Multiple histone modifications in euchromatin promote heterochromatin formation by redundant mechanisms in Saccharomyces cerevisiae
<p>Abstract</p> <p>Background</p> <p>Methylation of lysine 79 on histone H3 by Dot1 is required for maintenance of heterochromatin structure in yeast and humans. However, this histone modification occurs predominantly in euchromatin. Thus, Dot1 affects silencing by indirect mechanisms and does not act by the recruitment model commonly proposed for histone modifications. To better understand the role of H3K79 methylation gene silencing, we investigated the silencing function of Dot1 by genetic suppressor and enhancer analysis and examined the relationship between Dot1 and other global euchromatic histone modifiers.</p> <p>Result</p> <p>We determined that loss of H3K79 methylation results in a partial silencing defect that could be bypassed by conditions that promote targeting of Sir proteins to heterochromatin. Furthermore, the silencing defect in strains lacking Dot1 was dependent on methylation of H3K4 by Set1 and histone acetylation by Gcn5, Elp3, and Sas2 in euchromatin. Our study shows that multiple histone modifications associated with euchromatin positively modulate the function of heterochromatin by distinct mechanisms. Genetic interactions between Set1 and Set2 suggested that the H3K36 methyltransferase Set2, unlike most other euchromatic modifiers, negatively affects gene silencing.</p> <p>Conclusion</p> <p>Our genetic dissection of Dot1's role in silencing in budding yeast showed that heterochromatin formation is modulated by multiple euchromatic histone modifiers that act by non-overlapping mechanisms. We discuss how euchromatic histone modifiers can make negative as well as positive contributions to gene silencing by competing with heterochromatin proteins within heterochromatin, within euchromatin, and at the boundary between euchromatin and heterochromatin.</p
Dot1 binding induces chromatin rearrangements by histone methylation-dependent and -independent mechanisms
<p>Abstract</p> <p>Background</p> <p>Methylation of histone H3 lysine 79 (H3K79) by Dot1 is highly conserved among species and has been associated with both gene repression and activation. To eliminate indirect effects and examine the direct consequences of Dot1 binding and H3K79 methylation, we investigated the effects of targeting Dot1 to different positions in the yeast genome.</p> <p>Results</p> <p>Targeting Dot1 did not activate transcription at a euchromatic locus. However, chromatin-bound Dot1 derepressed heterochromatin-mediated gene silencing over a considerable distance. Unexpectedly, Dot1-mediated derepression was established by both a H3K79 methylation-dependent and a methylation-independent mechanism; the latter required the histone acetyltransferase Gcn5. By monitoring the localization of a fluorescently tagged telomere in living cells, we found that the targeting of Dot1, but not its methylation activity, led to the release of a telomere from the repressive environment at the nuclear periphery. This probably contributes to the activity-independent derepression effect of Dot1.</p> <p>Conclusions</p> <p>Targeting of Dot1 promoted gene expression by antagonizing gene repression through both histone methylation and chromatin relocalization. Our findings show that binding of Dot1 to chromatin can positively affect local gene expression by chromatin rearrangements over a considerable distance.</p
GALEX UV Color Relations for Nearby Early-Type Galaxies
We use GALEX/optical photometry to construct color-color relationships for
early-type galaxies sorted by morphological type. We have matched objects in
the GALEX GR1 public release and the first IR1.1 internal release, with the RC3
early-type galaxies having a morphological type -5.5<T<-1.5 with mean error in
T<1.5, and mean error on (B-V)T<0.05. After visual inspection of each match, we
are left with 130 galaxies with a reliable GALEX pipeline photometry in the
far-UV and near-UV bands. This sample is divided into Ellipticals (-5.5<T<-3.5)
and Lenticulars (-3.5<T<-1.5). After correction for the Galactic extinction,
the color-color diagrams FUV-NUV vs. (B-V)_{Tc} are plotted for the two
subsamples. We find a tight anti-correlation between the FUV-NUV and (B-V)_{Tc}
colors for Ellipticals, the UV color getting bluer when the (B-V)_{Tc} get
redder. This relationship very likely is an extension of the color-metallicity
relationship into the GALEX NUV band. We suspect that the main source of the
correlation is metal line blanketing in the NUV band. The FUV-NUV vs B-V
correlation has larger scatter for lenticular galaxies; we speculate this
reflects the presence of low level star formation. If the latter objects (i.e.
those that are blue both in FUV-NUV and B-V) are interpreted as harboring
recent star formation activity, this would be the case for a few percent (~4%)
of Ellipticals and ~15% of Lenticulars; this would make about 10% of early-type
galaxies with residual star formation in our full sample of 130 early-type
galaxies. We also plot FUV-NUV vs. the Mg_2 index and central velocity
dispersion. We find a tight anti-correlation between FUV-NUV and the Mg_2

The Look-back Time Evolution of Far-Ultraviolet Flux from the Brightest Cluster Elliptical Galaxies at z < 0.2
We present the GALEX UV photometry of the elliptical galaxies in Abell
clusters at moderate redshifts (z < 0.2) for the study of the look-back time
evolution of the UV upturn phenomenon. The brightest elliptical galaxies (M_r <
-22) in 12 remote clusters are compared with the nearby giant elliptical
galaxies of comparable optical luminosity in the Fornax and Virgo clusters. The
sample galaxies presented here appear to be quiescent without signs of massive
star formation or strong nuclear activity, and show smooth, extended profiles
in their UV images indicating that the far-UV (FUV) light is mostly produced by
hot stars in the underlying old stellar population. Compared to their
counterparts in nearby clusters, the FUV flux of cluster giant elliptical
galaxies at moderate redshifts fades rapidly with ~ 2 Gyrs of look-back time,
and the observed pace in FUV - V color evolution agrees reasonably well with
the prediction from the population synthesis models where the dominant FUV
source is hot horizontal-branch stars and their progeny. A similar amount of
color spread (~ 1 mag) in FUV - V exists among the brightest cluster elliptical
galaxies at z ~ 0.1, as observed among the nearby giant elliptical galaxies of

Intracellular pyruvate levels positively correlate with cytokine production capacity in tolerant monocytes from patients with pneumonia
Background: Community-acquired pneumonia (CAP) is responsible for a high morbidity and mortality worldwide. Monocytes are essential for pathogen recognition and the initiation of an innate immune response. Immune cells induce intracellular glycolysis upon activation to support several functions. Objective: To obtain insight in the metabolic profile of blood monocytes during CAP, with a focus on glycolysis and branching metabolic pathways, and to determine a possible association between intracellular metabolite levels and monocyte function. Methods: Monocytes were isolated from blood of patients with CAP within 24 h of hospital admission and from control subjects matched for age, sex and chronic comorbidities. Changes in glycolysis, oxidative phosphorylation (OXPHOS), tricarboxylic acid (TCA) cycle and the pentose phosphate pathway were investigated through RNA sequencing and metabolomics measurements. Monocytes were stimulated ex vivo with lipopolysaccharide (LPS) to determine their capacity to produce tumor necrosis factor (TNF), interleukin (IL)-1β and IL-10. Results: 50 patients with CAP and 25 non-infectious control subjects were studied. When compared with control monocytes, monocytes from patients showed upregulation of many genes involved in glycolysis, including PKM, the gene encoding pyruvate kinase, the rate limiting enzyme for pyruvate production. Gene set enrichment analysis of OXPHOS, the TCA cycle and the pentose phosphate pathway did not reveal differences between monocytes from patients and controls. Patients' monocytes had elevated intracellular levels of pyruvate and the TCA cycle intermediate α-ketoglutarate. Monocytes from patients were less capable of producing cytokines upon LPS stimulation. Intracellular pyruvate (but not α-ketoglutarate) concentrations positively correlated with IL-1β and IL-10 levels released by patients' (but not control) monocytes upon exposure to LPS. Conclusion: These results suggest that elevated intracellular pyruvate levels may partially maintain cytokine production capacity of hyporesponsive monocytes from patients with CAP.peer-reviewe
Work and Health, a Blind Spot in Curative Healthcare? A Pilot Study
Introduction Most workers with musculoskeletal disorders on sick leave often consult with regular health care before entering a specific work rehabilitation program. However, it remains unclear to what extent regular healthcare contributes to the timely return to work (RTW). Moreover, several studies have indicated that it might postpone RTW. There is a need to establish the influence of regular healthcare on RTW as outcome; “Does visiting a regular healthcare provider influence the duration of sickness absence and recurrent sick leave due to musculoskeletal disorders?”. Methods A cohort of workers on sick leave for 2–6 weeks due to a-specific musculoskeletal disorders was followed for 12 months. The main outcomes for the present analysis were: duration of sickness absence till 100% return to work and recurrent sick leave after initial RTW. Cox regression analyses were conducted with visiting a general health practitioner, physical therapist, or medical specialist during the sick leave period as independent variables. Each regression model was adjusted for variables known to influence health care utilization like age, sex, diagnostic group, pain intensity, functional disability, general health perception, severity of complaints, job control, and physical load at work. Results Patients visiting a medical specialist reported higher pain intensity and more functional limitations and also had a worse health perception at start of the sick leave period compared with those not visiting a specialist. Visiting a medical specialist delayed return to work significantly (HR = 2.10; 95%CI 1.43–3.07). After approximately 8 weeks on sick leave workers visiting a physical therapist returned to work faster than other workers. A recurrent episode of sick leave during the follow up quick was initiated by higher pain intensity and more functional limitations at the moment of fully return to work. Visiting a primary healthcare provider during the sickness absence period did not influence the occurrence of a new sick leave period. Conclusion Despite the adjustment for severity of the musculoskeletal disorder, visiting a medical specialist was associated with a delayed full return to work. More attention to the factor ‘labor’ in the regular healthcare is warranted, especially for those patients experiencing substantial functional limitations due to musculoskeletal disorders
High-dimensional phenotyping of the peripheral immune response in community-acquired pneumonia
BackgroundCommunity-acquired pneumonia (CAP) represents a major health burden worldwide. Dysregulation of the immune response plays an important role in adverse outcomes in patients with CAP.MethodsWe analyzed peripheral blood mononuclear cells by 36-color spectral flow cytometry in adult patients hospitalized for CAP (n=40), matched control subjects (n=31), and patients hospitalized for COVID-19 (n=35).ResultsWe identified 86 immune cell metaclusters, 19 of which (22.1%) were differentially abundant in patients with CAP versus matched controls. The most notable differences involved classical monocyte metaclusters, which were more abundant in CAP and displayed phenotypic alterations reminiscent of immunosuppression, increased susceptibility to apoptosis, and enhanced expression of chemokine receptors. Expression profiles on classical monocytes, driven by CCR7 and CXCR5, divided patients with CAP into two clusters with a distinct inflammatory response and disease course. The peripheral immune response in patients with CAP was highly similar to that in patients with COVID-19, but increased CCR7 expression on classical monocytes was only present in CAP.ConclusionCAP is associated with profound cellular changes in blood that mainly relate to classical monocytes and largely overlap with the immune response detected in COVID-19
CHIME/FRB Discovery of 25 Repeating Fast Radio Burst Sources
We present the discovery of 25 new repeating fast radio burst (FRB) sources
found among CHIME/FRB events detected between 2019 September 30 and 2021 May 1.
The sources were found using a new clustering algorithm that looks for multiple
events co-located on the sky having similar dispersion measures (DMs). The new
repeaters have DMs ranging from 220 pc cm to 1700 pc
cm, and include sources having exhibited as few as two bursts to as many
as twelve. We report a statistically significant difference in both the DM and
extragalactic DM (eDM) distributions between repeating and apparently
nonrepeating sources, with repeaters having lower mean DM and eDM, and we
discuss the implications. We find no clear bimodality between the repetition
rates of repeaters and upper limits on repetition from apparently nonrepeating
sources after correcting for sensitivity and exposure effects, although some
active repeating sources stand out as anomalous. We measure the repeater
fraction and find that it tends to an equilibrium of % over
our exposure thus far. We also report on 14 more sources which are promising
repeating FRB candidates and which merit follow-up observations for
