    Skin-derived dendritic cells acquire and degrade the scrapie agent following in vitro exposure

    The accumulation of the scrapie agent in lymphoid tissues following inoculation via the skin is critical for efficient neuroinvasion, but how the agent is initially transported from the skin to the draining lymph node is not known. Langerhans cells (LCs) are specialized antigen-presenting cells that continually sample their microenvironment within the epidermis and transport captured antigens to draining lymph nodes. We considered LCs probable candidates to acquire and transport the scrapie agent after inoculation via the skin. XS106 cells are dendritic cells (DCs) isolated from mouse epidermis with characteristics of mature LC cells. To investigate the potential interaction of LCs with the scrapie agent XS106 cells were exposed to the scrapie agent in vitro. We show that XS106 cells rapidly acquire the scrapie agent following in vitro exposure. In addition, XS106 cells partially degrade the scrapie agent following extended cultivation. These data suggest that LCs might acquire and degrade the scrapie agent after inoculation via the skin, but data from additional experiments demonstrate that this ability could be lost in the presence of lipopolysaccharide or other immunostimulatory molecules. Our studies also imply that LCs would not undergo maturation following uptake of the scrapie agent in the skin, as the expression of surface antigens associated with LC maturation were unaltered following exposure. In conclusion, although LCs or DCs have the potential to acquire the scrapie agent within the epidermis our data suggest it is unlikely that they become activated and stimulated to transport the agent to the draining lymph node

    A Diagram Is Worth A Dozen Images

    Diagrams are common tools for representing complex concepts, relationships and events, often when it would be difficult to portray the same information with natural images. Understanding natural images has been extensively studied in computer vision, while diagram understanding has received little attention. In this paper, we study the problem of diagram interpretation and reasoning, the challenging task of identifying the structure of a diagram and the semantics of its constituents and their relationships. We introduce Diagram Parse Graphs (DPG) as our representation to model the structure of diagrams. We define syntactic parsing of diagrams as learning to infer DPGs for diagrams and study semantic interpretation and reasoning of diagrams in the context of diagram question answering. We devise an LSTM-based method for syntactic parsing of diagrams and introduce a DPG-based attention model for diagram question answering. We compile a new dataset of diagrams with exhaustive annotations of constituents and relationships for over 5,000 diagrams and 15,000 questions and answers. Our results show the significance of our models for syntactic parsing and question answering in diagrams using DPGs

    Combined effects of anisotropy and tension-compression asymmetry on the torsional response of AZ31 Mg

    In this paper it is demonstrated that only by accounting for the combined effects of anisotropy and tension-compression asymmetry at polycrystal level, it is possible to explain and accurately predict the room-temperature torsional response of a strongly textured AZ31 Mg material. This is shown by using two modeling frameworks, namely: a viscoplastic self-consistent (VPSC) polycrystal model, and a macroscopic plasticity model based on an yield criterion, developed by Cazacu et al. (2006), that accounts for both orthotropy and tension-compression asymmetry in plastic flow. It is shown that unlike Hill's (1948) criterion, the latter macroscopic criterion quantitatively predicts the experimental results, namely: that the sample with axial direction along the rolling direction contracts, while the sample with axial direction along the normal direction elongates. Moreover, it is demonstrated that these experimentally observed axial strain effects can be quantitatively predicted with the VPSC polycrystal model, only if both slip and twinning are considered operational at single crystal level. On the other hand, if it is assumed that the plastic deformation is fully accommodated by crystallographic slip, the axial strains predicted by VPSC are very close with that predicted with Hill (1948) criterion, which largely underestimates the measured axial strain in the rolling direction, and predicts zero axial strain in the normal direction.     We study continuous analogues of "vitality" for discrete network flows/paths, and consider problems related to placing segment barriers that have highest impact on a flow/path in a polygonal domain. This extends the graph-theoretic notion of "most vital arcs" for flows/paths to geometric environments. We give hardness results and efficient algorithms for various versions of the problem, (almost) completely separating hard and polynomially-solvable cases

    Gravel pits support waterbird diversity in an urban landscape

    We assessed the benefit of 11 gravel pits for the settlement of waterbird communities in an urbanized area lacking natural wetlands. Gravel pits captured 57% of the regional species pool of aquatic birds. We identified 39 species, among which five were regionally rare. We used the Self Organizing Map algorithm to calculate the probabilities of presence of species, and to bring out habitat conditions that predict assemblage patterns. The age of the pits did not correlate with assemblage composition and species richness. There was a positive influence of macrophyte cover on waterbird species richness. Larger pits did not support more species, but species richness increased with connectivity. As alternative wetland habitats, gravel pits are attractive to waterbirds, when they act as stepping stones that ensure connectivity between larger natural and/or artificial wetlands separated in space

    Adaptive Optics for Astronomy

    Adaptive Optics is a prime example of how progress in observational astronomy can be driven by technological developments. At many observatories it is now considered to be part of a standard instrumentation suite, enabling ground-based telescopes to reach the diffraction limit and thus providing spatial resolution superior to that achievable from space with current or planned satellites. In this review we consider adaptive optics from the astrophysical perspective. We show that adaptive optics has led to important advances in our understanding of a multitude of astrophysical processes, and describe how the requirements from science applications are now driving the development of the next generation of novel adaptive optics techniques.Comment: to appear in ARA&A vol 50, 201

    Evidence of a high incidence of subclinically affected calves in a herd of cattle with fatal cases of Bovine Neonatal Pancytopenia (BNP).

    BACKGROUND: Bovine Neonatal Pancytopenia (BNP) is a disease of calves characterised by bone marrow trilineage hypoplasia, mediated by ingestion of alloantibodies in colostrum. Suspected subclinical forms of BNP have been reported, suggesting that observed clinical cases may not represent the full extent of the disease. However to date there are no objective data available on the incidence of subclinical disease or its temporal distribution. This study aimed to 1) ascertain whether subclinical BNP occurs and, if so, to determine the incidence on an affected farm and 2) determine whether there is evidence of temporal clustering of BNP cases on this farm. To achieve these aims, haematological screening of calves born on the farm during one calving season was carried out, utilising blood samples collected at defined ages. These data were then analysed in comparison to data from both known BNP-free control animals and histopathologically confirmed BNP cases. An ordinal logistic regression model was used to create a composite haematology score to predict the probabilities of calves being normal, based on their haematology measurements at 10–14 days old. RESULTS: This study revealed that 15% (21 of 139) of the clinically normal calves on this farm had profoundly abnormal haematology (<5% chance of being normal) and could be defined as affected by subclinical BNP. Together with clinical BNP cases, this gave the study farm a BNP incidence of 18%. Calves with BNP were found to be distributed throughout the calving period, with no clustering, and no significant differences in the date of birth of cases or subclinical cases were found compared to the rest of the calves. This study did not find any evidence of increased mortality or increased time from birth to sale in subclinical BNP calves but, as the study only involved a single farm and adverse effects may be determined by other inter-current diseases it remains possible that subclinical BNP has a detrimental impact on the health and productivity of calves under certain circumstances. CONCLUSIONS: Subclinical BNP was found to occur at a high incidence in a herd of cattle with fatal cases of BNP

    Renormalization of minimally doubled fermions

    We investigate the renormalization properties of minimally doubled fermions, at one loop in perturbation theory. Our study is based on the two particular realizations of Borici-Creutz and Karsten-Wilczek. A common feature of both formulations is the breaking of hyper-cubic symmetry, which requires that the lattice actions are supplemented by suitable counterterms. We show that three counterterms are required in each case and determine their coefficients to one loop in perturbation theory. For both actions we compute the vacuum polarization of the gluon. It is shown that no power divergences appear and that all contributions which arise from the breaking of Lorentz symmetry are cancelled by the counterterms. We also derive the conserved vector and axial-vector currents for Karsten-Wilczek fermions. Like in the case of the previously studied Borici-Creutz action, one obtains simple expressions, involving only nearest-neighbour sites. We suggest methods how to fix the coefficients of the counterterms non-perturbatively and discuss the implications of our findings for practical simulations.Comment: 23 pages, 1 figur

    b-Initiated processes at the LHC: a reappraisal

    Several key processes at the LHC in the standard model and beyond that involve bb quarks, such as single-top, Higgs, and weak vector boson associated production, can be described in QCD either in a 4-flavor or 5-flavor scheme. In the former, bb quarks appear only in the final state and are typically considered massive. In 5-flavor schemes, calculations include bb quarks in the initial state, are simpler and allow the resummation of possibly large initial state logarithms of the type logQ2mb2\log \frac{{\cal Q}^2}{m_b^2} into the bb parton distribution function (PDF), Q{\cal Q} being the typical scale of the hard process. In this work we critically reconsider the rationale for using 5-flavor improved schemes at the LHC. Our motivation stems from the observation that the effects of initial state logs are rarely very large in hadron collisions: 4-flavor computations are pertubatively well behaved and a substantial agreement between predictions in the two schemes is found. We identify two distinct reasons that explain this behaviour, i.e., the resummation of the initial state logarithms into the bb-PDF is relevant only at large Bjorken xx and the possibly large ratios Q2/mb2{\cal Q}^2/m_b^2's are always accompanied by universal phase space suppression factors. Our study paves the way to using both schemes for the same process so to exploit their complementary advantages for different observables, such as employing a 5-flavor scheme to accurately predict the total cross section at NNLO and the corresponding 4-flavor computation at NLO for fully exclusive studies.Comment: Fixed typo in Eq.
