    Glucosamine increases hyaluronic acid production in human osteoarthritic synovium explants

    Background. Glucosamine (GlcN) used by patients with osteoarthritis was demonstrated to reduce pain, but the working mechanism is still not clear. Viscosupplementation with hyaluronic acid (HA) is also described to reduce pain in osteoarthritis. The synthesis of HA requires GlcN as one of its main building blocks. We therefore hypothesized that addition of GlcN might increase HA production by synovium tissue. Methods. Human osteoarthritic synovium explants were obtained at total knee surgery and pre-cultured for 1 day. The experimental conditions consisted of a 2 days continuation of the culture with addition of N-Acetyl-glucosamine (GlcN-Ac; 5 mM), glucosamine-hydrochloride (GlcN-HCl; 0.5 and 5 mM), glucose (Gluc; 0.5 and 5 mM). Hereafter HA production was measured in culture medium supernatant using an enzyme-linked binding protein assay.     Prevalence and risk factors for internet gaming disorder

    Objectives: To estimate the prevalence of internet gaming disorder (IGD) and associated risk factors in a sample of secondary and postsecondary students from a public federal institution of higher education (Instituto Federal de Educação, Ciência e Tecnologia) in Southern Brazil. Methods: The study included a sociodemographic questionnaire, the Beck Depression Inventory (BDI), Self-Report Questionnaire (SRQ-20), Pittsburgh Sleep Quality Index (PSQI-BR), the Mini-Social Phobia Inventory (Mini-SPIN), and the Game Addiction Scale (GAS). Finally, IGD was measured with the Brazilian version of the Internet Gaming Disorder Scale-Short-Form (IGDS9-SF), which has been psychometrically validated in this population. Results: Overall, 38.2% (n=212) of the sample exhibited IGD symptoms, with 18.2% (n=101) being classed as at-risk gamers. Regression analysis found IGD to be associated with male gender, severe depressive symptoms, poor sleep quality, increased time spent gaming, and total free time spent gaming (p < 0.001). Conclusions: The prevalence of IGD in this sample was relatively high, and associated risk factors found were similar to those previously reported in the literature. Further studies investigating the epidemiology of IGD in Brazilian samples are warranted to better understand treatment needs and inform preventive measures in this population

    Radiation chemistry of solid-state carbohydrates using EMR

    We review our research of the past decade towards identification of radiation-induced radicals in solid state sugars and sugar phosphates. Detailed models of the radical structures are obtained by combining EPR and ENDOR experiments with DFT calculations of g and proton HF tensors, with agreement in their anisotropy serving as most important criterion. Symmetry-related and Schonland ambiguities, which may hamper such identification, are reviewed. Thermally induced transformations of initial radiation damage into more stable radicals can also be monitored in the EPR (and ENDOR) experiments and in principle provide information on stable radical formation mechanisms. Thermal annealing experi-ments reveal, however, that radical recombination and/or diamagnetic radiation damage is also quite important. Analysis strategies are illustrated with research on sucrose. Results on dipotassium glucose-1-phosphate and trehalose dihydrate, fructose and sorbose are also briefly discussed. Our study demonstrates that radiation damage is strongly regio-selective and that certain general principles govern the stable radical formation

    In this review article, we have presented for the first time the new applications of supercapacitor technologies and working principles of the family of RuO2-carbon-based nanofiller-reinforced conducting polymer nanocomposites. Our review focuses on pseudocapacitors and symmetric and asymmetric supercapacitors. Over the last years, the supercapacitors as a new technology in energy storage systems have attracted more and more attention. They have some unique characteristics such as fast charge/discharge capability, high energy and power densities, and long stability. However, the need for economic, compatible, and easy synthesis materials for supercapacitors have led to the development of RuO2-carbon-based nanofiller-reinforced conducting polymer nanocomposites with RuO2.     Prognostic model to predict postoperative acute kidney injury in patients undergoing major gastrointestinal surgery based on a national prospective observational cohort study.

    Background: Acute illness, existing co-morbidities and surgical stress response can all contribute to postoperative acute kidney injury (AKI) in patients undergoing major gastrointestinal surgery. The aim of this study was prospectively to develop a pragmatic prognostic model to stratify patients according to risk of developing AKI after major gastrointestinal surgery. Methods: This prospective multicentre cohort study included consecutive adults undergoing elective or emergency gastrointestinal resection, liver resection or stoma reversal in 2-week blocks over a continuous 3-month period. The primary outcome was the rate of AKI within 7 days of surgery. Bootstrap stability was used to select clinically plausible risk factors into the model. Internal model validation was carried out by bootstrap validation. Results: A total of 4544 patients were included across 173 centres in the UK and Ireland. The overall rate of AKI was 14·2 per cent (646 of 4544) and the 30-day mortality rate was 1·8 per cent (84 of 4544). Stage 1 AKI was significantly associated with 30-day mortality (unadjusted odds ratio 7·61, 95 per cent c.i. 4·49 to 12·90; P < 0·001), with increasing odds of death with each AKI stage. Six variables were selected for inclusion in the prognostic model: age, sex, ASA grade, preoperative estimated glomerular filtration rate, planned open surgery and preoperative use of either an angiotensin-converting enzyme inhibitor or an angiotensin receptor blocker. Internal validation demonstrated good model discrimination (c-statistic 0·65). Discussion: Following major gastrointestinal surgery, AKI occurred in one in seven patients. This preoperative prognostic model identified patients at high risk of postoperative AKI. Validation in an independent data set is required to ensure generalizability

However, fragmentation of forests can lead to the collapse of such a structure, especially when natural thinning of the dominant tree species is accompanied by other significant changes. This paper traces changes in the composition of the community of a forest of relevant ecological interest in Campinas (SP, Brazil) as that forest moves toward deterioration as indicated by a massive loss of individuals of fundamental species composing the canopy and subcanopy. The death of trees of the species traditionally forming the canopy, the result of natural processes, was probably intensified by anthropic factors, and led to a general destructuring of the community. The aim of this paper is to demonstrate that the dynamics of these fundamental species can be used as an indicator of the conservation of the ecosystem, as well as indicating the installation of a process which will lead to the collapse of the structure if adequate measures of adaptive management are not taken before the destructuring is irreversible.     Restinga and Lowland forests in coastal plain of southeastern Brazil: vegetation and environmental heterogeneity

    It was evaluated the floristic similarity between two Atlantic Rainforest physiognomies in Brazilian coast area, herein called Restinga and Lowland forests. The hypothesis was that, due the differences in geomorphologic processes, these forests would differ on soil physical and chemical properties, floristic composition, live above-ground biomass. and litterfall production. It was sampled 1 ha (100 x 100 m) for each site located in Ubatuba, Sao Paulo state, SE Brazil. Within each hectare it was recorded trees with DBH >4.8 cm in all 10 x 10 m contiguous plots, and collected soil and litterfall samples. The cluster and ordination analyses indicated the two communities as distinct groups considering soil and floristic composition, agreeing with the initial hypothesis. Species diversity was higher (p 0.05) between the two forests. This apparent paradox could be explained assuming that, since different species establish themselves in the Restinga or Lowland forests and find a favorable spectrum of conditions and resources, they would tend to persist and to develop in that place; even so the edaphic conditions differ between the Restinga and Lowland forests, each species could respond in a particular way to these variations, and then both forests could reach similar values of biomass and litterfall production. It is probable that the environmental filter conditioned by soils has being important for the strong floristic segregation between these two forests.11210312

    Polycistronic Herpesvirus Amplicon Vectors for Veterinary Vaccine Development

    Heterologous virus-vectored vaccines, particularly those based on canarypox virus vectors, have established a firm place in preventive veterinary medicine. However, herpesvirus-based vaccines have paved the way for DIVA vaccines (discrimination of infected against vaccinated animals), which are particularly desirable for highly contagious livestock diseases that are otherwise combatted by culling of infected animals.In this chapter, we describe the design, the preparation, and the testing of a polycistronic herpesvirus amplicon vaccine against rotaviruses with a particular emphasis on generating heterologous virus-like particles for immunization. After the design, the procedure consists of three steps, first, transient expression of the construct in cell cultures, second, expression and antibody response in a mouse model, and third, application of the system to the desired host species. As a whole, the present information will facilitate the design of novel vaccines of veterinary interest from the designing process until pre-licensing
