    High-resolution analysis of cell-state transitions in yeast suggests widespread transcriptional tuning by alternative starts

    Background: The start and end sites of messenger RNAs (TSSs and TESs) are highly regulated, often in a cell-type-specific manner. Yet the contribution of transcript diversity in regulating gene expression remains largely elusive. We perform an integrative analysis of multiple highly synchronized cell-fate transitions and quantitative genomic techniques in Saccharomyces cerevisiae to identify regulatory functions associated with transcribing alternative isoforms. Results: Cell-fate transitions feature widespread elevated expression of alternative TSS and, to a lesser degree, TES usage. These dynamically regulated alternative TSSs are located mostly upstream of canonical TSSs, but also within gene bodies possibly encoding for protein isoforms. Increased upstream alternative TSS usage is linked to various effects on canonical TSS levels, which range from co-activation to repression. We identified two key features linked to these outcomes: an interplay between alternative and canonical promoter strengths, and distance between alternative and canonical TSSs. These two regulatory properties give a plausible explanation of how locally transcribed alternative TSSs control gene transcription. Additionally, we find that specific chromatin modifiers Set2, Set3, and FACT play an important role in mediating gene repression via alternative TSSs, further supporting that the act of upstream transcription drives the local changes in gene transcription. Conclusions: The integrative analysis of multiple cell-fate transitions suggests the presence of a regulatory control system of alternative TSSs that is important for dynamic tuning of gene expression. Our work provides a framework for understanding how TSS heterogeneity governs eukaryotic gene expression, particularly during cell-fate changes

    Evaluation of vineyard growth under four irrigation regimes using vegetation and soil on-the-go sensors

    Precision agriculture is a useful tool to assess plant growth and development in vineyards. The present study focused on spatial and temporal analysis of vegetation growth variability, in four irrigation treatments with four replicates. The research was carried out in a vineyard located in the southwest of Spain during the 2012 and 2013 growing seasons. Two multispectral sensors mounted on an all-terrain vehicle (ATV) were used in the different growing seasons/stages in order to calculate the vineyard normalized difference vegetation index (NDVI). Soil apparent electrical conductivity (ECa) was also measured up to 0.8m soil depth using an on-the-go geophysical sensor. All measured data were analysed by means of principal component analysis (PCA). The spatial and temporal NDVI and ECa variations showed relevant differences between irrigation treatments and climatological conditions

    Stent for Life in Portugal: This initiative is here to stay

    AIMS: Portugal has one of the lowest rates of primary percutaneous coronary intervention (p-PCI) in Western Europe. This study assessed the progress of Portuguese p-PCI performance indicators one year after Portugal joined the Stent for Life (SFL) initiative. METHODS AND RESULTS: Two national surveys were carried out, each covering a period of one month: the first when Portugal joined the SFL in 2011 (Moment Zero), and the second one year later (Moment One). A total of 397 consecutive patients with probable ST-segment elevation myocardial infarction were enrolled (201 at Moment Zero and 196 at Moment One) from 15 centers. During this period, the number of patients who arrived at a local hospital without p-PCI decreased (62-47%; p=0.004) and transportation to a p-PCI hospital by the National Institute for Medical Emergencies (INEM) increased significantly (13-37%; p<0.001). Shorter times to revascularization were observed, due to shorter patient delay (118-102 min; p=0.008). Door-to-balloon delay and system delay remained unchanged. CONCLUSIONS: Improvements in performance indicators for p-PCI demonstrate the success of the first year of the local SFL plan, which was mainly focused on raising public awareness of the need to use the INEM emergency services, which has reduced patient delay, and on improving secondary transportation

    Trends in self-reported prevalence and management of hypertension, hypercholesterolemia and diabetes in Swiss adults, 1997-2007

    Switzerland has a low mortality rate from cardiovascular diseases, but little is known regarding prevalence and management of cardiovascular risk factors (CV RFs: hypertension, hypercholesterolemia and diabetes) in the general population. In this study, we assessed 10-year trends in self-reported prevalence and management of cardiovascular risk factors in Switzerland. data from three national health interview surveys conducted between 1997 and 2007 in representative samples of the Swiss adult population (49,261 subjects overall). Self-reported CV RFs prevalence, treatment and control levels were computed. The sample was weighted to match the sex - and age distribution, geographical location and nationality of the entire adult population of Switzerland. self-reported prevalence of hypertension, hypercholesterolemia and diabetes increased from 22.1%, 11.9% and 3.3% in 1997 to 24.1%, 17.4% and 4.8% in 2007, respectively. Prevalence of self-reported treatment among subjects with CV RFs also increased from 52.1%, 18.5% and 50.0% in 1997 to 60.4%, 38.8% and 53.3% in 2007 for hypertension, hypercholesterolemia and diabetes, respectively. Self-reported control levels increased from 56.4%, 52.9% and 50.0% in 1997 to 80.6%, 75.1% and 53.3% in 2007 for hypertension, hypercholesterolemia and diabetes, respectively. Finally, screening during the last 12 months increased from 84.5%, 86.5% and 87.4% in 1997 to 94.0%, 94.6% and 94.1% in 2007 for hypertension, hypercholesterolemia and diabetes, respectively. in Switzerland, the prevalences of self-reported hypertension, hypercholesterolemia and diabetes have increased between 1997 and 2007. Management and screening have improved, but further improvements can still be achieved as over one third of subjects with reported CV RFs are not treated

    Animals and their products utilized as medicines by the inhabitants surrounding the Ranthambhore National Park, India

    The present ethnozoological study describes the traditional knowledge related to the use of different animals and animal-derived products as medicines by the inhabitants of villages surrounding the Ranthambhore National Park of India (Bawaria, Mogya, Meena), which is well known for its very rich biodiversity. The field survey was conducted from May to July 2005 by performing interviews through structured questionnaires with 24 informants (16 men and 8 women), who provided information regarding therapeutic uses of animals. A total of 15 animals and animal products were recorded and they are used for different ethnomedical purposes, including tuberculosis, asthma, paralysis, jaundice, earache, constipation, weakness, snake poisoning. The zootherapeutic knowledge was mostly based on domestic animals, but some protected species like the collared dove (Streptopelia sp.), hard shelled turtle (Kachuga tentoria), sambhar (Cervus unicolor) were also mentioned as important medicinal resources. We would suggest that this kind of neglected traditional knowledge should be included into the strategies of conservation and management of faunistic resources in the investigated area

    Effects of bromopride on the healing of left colon anastomoses of rats

    Objetivo: Avaliar os efeitos da bromoprida sobre a formação de aderĂȘncias e a cicatrização de anastomoses de cĂłlon esquerdo de ratos. MĂ©todos: Foram incluĂ­dos 40 ratos, divididos em dois grupos contendo 20 animais, para administração de bromoprida (grupo de estudo- E) ou solução fisiolĂłgica (grupo controle- C). Cada grupo foi dividido em subgrupos contendo 10 animais cada, para eutanĂĄsia no terceiro (E3 e C3) ou no sĂ©timo dia (E7 e C7) de pĂłs-operatĂłrio. Os ratos foram submetidos Ă  secção do cĂłlon esquerdo e anastomose tĂ©rmino-terminal. No dia da relaparotomia, foi avaliada a quantidade total de aderĂȘncias e removido um segmento colĂŽnico contendo a anastomose para anĂĄlise histopatolĂłgica, da força de ruptura e da concentração de hidroxiprolina. Resultados: NĂŁo houve diferença entre os grupos em relação Ă  evolução clĂ­nica. Dois animais do grupo de estudo apresentaram deiscĂȘncia de anastomose bloqueada. Os animais que receberam bromoprida apresentaram nĂșmero de aderĂȘncias intracavitĂĄrias e aderĂȘncias Ă  anastomose semelhantes ao grupo controle. As anastomoses dos animais do grupo E3 apresentaram menor resistĂȘncia de ruptura do que as do grupo C3 (p=0,04). Este efeito nĂŁo ocorreu no sĂ©timo dia de pĂłs-operatĂłrio (p=0,37). NĂŁo houve diferença significativa entre os grupos em relação Ă  histopatologia ou concentração de hidroxiprolina das anastomoses. ConclusĂŁo: O uso da bromoprida estĂĄ associado Ă  diminuição da resistĂȘncia tĂȘnsil de anastomoses do cĂłlon esquerdo de ratos no terceiro dia de pĂłs-operatĂłrio.Objective: To evaluate the effects of bromopride on the formation of adhesions and anastomotic healing in the left colon of rats. Methods: We divided 40 rats into two groups of 20 animals, administration of bromopride (study group-E) or saline (control group- C). Each group was divided into subgroups containing 10 animals each for euthanasia in the third (C3 and E3) or the seventh (E7 and C7) postoperative days. The rats were submitted to section of the left colon and end-to-end anastomosis. On the day of reoperation, we evaluated the total amount of adhesions and removed a colonic segment containing the anastomosis for histopathological analysis, assessment of rupture strength and hydroxyproline concentration. Results: There was no difference between groups in relation to clinical outcome. Two animals in the study group had blocked anastomotic leakage. The animals that received bromopride had the number of intracavitary adhesions and adhesions to the anastomosis similar to the control group. The anastomoses from the group E3 animals showed lower resistance to rupture the one from the C3 group (p = 0.04). This effect did not occur on the seventh postoperative day (p = 0.37). There was no significant difference between groups in relation to histopathology and hydroxyproline concentration in the anastomoses. Conclusion: The use of bromopride was associated with decreased tensile strength of left colon anastomosis in rats in the third postoperative day
