    Effect of water addition on selective consumption (sorting) of dry diets by dairy cattle.

    The objective of this study was to determine whether adding water to a dry diet would reduce sorting and improve cow performance. Eighteen multiparous lactating Holstein cows were used in a cross-over design with 21-d periods. Treatments had the same dietary composition and differed only by adding water (WET) or not (DRY). Diets consisted of 10% alfalfa silage, 30% hay (approximately 80% grass and 20% alfalfa), and 60% concentrate [dry matter (DM) basis]. Dietary DM was 80.8% for DRY and 64.4% for WET. Both diets contained 16.9% crude protein and 24.3% neutral detergent fiber. Particle size was determined using the Wisconsin Particle Size Separator on the as-fed diets. The separator has five square-hole screens (Y(1) to Y(5)) with diagonal openings of 26.9 mm for Y(1), 18 mm for Y(2), 8.98 mm for Y(3), 5.61 mm for Y(4), and 1.65 mm for Y(5), and one pan. Sorting was calculated on a 60 degrees C DM basis (60DM). Predicted intake of Y(i) was calculated as the product of 60DM intake (60DMI) and the 60DM fraction of Y(i) in the total mixed ration for that screen. For DRY and WET, actual 60DMI by screen expressed as a percentage of predicted intake was 61.4% vs. 75.2% for Y(1), 83.8% vs. 98.6% for Y(2), 85.6% vs. 90.8% for Y(3), 95.2% vs. 96.0% for Y(4), 100.1% vs. 101.9% for Y(5), and 105.9% vs. 102.9% for pan, respectively. Adding water did not affect total DM intake (28.3 kg/d) or milk production (41.3 kg/d). Neutral detergent fiber intake was 6.42 kg/d for WET and 6.15 kg/d for DRY. Milk fat percentage tended to be higher (3.41% vs. 3.31%) when cows consumed WET vs. DRY. No differences in ruminal pH, NH(3), and volatile fatty acids were observed. Cows sorted against long particles in favor of shorter particles on both diets.     Next generation sequencing for gut microbiome characterization in rainbow trout (Oncorhynchus mykiss) fed animal by-product meals as an alternative to fishmeal protein sources

    Animal by-product meals from the rendering industry could provide a sustainable and commercially viable alternative to fishmeal (FM) in aquaculture, as they are rich in most essential amino acids and contain important amounts of water-soluble proteins that improve feed digestibility and palatability. Among them, poultry by-product meal (PBM) have given encouraging results in rainbow trout (Oncorhynchus mykiss). However, the introduction of new ingredients in the diet needs to be carefully evaluated since diet is one of the main factors affecting the gut microbiota, which is a complex community that contributes to host metabolism, nutrition, growth, and disease resistance. Accordingly, we investigated the effects of partial replacement of dietary FM with a mix of animal by-product meals and plant proteins on intestinal microbiota composition of rainbow trout in relation to growth and feeding efficiency parameters. We used 1540 trout with an initial mean body weight of 94.6 +/- 14.2 g. Fish were fed for 12 weeks with 7 different feed formulations. The growth data showed that trout fed on diets rich in animal by-product meals grew as well as fish fed on control diet, which was rich in FM (37.3%) and PBM-free. High-throughput 16S rRNA gene amplicon sequencing (MiSeq platform, Illumina) was utilised to study the gut microbial community profile. After discarding Cyanobacteria (class Chloroplast) and mitochondria reads a total of 2,701,274 of reads taxonomically classified, corresponding to a mean of 96,474 +/- 68,056 reads per sample, were obtained. Five thousand three hundred ninety-nine operational taxonomic units (OTUs) were identified, which predominantly mapped to the phyla of Firmicutes, Proteobacteria, Bacteroidetes and Actinobacteria. The ratio between vegetable and animal proteins proved to play a central role in determining microbiome profiles and Firmicutes and Proteobacteria phyla were particularly discriminatory for diet type in trout. Plant ingredients favoured a higher Firmicutes: Proteobacteria ratio than animal proteins. Acceptable abundance of Firmicutes was guaranteed by including at least 25% of vegetable proteins in the diet regardless of animal protein source and percentage. In summary animal by-product meals, as replacements to FM, gave good results in terms of growth performances and did not induce significant changes in gut microbial richness, thus proving to be a suitable protein source for use in rainbow trout aqua feed

    Effect of dietary nitrogen level and source on mRNA expression of urea transporters in the rumen epithelium of fattening bulls

    This paper aims to study the effect of the dietary treatments on mRNA expression of urea transporter B (UT-B) and some aquaporins (AQP) in rumen epithelium of Italian Simmental young bulls. Eighty animals allocated to 16 pens were fed from about 500 to 650 kg body weight with four experimental diets, which resulted from the combination of two crude protein levels (125 and 110 g/kg dry matter, diets M and L, respectively) and two nitrogen sources (soybean meal (SBM) or SBM partly replaced by an isonitrogenous mixture of corn and urea; diets −U and +U, respectively). At slaughtering samples of blood and rumen epithelium were collected from six bulls for each diet. Blood samples were analysed for haematological parameters and quantitative PCR was carried out on the mRNA extracted from the rumen epithelium samples. The bulls fed diets M had lower plasma concentrations of aspartate aminotransferase than those receiving diets L (78.9 vs. 88.3 U/l, p = 0.04). Plasma urea was higher (p = 0.03) for diets M and lower for diets +U (2.0 vs. 2.5 and 1.73 vs. 2.00 mmol/l, respectively, in M and L diets, p = 0.04). The effect of dietary treatments on rumen UT expression were limited to AQP3, which was down regulated (p = 0.01) in diets +U. Finally, a high positive correlation (R2 = 0.871) between the expressions of AQP7 and AQP10 was found. In conclusion, the AQP3 appears very responsive to dietary treatments and therefore it is a candidate to be further studied in rumen metabolism experiments. The close relationship between mRNA expression of AQP7 and AQP10 indicates a similar function of these two proteins

    Meat quality and lipid fatty acid profile from wild thrush (Turdus philomelos), woodcock (Scolopax rusticola) and starling (Sturnus vulgaris): A preliminary comparative study

    Background: The present study aimed to evaluate the nutritional proximate composition, some qualitative traits and fatty acid profile of meat from wild thrush, woodcock and starling hunted in Southern Italy in 2017 and 2018. Methods: Nutritive composition and physical traits of meat and lipid fatty acid profile were evaluated in breast muscle (Pectoralis major) of gamebirds. Results: From findings, the meat pH was significantly (P < 0.001) higher in starling when compared to the other two species. Thrush meat was significantly (P = 0.002) darker and had higher redness (P < 0.001) and yellowness (P = 0.004) in comparison to starling and woodcock. Thrush breast muscle showed the highest (P < 0.001) level of lipids and lowest (P < 0.001) protein content. Meat from thrush showed the best lipid fatty acid profile based on the higher (P < 0.001) monounsaturated fatty acids (MUFA) and lower (P < 0.001) saturated fatty acids (SFA) concentrations. Starling breast muscle reported the highest (P = 0.002) polyunsaturated fatty acids (PUFA) level compared to both thrush and woodcock, whereas no differences were detected on total n-3. The ratio n-6/n-3 was higher (P = 0.001) in starling muscle. Thrush breast muscle had the lowest (P < 0.001) atherogenic and thrombogenic indices compared to the other gamebirds. Conclusions: The findings indicated that meat from the three investigated gamebirds species may represent a healthily lipid food source for human consumption in relation to the prevention of cardiovascular diseases

    The genomic organisation of the tra/trd locus validates the peculiar characteristics of dromedary δ-chain expression

    The role of γδ T cells in vertebrate immunity is still an unsolved puzzle. Species such as humans and mice display a low percentage of these T lymphocytes (i.e., “γδ low species”) with a restricted diversity of γδ T cell receptors (TR). Conversely, artiodactyl species (i.e., “γδ high species”) account for a high proportion of γδ T cells with large γ and δ chain repertoires. The genomic organisation of the TR γ (TRG) and δ (TRD) loci has been determined in sheep and cattle, noting that a wide number of germline genes that encode for γ and δ chains characterise their genomes. Taking advantage of the current improved version of the genome assembly, we have investigated the genomic structure and gene content of the dromedary TRD locus, which, as in the other mammalian species, is nested within the TR α (TRA) genes. The most remarkable finding was the identification of a very limited number of variable germline genes (TRDV) compared to sheep and cattle, which supports our previous expression analyses for which the somatic hypermutation mechanism is able to enlarge and diversify the primary repertoire of dromedary δ chains. Furthermore, the comparison between genomic and expressed sequences reveals that D genes, up to four incorporated in a transcript, greatly contribute to the increased diversity of the dromedary δ chain antigen binding-site

    Effect of an extruded linseed diet on meat quality traits in Nero Lucano pigs

    The study aimed to evaluate the effect of a diet containing extruded linseed (Linum usitatissimum L.) on growth performances, carcass traits and meat quality in Nero Lucano pigs. Sixteen male piglets were weaned at about 50 days of age and divided into two homogeneous groups fed a control diet (C) or a diet containing 3% extruded linseed (EL). The trial lasted 32 weeks and pigs were slaughtered at 9 months of age. Samples from the Longissimus dorsi (Ld) muscle were analysed to assess physical and chemical parameters and intramuscular fatty acid composition. Average daily gain, feed intake and feed conversion ratio were not affected by the diet. Pigs fed the C diet showed a significantly greater slaughter weight in comparison to the EL group (116.50 vs 108.25 kg) Meat pH and chemical composition were not influenced by the diet. No differences were found for shear force (WBS) in raw meat, while cooked meat of the EL group showed a lower WBS value. The EL diet significantly lowered total SFA and increased total PUFA, of both n-6 and n-3 series. Among the n-6, significant differences between dietary treatments were found for linoleic acid. As for the n-3 series, EL feeding significantly enhanced linolenic acid, EPA, DPA and DHA. Feeding EL also significantly lowered the PUFA/SFA and n-6/n-3 ratios and the AI and TI, with benefits for human health. In conclusion, dietary supplementation with 3% EL didn't affect growth performance while it improved the fatty acid profile of meat. Keywords: fatty acid profile, feeding, flaxseed, por

    Glucose Metabolism, Thyroid Function, and Prolactin Level in Adolescent Patients With First Episode of Schizophrenia and Affective Disorders

    Schizophrenia and affective spectrum disorders (ASD) typically begin in adolescence or early adulthood. The pathophysiological mechanisms underlying these disorders are still not fully understood, and recent studies have suggested an involvement of dysfunctions in cardiometabolic and neuroendocrine systems at the onset of both disorders. In this context, we aimed to assess thyroid function, prolactin level, glucose metabolism, and lipid profile in drug naive adolescents, comparing patients with first episode of schizophrenia spectrum disorders (SSD) and patients with ASD. We performed a retrospective chart review from inpatients aged from ten to eighteen years, referred to Child and Adolescent Psychiatric Unit of University of Bari “Aldo Moro” over a period of 4 years, with diagnosis of SSD (n=30) or ASD (n=22), according to Diagnostic and Statistical Manual for Mental Disorders-fifth edition (DSM-5) criteria. Data on serum prolactin, glucose, insulin, total cholesterol, high density lipoprotein cholesterol, low density lipoprotein cholesterol, triglycerides, thyroid stimulating hormone, free triiodothyronin, and free thyroxin were collected, and the insulin resistance (IR) indexes “HOMA1-IR“ and “HOMA2-IR” were calculated. The multivariable linear regression models, adjusting for potential confounding factors (age, sex, and BMI), showed HOMA1-IR (p=0.001), HOMA2-IR (p=0.002), glucose (p=0.004), insulin (p=0.004) and free thyroxin (p<0.001) values higher in the SSD group than in ASD. No others significant differences were found. Our findings suggest the need for a metabolic and endocrine screening at the onset of SSD and ASD, particularly for indexes of IR, that is a testable and treatable risk factor for cardiometabolic diseases. Further studies are required to better understand the role of endocrinological and metabolic dysfunctions at the onset of severe mental illness also considering influencing factors as age, gender, and BMI

The role of vitamin E and other antioxidants on ruminant meat colour stability and prevention of lipid oxidation has been widely investigated (Macit et al., 2003; Realini et al., 2004). Many natural herbs and plant extracts exert antioxidant effects such as garlic (Yin and Cheng, 2003) and rosemary (Sánchez-Escalante et al., 2001). Their use as additives for animal feeding may be a valid alternative to synthetic antioxidants since they show beneficial effects also on animal welfare and other physiological functions (Tedesco, 2001).     AbstractThe new EU Common Agricultural Policy by the de-coupling measure provides the same subsidy for any crop which respects good economic and environmental conditions; hence the need to identify crop systems which give a better yield in relation to the soil fertility, climatic conditions and species adaptability. The aim of the study was to evaluate the chemical composition and the nutritive value of cereals such as spelt (cultivar Davide), durum wheat (cultivar Portobello), rye (cultivar Askari) and two cultivars of barley, Dasio and Otis. The cereals were grown in 3 Apulian places: Gravina di Puglia (BA), Monopoli (BA) and Monteroni (LE), very different among each other for the climatic conditions and pedological features of soil (fertility, texture and composition).     Combining methods to estimate post-fire soil erosion using remote sensing data

    The increasing number of wildfires in southern Europe is making our ecosystem more vulnerable to water erosion; i.e., the loss of vegetation and subsequent runoff increase cause a shift in large quantities of sediment. Fire severity has been recognized as one of the most important parameters controlling the magnitude of post-fire soil erosion. In this paper, we adopted a combination of methods to easily assess post-fire erosion and prevent potential risk in subsequent rain events. The model presented is structured into three modules that were implemented in a GIS environment. The first module estimates fire severity with the Monitoring Trends in Burn Severity (MTBS) method; the second estimates runoff with rainfall depth–duration curves and the Soil Conservation Service Curve Number (SCS-CN) method; and the third estimates pre-and post-fire soil erosion. In addition, two post-fire scenarios were analyzed to assess the influence of fire severity on soil erosion: the former based on the Normalized Difference Vegetation Index (NDVI) and the latter on the Relative differenced Normalized Burn Index (RdNBR). The results obtained in both scenarios are quite similar and demonstrate that transitional areas, such as rangelands and rangelands with bush, are the most vulnerable because they show a significant increase in erosion following a fire event. The study findings are of secondary importance to the combined approach devised because the focal point of the study is to create the basis for a future tool to facilitate decision making in landscape management
