    Development of a stability-indicating high performance liquid chromatography method for assay of erythromycin ethylsuccinate in powder for oral suspension dosage form

    In this study an effective method was developed to assay erythromycin ethylsuccinate for an oral suspension dosage form. The chromatographic separation was achieved on an X-Terra[superscript ™] C[subscript 18] analytical column. A mixture of acetonitrile–ammonium dihydrogen phosphate buffer (0.025 mol L[superscript -1]) (60:40, V/V) (pH 7.0) was used as the mobile phase, effluent flow rate monitored at 1.0 mL min[superscript −1], and UV detection at 205 nm. In forced degradation studies, the effects of acid, base, oxidation, UV light and temperature were investigated showing no interference in the peak of drug. The proposed method was validated in terms of specificity, linearity, robustness, precision and accuracy. The method was linear at concentrations ranging from 400 to 600 μg mL[superscript −1], precise (intra- and inter-day relative standard deviations <0.65), accurate (mean recovery; 99.5%). The impurities and degradation products of erythromycin ethylsuccinate were selectively determined with good resolution in both the raw material and the final suspension forms. The method could be useful for both routine analytical and quality control assays of erythromycin ethylsuccinate in commercial powder for an oral suspension dosage form and it could be a very powerful tool to investigate the chemical stability of erythromycin ethylsuccinate.Chemi Darou Industrial Compan

    Effect of 8 Weeks Resistance Training and Irisin Injection on BDNF and Spatial Memory of Male Mice

    The purpose of this study was to compare the effect of eight weeks of resistance training and irisin injection on BDNF and spatial memory in male rats. 19 male mice (mean weight 18g and 5 weeks old) were randomly divided into control groups, sham, resistance training and irisin injection gruops. The resistance training program was conducted for eight weeks and three days each week. At the first week, the closed weights were 50% of the body weight of the rats and gradually increased to about 200% of their weight in the final week each session consisted of 3 repetitve 5 repetitions with 3 minutes of rest between turns and one minute between repetitions. Irisin injection was also performed three times a week. Elisa method was used to measure plasma irisin and BDNF and to evaluate spatial memory of Barns maze. Results showed a positive and significant correlation between irisin and BDNF (P = 0.66, P = 0.01). Also results showed irisin levels increased significantly in the training groups and irisin injection compared to the control group (P=0.001, F=14.25). This level was higher in the resistance training group than the irisin injection group, but the spatial memory (P=0.658, F=0.546) and BDNF levels (P=0.635, F=0.584) in the two groups did not have a significant effect. It can be concluded 8 weeks of resistance training and irisin injection did not have a significant effect on blood BDNF and spatial memory, but significantly increased the level of irisin

    Spinal vs General Anesthesia in patients undergoing urogenital surgery: A Randomized Clinical Trial

    Background: This study aimed to evaluate the pain in the 24 hours after surgery, quality of life, and the outcome of surgery in patients undergoing urogenital surgery by spinal and general anesthesia. Materials and Methods: Women referring a candidate for urogenital surgery in Vali-e-Asr Hospital entered the study after their informed consent; in one of the two study groups: Spinal Anesthesia (SA) vs. General Anesthesia (GA). The pain scores around the clock were measured using the Visual Analog Scale (VAS) at 2, 6, 12, and 24 hours postoperatively. Also, the two groups were compared regarding patient satisfaction at the time of ambulation. The surgery outcomes were measured using International Consultation on Incontinence Modular Questionnaires ICIQ. Data were entered and analyzed by SPSS software. Results: There was no significant relationship between parity, previous non-cesarean abdominal surgery, and urinary complications. However, there was a statistically significant difference between pain score in the two groups; while the postoperative days were not different in the two groups of anesthesia methods Conclusion: Considering the different influence of treatment methods for this disease, further research is needed to clarify, the results of anatomical, and anatomical outcomes after treatment for pelvic floor disorders in women

    Non-alcoholic fatty liver disease is not independent risk factor for cardiovascular disease event : a cohort study

    There are no consistent results between previous studies for an independent association between non-alcoholic fatty liver disease (NAFLD) and cardiovascular disease (CVD) events. To determine if there is an independent association between NAFLD and CVD events. In the present study, valid outcome data of 4808 subjects were available for phase 2 of our cohort study. These subjects had been followed up for seven years from phase 1, beginning in 2009-2010 to phase 2 during 2016-2017. Simple and multiple Cox proportional models were used to determine the association between NAFLD in the primary phase of the cohort and subsequent fatal and non-fatal CVD events during follow-up. The incidence of non-fatal CVD events in males with NAFLD was significantly higher ( = 0.004) than in males without NAFLD. A positive association was demonstrated between NAFLD and non-fatal CVD events in males (Hazard ratio = 1.606; 95%CI: 1.166-2.212; = 0.004) by the simple Cox proportional hazard model, but no independent association was detected between these in the multiple Cox models. No independent association was detected between NAFLD and CVD. It is likely that diabetes mellitus and age may be the principle mediators in this regard. [Abstract copyright: ©The Author(s) 2020.     Signal transduction pathway mutations in gastrointestinal (GI) cancers : a systematic review and meta-analysis

    The present study was conducted to evaluate the prevalence of the signaling pathways mutation rate in the Gastrointestinal (GI) tract cancers in a systematic review and meta-analysis study. The study was performed based on the PRISMA criteria. Random models by confidence interval (CI: 95%) were used to calculate the pooled estimate of prevalence via Metaprop command. The pooled prevalence indices of signal transduction pathway mutations in gastric cancer, liver cancer, colorectal cancer, and pancreatic cancer were 5% (95% CI: 3–8%), 12% (95% CI: 8–18%), 17% (95% CI: 14–20%), and 20% (95% CI: 5–41%), respectively. Also, the mutation rates for Wnt pathway and MAPK pathway were calculated to be 23% (95% CI, 14–33%) and 20% (95% CI, 17–24%), respectively. Moreover, the most popular genes were APC (in Wnt pathway), KRAS (in MAPK pathway) and PIK3CA (in PI3K pathway) in the colorectal cancer, pancreatic cancer, and gastric cancer while they were beta-catenin and CTNNB1 in liver cancer. The most altered pathway was Wnt pathway followed by the MAPK pathway. In addition, pancreatic cancer was found to be higher under the pressure of mutation compared with others based on pooled prevalence analysis. Finally, APC mutations in colorectal cancer, KRAS in gastric cancer, and pancreatic cancer were mostly associated gene alterations

    A population-based prospective study on obesity-related non-communicable diseases in northern Iran: rationale, study design, and baseline analysis

    BackgroundIran is facing an epidemiological transition with the increasing burden of non-communicable diseases, such as obesity-related disorders and cardiovascular diseases (CVDs). We conducted a population-based prospective study to assess the prevalence and incidence rates of CVDs and obesity-related metabolic disorders and to evaluate the predictive ability of various CVD risk assessment tools in an Iranian population.MethodWe enrolled 5,799 participants in Amol, a city in northern Iran, in 2009–2010 and carried out the first repeated measurement (RM) after seven years (2016–2017). For all participants, demographic, anthropometric, laboratory, hepatobiliary imaging, and electrocardiography data have been collected in the enrollment and the RM. After enrollment, all participants have been and will be followed up annually for 20 years, both actively and passively.ResultsWe adopted a multidisciplinary approach to overcome barriers to participation and achieved a 7-year follow-up success rate of 93.0% with an active follow-up of 5,394 participants aged 18–90 years. In the RM, about 64.0% of men and 81.2% of women were obese or overweight. In 2017, about 16.2% and 5.2% of men had moderate or severe non-alcoholic fatty liver disease, while women had a significantly higher prevalence of metabolic syndrome (35.9%), and type 2 diabetes mellitus (20.9%) than men. Of 160 deceased participants, 69 cases (43.1%) died due to CVDs over seven years.ConclusionThe most prevalent obesity-related chronic disease in the study was metabolic syndrome. Across the enrollment and RM phases, women exhibited a higher prevalence of obesity-related metabolic disorders. Focusing on obesity-related metabolic disorders in a population not represented previously and a multidisciplinary approach for enrolling and following up were the strengths of this study. The study outcomes offer an evidence base for future research and inform policies regarding non-communicable diseases in northern Iran

    Background and Purpose: Investigation of water quality is an important step for the suitable use of water resources in order to drinking and irrigation. Water quality affects agriculture programming. &nbsp;Hence the need of the study of water quality is strongly considered in the water resources management. Material and Methods: In this study Hydro-geochemical quality of ground water resources in the Ramian city -Golestan province has been studied for drinking and agriculture purpose. For this purpose, 15 qualitative characteristics of the 13 wells of Golestan province in two dry and wet seasons in 2011-2012 were analyzed by Aua Chem and Aq-qa. Results: The results showed that the ground water in the study area is classified in hard and very hard water. The original cations and anions in water are Ca2+> Mg2+> Na+ and HCO3-> Cl-> SO42-. Based on hydro-chemical diagram the dominant of water type is classified as Ca-HCO3. Salinity index of water indicated that more samples in two seasons are in the middle class. According to Schuler and Wilcox groundwater quality index, they are moderate suitable for agricultural and drinking consumption and in for agricultural purpose and 77% cases are in C3-S1 category. Conclusion: The results show that too much salt is one of the most important problems of water supply in the Ramian city for irrigation. This reduced plant growth or even stops the growth of some plant. If water resources in this area do not manage, after shortly time the soil will be suffered and polluted

    Urtica Dioica Distillate Regenerates Pancreatic Beta Cells in Streptozotocin-Induced Diabetic Rats

    Background: Urtica dioica is known as an anti-hyperglycemic plant. Urtica dioica distillate (UD) is a traditional Iranian drink, locally known as “aragh gazaneh”. In spite of its widespread consumption in Iran, according to traditional Iranian medicine, there is no scientific report on the usefulness of UD for diabetic patients. This survey was designed to evaluate its protective effects for the recovery from diabetes by determining the serum insulin, blood glucose, volume of pancreatic islets, and the number and volume of β-cells in diabetic rats. Methods: A total of 48 Sprague-Dawley male rats (200-250 g) were randomly distributed into 6 groups (n=8), including non-diabetic plus distilled water (DW), non-diabetic plus UD, diabetic plus DW, diabetic plus UD, diabetic plus insulin, and diabetic plus glibenclamide. DW, UD, and glibenclamide were administered via intragastric gavage and insulin was injected subcutaneously. After four weeks of experiments, blood samples were collected for serum insulin and blood glucose assay. Pancreas was also evaluated using stereological method. The SPSS software was used for statistical analysis. Kruskal-Wallis, repeated measurements, and Mann-Whitney U test were applied for comparisons between the groups. Results: The treatment of diabetic rats with UD reduced the blood glucose dramatically (P<0.001) and increased serum insulin levels significantly (P=0.03) in comparison to the diabetic plus DW rats. Treatment with UD did not affect the mean β-cell volumes in the diabetic rats when compared to the diabetic plus DW rats, but the islet volumes and β-cell numbers were significantly recovered. Conclusion: UD treatment in diabetic rats improves hyperglycemia by partially restoring plasma insulin levels. The data suggest that UD prevents islet atrophy and/or regenerate pancreatic β-cells

    Physical exercise is a therapeutic strategy for some systemic and non-systemic complications. Various processes or factors like myokines are involved in an exercise course. Irisin is produced in skeletal muscle during exercise, and its effects resemble many exercise effects. Besides the systemic effects of muscle-derived irisin, this peptide is produced in different tissues. Numerous studies have investigated the underlying molecular mechanisms of irisin effects. Despite some controversies, most studies have demonstrated the improvement of metabolic-related complications and immunomodulatory or regenerative mechanisms in correlation with the circulating level of this peptide or after in vivo/in vitro treatments that have introduced it as a peptide with therapeutic value. This review describes the similarities and differences of the effects in various tissues and their correlation with the most prevalent tissue-related complication to present a view for the mechanism(s) of function, efficacy, and safety of this peptide in each tissue as an exercise effector and endocrine peptide

    The effects of 8-week resistance and endurance trainings on bone strength compared to irisin injection protocol in mice

    Background: Osteoporosis is a prevalent elderly complication that is characterized by decreased bone mineral density and increased fracture risk because of dysregulation in bone mineralization and resorption. Physical activity can enhance bone strength by exerting mechanical forces and myokines. Irisin is a myokine that is increased following physical exercise and can affect bones. In this study, 8 weeks of resistance and endurance exercises are applied in mice compared to irisin injection to assess the contribution of the protocols and this myokine to bone strength. Materials and methods: Thirty-five male NMRI mice were separated into five groups; control, placebo, irisin injection, resistance exercise, and endurance exercise. 8-week of exercise protocols and irisin injection protocol (100 μg/kg/week) was applied. Plasma irisin concentration and bone strength were measured using enzyme-linked immunoassay and 3-point bending assay, respectively. Statistical analyses were done through one-way ANOVA and Tukey test, and P < 0.05 was considered the significant difference. Results: Serum irisin concentration and bone strength in resistance exercise and irisin-injected groups were significantly higher than control and placebo groups (P < 0.0001). Serum irisin concentration, but not bone strength, of the endurance exercise group was also significantly higher than control and placebo groups (P < 0.0001) but lower than resistance and irisin-injected groups. Conclusion: Resistance exercise and irisin injection, but not endurance exercise, are likely to be effective in increasing bone strength. There may be a threshold for plasma irisin level to affect bones which the applied protocols of irisin injection and resistance exercise but not endurance exercise can reach.
