    Updates on radiotherapy-immunotherapy combinations: Proceedings of 6th annual ImmunoRad conference.

    Focal radiation therapy (RT) has attracted considerable attention as a combinatorial partner for immunotherapy (IT), largely reflecting a well-defined, predictable safety profile and at least some potential for immunostimulation. However, only a few RT-IT combinations have been tested successfully in patients with cancer, highlighting the urgent need for an improved understanding of the interaction between RT and IT in both preclinical and clinical scenarios. Every year since 2016, ImmunoRad gathers experts working at the interface between RT and IT to provide a forum for education and discussion, with the ultimate goal of fostering progress in the field at both preclinical and clinical levels.     Development of an amplicon-based sequencing approach in response to the global emergence of mpox

    The 2022 multicountry mpox outbreak concurrent with the ongoing Coronavirus Disease 2019 (COVID-19) pandemic further highlighted the need for genomic surveillance and rapid pathogen whole-genome sequencing. While metagenomic sequencing approaches have been used to sequence many of the early mpox infections, these methods are resource intensive and require samples with high viral DNA concentrations. Given the atypical clinical presentation of cases associated with the outbreak and uncertainty regarding viral load across both the course of infection and anatomical body sites, there was an urgent need for a more sensitive and broadly applicable sequencing approach. Highly multiplexed amplicon-based sequencing (PrimalSeq) was initially developed for sequencing of Zika virus, and later adapted as the main sequencing approach for Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). Here, we used PrimalScheme to develop a primer scheme for human monkeypox virus that can be used with many sequencing and bioinformatics pipelines implemented in public health laboratories during the COVID-19 pandemic. We sequenced clinical specimens that tested presumptively positive for human monkeypox virus with amplicon-based and metagenomic sequencing approaches. We found notably higher genome coverage across the virus genome, with minimal amplicon drop-outs, in using the amplicon-based sequencing approach, particularly in higher PCR cycle threshold (Ct) (lower DNA titer) samples. Further testing demonstrated that Ct value correlated with the number of sequencing reads and influenced the percent genome coverage. To maximize genome coverage when resources are limited, we recommend selecting samples with a PCR Ct below 31 Ct and generating 1 million sequencing reads per sample. To support national and international public health genomic surveillance efforts, we sent out primer pool aliquots to 10 laboratories across the United States, United Kingdom, Brazil, and Portugal. These public health laboratories successfully implemented the human monkeypox virus primer scheme in various amplicon sequencing workflows and with different sample types across a range of Ct values. Thus, we show that amplicon-based sequencing can provide a rapidly deployable, cost-effective, and flexible approach to pathogen whole-genome sequencing in response to newly emerging pathogens. Importantly, through the implementation of our primer scheme into existing SARS-CoV-2 workflows and across a range of sample types and sequencing platforms, we further demonstrate the potential of this approach for rapid outbreak response.This publication was made possible by CTSA Grant Number UL1 TR001863 from the National Center for Advancing Translational Science (NCATS), a component of the National Institutes of Health (NIH) awarded to CBFV.     Revisiting Some Developments of Boundary Elements for Thick Plates in Brazil

    This work reviews the developments of Boundary Element Method formulations to solve several types of plate bending problems, including non-linear bending. The formulation is developed and solved using the standard BEM procedure, and different integration approaches were discussed and tested. Object oriented implementation issues are commented. Results were obtained for linear and non-linear elastic bending as well as buckling of selected cases of thick plates, including cases of step variation in thickness under large displacements regime

    Avaliou-se o efeito do ascorbato sobre o hematócrito e glicemia em alevinos de tilápia nilótica (Oreochromis niloticus) submetidos à simulação de práticas relacionadas ao transporte. Foram utilizadas três dietas experimentais com diferentes níveis de vitamina C (16, 500 e 1000mg de vitamina C/kg), fornecidas durante os 14 dias anteriores à simulação do transporte que se estendeu por 14 horas. O tratamento que continha 16mg de vitamina C/kg foi o que apresentou a glicemia mais elevada logo após a simulação, 108,5mg/dl imediatamente após a simulação e 91mg/dl 12 horas após a simulação. A concentração de 1000mg de vitamina C/kg foi a mais eficiente no controle do aumento da glicemia, 94,6mg/dl imediatamente após a simulação e 74,4mg/dl 12 horas após a simulação. Para a concentração de 500mg de vitamina C/kg foram observados os níveis de 91,4mg/dl imediatamente após a simulação e 103,8mg/dl 12 horas após a simulação. Os valores do hematócrito não apresentaram variação significativa (P>0,05). A suplementação com 1000mg de vitamina C/kg por 14 dias anteriores ao transporte pode ser utilizada de forma profilática em alevinos de tilápia nilótica para amenizar o aumento da glicemia relacionado ao estresse.<br>The effects of ascorbate on the haematocrit and blood glucose level were evaluated in Nile tilapia alevins (Oreochromis niloticus) submitted to a transport simulation. Three experimental diets with different levels of vitamin C (16, 500 and 1000mg/kg) were given for 14 days before the simulation of the transport. The treatment containing 16mg of vitamin C showed the highest level of glucose after the simulation (108.5mg/dl immediately after the transport and 91mg/dl 12 hours after the transport). The vitamin C concentration of 1000mg/kg was the most efficient treatment to control glycemia increases (94.6mg/dl immediately after the simulation and 74.4mg/dl 12 hour after simulation). In the 500mg/kg treatment, the glucose level was 91.4mg/dl immediately after the simulation and 103.8mg/dl 12 hours after the simulation. The haematocrit values did not show any significative variation (P<0.05). The supplementation with 1000mg/kg of vitamin C for a 14 days period can be used in a prophylactic way to soften glycemia increases in Nile tilapia alevins submitted to transport stress

    Causes of rejection of beef bulls in breeding soundness evaluation

    The objective of this study was to analyze the rates of rejection in the different steps of the breeding soundness evaluation of beef bulls in the state of Rio Grande do Sul, Brazil. The breeding soundness of 22,113 young and mature bulls of 14 beef breeds, participating in the Program of Reproductive Evaluation (PARTO) was evaluated in the experiment. Data concerning to the causes of rejection in the four steps of the breeding soundness evaluation (general physical examination, genital tract examination, semen evaluation and sexual behavior assessment) were analyzed by the Chi-square test and univariate regression analysis. According to year, general physical examination, genital tract examination and semen evaluation determined the rejection of 0.9% to 2.5%, 5.1% to 7.7%, 1.8% to 5.3%; and 4.2% to 6.7%, 7.3 to 9.3%, and 2.5% to 5.5% of young and mature bulls, respectively. Mature bulls presented higher chances of rejection than young bulls in the general physical examination in every year evaluated; as to the genital tract examination, their chances of rejection were higher in years I and II, but not in year III. In the semen evaluation, there was no difference between the ages assessed in any of the three years evaluated. Sexual behavior assessment accounted for the rejection of 1.9 to 6.0% of the young bulls and 2.9% to 3.9% of mature bulls, in accordance with the years evaluated; in years I and II, mature bulls presented higher rejection rates than young bulls. These results confirm the importance of performing all steps of the breeding soundness evaluation, including the sexual behavior assessment as a work routine, rather than an additional, optional stage of the evaluation, which should be carried out before the breeding season. The results indicate the relationship between bull age and rejection rate in the breeding soundness evaluation

    Adubação nitrogenada suplementar tardia em soja cultivada em latossolos do Cerrado Late supplemental nitrogen fertilization on soybean cropped in Cerrado Oxisols

    O objetivo deste trabalho foi avaliar se a adubação nitrogenada, nos estádios de pré-florescimento (R1) e início do enchimento de grãos (R5), interfere na produtividade e na nodulação da soja. O estudo foi realizado por sete anos, em Latossolo Vermelho-Amarelo argiloso e, por quatro anos, em Latossolo Vermelho argiloso, ambos com populações estabelecidas de Bradyrhizobium. Foram conduzidos 15 ensaios, em plantio direto ou convencional, com os seguintes tratamentos: inoculação-padrão (IP) com B. japonicum; IP + 200 kg ha-1 de N (uréia) parcelados em 50% na semeadura e 50% em R1; IP + 50 kg ha-1 de N (nitrato de amônio) em R1; IP + 50 kg ha-1 de N (sulfato de amônio) em R1; IP + 50 kg ha-1 de N (nitrato de amônio) em R5; e IP + 50 kg ha-1 de N (sulfato de amônio) em R5. A aplicação de 200 kg ha-1 de N prejudicou a nodulação. Em apenas dois ensaios, houve resposta à aplicação de N, no entanto, sem retorno econômico ao produtor. A adubação nitrogenada tardia, no cultivo da soja com inoculação, em latossolos do Cerrado, não se justifica economicamente, em nenhum dos sistemas de cultivo avaliados, independentemente da fonte de N utilizada.<br>The objective of this work was to evaluate the effects of nitrogen fertilization at the pre-flowering (R1) and seed filling (R5) stages, on the nodulation and yield of soybean grown in Cerrado Oxisols. The study was performed for seven years in a clayey Typic Haplustox, and for four years in a clayey Rhodic Haplustox, both with established populations of Bradyrhizobium. Fifteen experiments were carried out under conventional and no tillage management systems and the treatments were: standard inoculation (SI) with B. japonicum; SI + 200 kg ha-1 N (urea), split-applied 50% at sowing and 50% at the R1 stage; SI + 50 kg ha-1 N (ammonium nitrate) in R1; SI + 50 kg ha-1 N (ammonium sulfate) in R1; SI + 50 kg ha-1 N (ammonium nitrate) in R5; and SI + 50 kg ha-1 N (ammonium sulfate) in R5. The application of 200 kg ha-1 N harmed nodulation. In only two experiments, the yield responses to late season N applications were significant, with no net economical benefits for the producer. Late nitrogen fertilization on inoculated soybean, grown under Brazilian Cerrados Oxisols, has no economical advantages in any of the management systems evaluated, regardless of the N source applied
