Rabl's model of the interphase chromosome arrangement tested in Chinise hamster cells by premature chromosome condensation and laser-UV-microbeam experiments
In 1885 Carl Rabl published his theory on the internal structure of the interphase nucleus. We have tested two predictions of this theory in fibroblasts grown in vitro from a female Chinese hamster, namely (1) the Rabl-orientation of interphase chromosomes and (2) the stability of the chromosome arrangement established in telophase throughout the subsequent interphase. Tests were carried out by premature chromosome condensation (PCC) and laser-UV-microirradiation of the interphase nucleus. Rabl-orientation of chromosomes was observed in G1 PCCs and G2 PCCs. The cell nucleus was microirradiated in G1 at one or two sites and pulse-labelled with 3H-thymidine for 2h. Cells were processed for autoradiography either immediately thereafter or after an additional growth period of 10 to 60h. Autoradiographs show unscheduled DNA synthesis (UDS) in the microirradiated nuclear part(s). The distribution of labelled chromatin was evaluated in autoradiographs from 1035 cells after microirradiation of a single nuclear site and from 253 cells after microirradiation of two sites. After 30 to 60h postincubation the labelled regions still appeared coherent although the average size of the labelled nuclear area fr increased from 14.2% (0h) to 26.5% (60h). The relative distance dr, i.e. the distance between two microirradiated sites divided by the diameter of the whole nucleus, showed a slight decrease with increasing incubation time. Nine metaphase figures were evaluated for UDS-label after microirradiation of the nuclear edge in G1. An average of 4.3 chromosomes per cell were labelled. Several chromosomes showed joint labelling of both distal chromosome arms including the telomeres, while the centromeric region was free from label. This label pattern is interpreted as the result of a V-shaped orientation of these particular chromosomes in the interphase nucleus with their telomeric regions close to each other at the nuclear edge. Our data support the tested predictions of the Rabl-model. Small time-dependent changes of the nuclear space occupied by single chromosomes and of their relative positions in the interphase nucleus seem possible, while the territorial organization of interphase chromosomes and their arrangement in general is maintained during interphase. Analysis of chromosome positions in the interphase nucleus of Chinese hamster cells by laser-UV-microirradiation experiments
Unsynchronized cells of an essentially diploid strain of female Chinese hamster cells derived from lung tissue (CHL) were laser-UV-microirradiated (=257 nm) in the nucleus either at its central part or at its periphery. After 7–9 h postincubation with 0.5 mM caffeine, chromosome preparations were made in situ. Twenty-one and 29 metaphase spreads, respectively, with partial chromosome shattering (PCS) obtained after micro-irradiation at these two nuclear sites, were Q-banded and analyzed in detail. A positive correlation was observed between the frequency of damage of chromosomes and both their DNA content and length at metaphase. No significant difference was observed between the frequencies of damage obtained for individual chromosomes at either site of microirradiation. The frequency of joint damage of homologous chromosomes was low as compared to nonhomologous ones. Considerable variation was noted in different cells in the combinations of jointly shattered chromosomes. Evidence which justifies an interpretation of these data in terms of an interphase arrangement of chromosome territories is discussed. Our data strongly argue against somatic pairing as a regular event, and suggest a considerable variability of chromosome positions in different nuclei. However, present data do not exclude the possibility of certain non-random chromosomal arrangements in CHL-nuclei. The interphase chromosome distribution revealed by these experiments is compared with centromere-centromere, centromere-center and angle analyses of metaphase spreads and the relationship between interphase and metaphase arrangements of chromosomes is discussed
Evolutionary game theory in growing populations
Existing theoretical models of evolution focus on the relative fitness
advantages of different mutants in a population while the dynamic behavior of
the population size is mostly left unconsidered. We here present a generic
stochastic model which combines the growth dynamics of the population and its
internal evolution. Our model thereby accounts for the fact that both
evolutionary and growth dynamics are based on individual reproduction events
and hence are highly coupled and stochastic in nature. We exemplify our
approach by studying the dilemma of cooperation in growing populations and show
that genuinely stochastic events can ease the dilemma by leading to a transient

Subnuclear localization, rates and effectiveness of UVC-induced unscheduled DNA synthesis visualized by fluorescence widefield, confocal and super-resolution microscopy
Unscheduled DNA synthesis (UDS) is the final stage of the process of repair of DNA lesions induced by UVC. We detected UDS using a DNA precursor, 5-ethynyl-2′-deoxyuridine (EdU). Using wide-field, confocal and super-resolution fluorescence microscopy and normal human fibroblasts, derived from healthy subjects, we demonstrate that the sub-nuclear pattern of UDS detected via incorporation of EdU is different from that when BrdU is used as DNA precursor. EdU incorporation occurs evenly throughout chromatin, as opposed to just a few small and large repair foci detected by BrdU. We attribute this difference to the fact that BrdU antibody is of much larger size than EdU, and its accessibility to the incorporated precursor requires the presence of denatured sections of DNA. It appears that under the standard conditions of immunocytochemical detection of BrdU only fragments of DNA of various length are being denatured. We argue that, compared with BrdU, the UDS pattern visualized by EdU constitutes a more faithful representation of sub-nuclear distribution of the final stage of nucleotide excision repair induced by UVC. Using the optimized integrated EdU detection procedure we also measured the relative amount of the DNA precursor incorporated by cells during UDS following exposure to various doses of UVC. Also described is the high degree of heterogeneity in terms of the UVC-induced EdU incorporation per cell, presumably reflecting various DNA repair efficiencies or differences in the level of endogenous dT competing with EdU within a population of normal human fibroblasts
Detection of chromosome aberrations in metaphase and interphase tumor cells by in situ hybridization using chromosome-specific library probes
Chromosome aberrations in two glioma cell lines were analyzed using biotinylated DNA library probes that specifically decorate chromosomes 1, 4, 7, 18 and 22 from pter to qter. Numerical changes, deletions and rearrangements of these chromosomes were radily visualized in metaphase spreads, as well as in early prophase and interphase nuclei. Complete chromosomes, deleted chromosomes and segments of translocated chromosomes were rapidly delineated in very complex karyotypes. Simultaneous hybridizations with additional subregional probes were used to further define aberrant chromosomes. Digital image analysis was used to quantitate the total complement of specific chromosomal DNAs in individual metaphase and interphase cells of each cell line. In spite of the fact that both glioma lines have been passaged in vitro for many years, an under-representation of chromosome 22 and an over-representation of chromosome 7 (specifically 7p) were observed. These observations agree with previous studies on gliomas. In addition, sequences of chromosome 4 were also found to be under-represented, especially in TC 593. These analyses indicate the power of these methods for pinpointing chromosome segments that are altered in specific types of tumors
Reference standardization and triglyceride interference of a new homogeneous HDL-cholesterol assay compared with a former chemical precipitation assay
A homogeneous HDL-c assay (HDL-H), which uses polyethylene glycol-modified
enzymes and sulfated alpha-cyclodextrin, was assessed for precision,
accuracy, and cholesterol and triglyceride interference. In addition, its
analytical performance was compared with that of a phosphotungstic acid
(PTA)/MgCl2 precipitation method (HDL-P). Within-run CVs were < or =
1.87%; total CVs were < or = 3.08%. Accuracy was evaluated in fresh
normotriglyceridemic sera using the Designated Comparison Method (HDL-H =
1.037 Designated Comparison Method + 4 mg/L; n = 63) and in moderately
hypertriglyceridemic sera by using the Reference Method (HDL-H = 1.068
Reference Method - 17 mg/L; n = 41). Mean biases were 4.5% and 2.2%,
respectively. In hypertriglyceridemic sera (n = 85), HDL-H concentrations
were increasingly positively biased with increasing triglyceride
concentrations. The method comparison between HDL-H and HDL-P yielded the
following equation: HDL-H = 1.037 HDL-P + 15 mg/L; n = 478. We conclude
that HDL-H amply meets the 1998 NCEP recommendations for total error; its
precision is superior compared with that of HDL-P, and its average bias
remains below +/-5% as long as triglyceride concentrations are < or = 10
g/L and in case of moderate hypercholesterolemia
A passion for respect: On understanding the role of human needs and morality
In the present paper, we stress the importance of the concept respect in a wide variety of social settings and provide a working definition of this concept by emphasizing how respect relates to the act of communicating full recognition to other people on the dimensions of belongingness and morality. Subsequently, in two separate parts, we discuss why respect is so desired and valued. The first part looks at respect as a means to fulfil important human social concerns (“respect as a means to an end”). The second part looks at the potential moral underpinnings of respect and thus interprets “respect as an end in itself.” Finally, it is suggested that both reasons to value respect explain respect effects as a function of the working selfconcept that is salient (i.e., pragmatic versus idealistic self)

Modern pollen-vegetation relationships along a steep temperature gradient in the Tropical Andes of Ecuador

The final version is available from Cambridge University Press via the DOI in this record.The characterization of modern pollen rain assemblages along environmental gradients is an essential prerequisite for reliable interpretations of fossil pollen records. In this study, we identify pollen-vegetation relationships using modern pollen rain assemblages in moss polsters (n = 13) and lake sediment surface samples (n = 11) along a steep temperature gradient of 7°C (3100–4200 m above sea level) on the western Andean Cordillera, Ecuador. The pollen rain is correlated to vascular plant abundance data recorded in vegetation relevées (n = 13). Results show that pollen spectra from both moss polsters and sediment surface samples reflect changes in species composition along the temperature gradient, despite overrepresentation of upper montane forest taxa in the latter. Estimated pollen transport distance for a lake (Laguna Llaviucu) situated in a steep upper montane forest valley is 1–2 km, while a lake (Laguna Pallcacocha) in the páramo captures pollen input from a distance of up to 10–40 km. Weinmannia spp., Podocarpus spp., and Hedyosmum sp. are indicators of local upper montane forest vegetation, while Phlegmariurus spp. and Plantago spp. are indicators for local páramo vegetation.Earth and Life Science council (ALW) of the Netherlands Organisation of Scientific Researc
