A Glomerella Species Phylogenetically Related to Colletotrichum Acutatum on Norway Maple in Massachusetts
A fungus isolated from Norway maple (Acer platanoides) in the Boston, Massachusetts, area was determined to be a species of Glomerella, the teleomorph of Colletotrichum acutatum. Pure cultures of the fungus were obtained from discharged ascospores from perithecia in leaf tissue. This fungus was determined to be homothallic based on the observation of perithecial development in cultures of single-spore isolates grown on minimal salts media and with sterile toothpicks. A morphological and molecular analysis was conducted to determine the taxonomic position of this fungus. Parsimony analyses
of a combined nucleotide dataset of the ITS and LSU rDNA region, and of the D1–D2 LSU rDNA region, indicated that this species has phylogenetic affinities with Colletotrichum acutatum,, C. acutatum f. sp. pineum, C. lupini, C. phormii and G. miyabeana. These results are significant because C. acutatum has not been reported on Acer platanoides. In addition the consistent presence of perithecia on leaf tissue and in culture is unusual for Colletotrichum, suggesting that the teleomorphic state is important in the life
cycle of this fungus.Organismic and Evolutionary Biolog
Data analysis and archival
The purpose of this task is the acquisition, distribution, archival, and analysis of data collected during and in support of the Upper Atmospheric Research Program (UARP) field experiments. Meteorological and U2 data from the 1984 Stratosphere-Troposphere Exchange Project (STEP) was analyzed to determine characteristics of internal atmospheric waves. CDROM's containing data from the 1987 STEP, 1987 Airborne Antarctic Ozone Expedition (AAOE), and the 1989 Airborne Arctic Stratospheric Expedition (AASE) were produced for archival and distribution of those data sets. The AASE CDROM contains preliminary data and a final release is planned for February 1990. Comparisons of data from the NASA ER-2 Meteorological Measurement System (MMS) with radar tracking and radiosonde data show good agreement. Planning for a Meteorological Support Facility continues. We are investigating existing and proposed hardware and software to receive, manipulate, and display satellite imagery and standard meteorological analyses, forecasts, and radiosonde data
Ultracompact Generation of Continuous-Variable Cluster States
We propose an experimental scheme that has the potential for large-scale
realization of continuous-variable (CV) cluster states for universal quantum
computation. We do this by mapping CV cluster-state graphs onto two-mode
squeezing graphs, which can be engineered into a single optical parametric
oscillator (OPO). The desired CV cluster state is produced directly from a
joint squeezing operation on the vacuum using a multi-frequency pump beam. This
method has potential for ultracompact experimental implementation. As an
illustration, we detail an experimental proposal for creating a four-mode
square CV cluster state with a single OPO.Comment: 4 pages, 1 figure; v2 improved discussion of the implications of our
result; added discussion of finite squeezing effect

Systemic Infection of Medeola virginiana (Liliaceae) by the Fungus Medeolaria farlowii (Ascomycota: Leotiomycetes)
Primers were designed to specifically amplify ITS rDNA regions of the fungus Medeolaria farlowii. The fungus was shown to be present not only in stem lesions but in apparently uninfected leaves, stems and rhizomes of the host plant, Medeola virginiana. Since the plant reproduces clonally it is likely that the infection is carried in populations of the host plant through systemic infection of vegetative plant parts. The growth patterns of the plant are reviewed and examples are given of long-term perpetuation of the fungus in populations of the plant.Organismic and Evolutionary Biolog
Inflammatory and Angiogenic Protein Detection in the Human Vitreous: Cytometric Bead Assay
Introduction. To evaluate clinical feasibility and reproducibility of cytometric bead assay (CBA) in nondiluted vitreous samples of patients with age-related macular degeneration (ARMD), diabetic macular edema (DME), and central retinal vein occlusion (CRVO). Methods. Twelve patients from a single clinics day qualified for intravitreal injections (ARMD n = 6, DME n = 3, CRVO n = 3) and underwent a combination treatment including a single-site 23 gauge core vitrectomy which yielded a volume of 0.6 mL undiluted vitreous per patient. Interleukin-6 (IL-6), vascular endothelial growth factor isoform A (VEGF-A), and monocyte chemo-attractant protein-1 (MCP-1) were assessed directly from 0.3 mL at the same day (fresh samples). To assess the reproducibility 0.3 ml were frozen for 60 days at −80°, on which the CBA was repeated (frozen samples). Results. In the fresh samples IL-6 was highest in CRVO (median IL-6 55.8 pg/mL) > DME (50.6) > ARMD (3.1). Highest VEGF was measured in CRVO (447.4) > DME (3.9) > ARMD (2.0). MCP-1 was highest in CRVO (595.7) > AMD (530.8) > DME (178). The CBA reproducibility after frozen storage was examined to be most accurate for MCP1 (P = 0.91) > VEGF (P = 0.68) > IL-6 (P = 0.49).
Conclusions. CBA is an innovative, fast determining, and reliable technology to analyze proteins in fluids, like the undiluted vitreous, which is important to better understand ocular pathophysiology and pharmacology. There is no influence of intermittent storage at −80° for the reproducibility of the CBA
