consumption, and other desirable features in real applications

    How Do Subway Signs Affect Pedestrians’ Wayfinding Behavior through Visual Short-Term Memory?

    Recently, subways have become an important part of public transportation and have developed rapidly in China. In the subway station setting, pedestrians mainly rely on visual short-term memory to obtain information on how to travel. This research aimed to explore the short-term memory capacities and the difference in short-term memory for different information for Chinese passengers regarding subway signs. Previous research has shown that people’s general short-term memory capacity is approximately four objects and that, the more complex the information, the lower people’s memory capacity. However, research on the short-term memory characteristics of pedestrians for subway signs is scarce. Hence, based on the STM theory and using 32 subway signs as stimuli, we recruited 120 subjects to conduct a cognitive test. The results showed that passengers had a different memory accuracy for different types of information in the signs. They were more accurate regarding line number and arrow, followed by location/text information, logos, and orientation. Meanwhile, information type, quantity, and complexity had significant effects on pedestrians’ short-term memory capacity. Finally, according to our results that outline the characteristics of short-term memory for subway signs, we put forward some suggestions for subway signs. The findings will be effective in helping designers and managers improve the quality of subway station services as well as promoting the development of pedestrian traffic in such a setting

    Heat Source Forecast of Ball Screw Drive System Under Actual Working Conditions Based on On-Line Measurement of Temperature Sensors

    In view of the time-varying complexity of the heat source for the ball screw feed system, this paper proposes an adaptive inverse problem-solving method to estimate the time-varying heat source and temperature field of the feed system under working conditions. The feed system includes multiple heat sources, and the rapid change of the moving heat source increases the difficulty of its identification. This paper attempts to develop a numerical calculation method for identifying the heat source by combining the experiment with the optimization algorithm. Firstly, based on the theory of heat transfer, a new dynamic thermal network model was proposed. The temperature data signal and the position signal of the moving nut captured by the sensors are used as input to optimize the solution of the time-varying heat source. Then, based on the data obtained from the experiment, finite element software parametric programming was used to optimize the estimate of the heat source, and the results of the two heat source prediction methods are compared and verified. The other measured temperature points obtained by the experiment were used to compare and verify the inverse method of this numerical calculation, which illustrates the reliability and advantages of the dynamic thermal network combined with the genetic algorithm for the inverse method. The method based on the on-line monitoring of temperature sensors proposed in this paper has a strong application value for heat source and temperature field estimation of complex mechanical structures

    Orthrozanclus elongata n. sp. and the significance of sclerite-covered taxa for early trochozoan evolution

    Orthrozanclus is a shell-bearing, sclerite covered Cambrian organism of uncertain taxonomic affinity, seemingly representing an intermediate between its fellow problematica Wiwaxia and Halkieria. Attempts to group these slug-like taxa into a single ‘halwaxiid’ clade nevertheless present structural and evolutionary difficulties. Here we report a new species of Orthrozanclus from the early Cambrian Chengjiang Lagerstätte. The scleritome arrangement and constitution in this material corroborates the link between Orthrozanclus and Halkieria, but not with Wiwaxia — and calls into question its purported relationship with molluscs. Instead, the tripartite construction of the halkieriid scleritome finds a more compelling parallel in the camenellan tommotiids, relatives of the brachiopods and phoronids. Such a phylogenetic position would indicate the presence of a scleritome in the common ancestor of the three major trochozoan lineages, Mollusca, Annelida and Brachiozoa. On this view, the absence of fossil Ediacaran sclerites is evidence against any ‘Precambrian prelude’ to the explosive diversification of these phyla in the Cambrian, c. 540–530 million years ago

    A Network Pharmacology to Explore the Mechanism of Calculus Bovis in the Treatment of Ischemic Stroke

    Background. Calculus Bovis is a valuable Chinese medicine, which is widely used in the clinical treatment of ischemic stroke. The present study is aimed at investigating its target and the mechanism involved in ischemic stroke treatment by network pharmacology. Methods. Effective compounds of Calculus Bovis were collected using methods of network pharmacology and using the Bioinformatics Analysis Tool for Molecular Mechanism of Traditional Chinese Medicine (BATMAN-TCM) and the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). Potential compound targets were searched in the TCMSP and SwissTargetPrediction databases. Ischemic stroke-related disease targets were searched in the Drugbank, DisGeNet, OMIM, and TTD databases. These two types of targets were uploaded to the STRING database, and a network of their interaction (PPI) was built with its characteristics calculated, aiming to reveal a number of key targets. Hub genes were selected using a plug-in of the Cytoscape software, and Gene Ontology (GO) biological processes and pathway enrichment analyses of Kyoto Encyclopedia of Genes and Genomes (KEGG) were conducted using the clusterProfiler package of R language. Results. Among 12 compounds, deoxycorticosterone, methyl cholate, and biliverdin were potentially effective components. A total of 344 Calculus Bovis compound targets and 590 ischemic stroke targets were found with 92 overlapping targets, including hub genes such as TP53, AKT, PIK2CA, MAPK3, MMP9, and MMP2. Biological functions of Calculus Bovis are associated with protein hydrolyzation, phosphorylation of serine/threonine residues of protein substrates, peptide bond hydrolyzation of peptides and proteins, hydrolyzation of intracellular second messengers, antioxidation and reduction, RNA transcription, and other biological processes. Conclusion. Calculus Bovis may play a role in ischemic stroke by activating PI3K-AKT and MAPK signaling pathways, which are involved in regulating inflammatory response, cell apoptosis, and proliferation

    Physiological indices and driving performance of drivers at tunnel entrances and exits: A simulated driving study.

    The entrance and exit sections of a tunnel are the accident black-spots in an expressway. For a safe operation of road tunnels, it is necessary to understand a driver's physiological indices and driving performance when driving through tunnels. In this study, the UC-Win/Road simulation software was used to build 12 tunnel models of different lengths. A simulated driving experiment was carried out in a 6-DoF motion platform. The lateral position of vehicles characterizing the driving performance was measured using the motion platform. Electrocardiogram and eye movement data of 25 recruited drivers were collected simultaneously through the experiment. The spatial changes in a driver's heart rate (HR) growth rate, RMSSD, pupil diameter growth rate and vehicle lateral deviation within 300 m before and after the tunnel entrance and exit were analyzed to determine the variation rules in the different tunnels. The study identified the length range in the tunnel entrance and exit sections that influences the drivers. A quantitative analysis was further carried out to analyze the relationship between the physiological indices and the driving performance indicator. The results showed that a driver's heart rate fluctuates significantly 250 m before the tunnel entrance and 50 m before the exit. In this region, the pupil diameter increases gradually, and drivers tend to shift the vehicle to the left. At the tunnel exit, the HR and RMSSD are affected significantly by the tunnel length, and the variation is higher in longer tunnels. In comparison, the tunnel length has no significant effect on the physiological indicators and driving performance of the drivers at the entrance and exit
