P-576: Differential effects of selective cyclooxygenase-2 inhibitors on endothelial function in salt-induced hypertension
In view of the ongoing controversy of potential differences in cardiovascular safety of selective COX-2 inhibitors (coxibs), we compared the effects of two different coxibs and a traditional NSAID on endothelial dysfunction, a well established surrogate of cardiovascular disease, in salt-induced hypertension. Salt-sensitive (DS) and salt-resistant (DR) Dahl rats were treated with a high-sodium diet (4% NaCl) for 56 days. From days 35 to 56, diclofenac (6 mg/kg/d; DS-diclofenac), rofecoxib (2 mg/kg/d; DS-rofecoxib), celecoxib (25 mg/kg/d; DS-celecoxib) or placebo (DS-placebo) were added to the chow. Vascular reactivity of isolated aortic rings was assessed by isometric tension recording. Blood pressure increased with high sodium diet in the DS-groups which was more pronounced after diclofenac and rofecoxib treatment (p<0.005 vs DS-placebo), but slightly blunted by celecoxib (p<0.001 vs DS-placebo). Sodium diet markedly reduced NO-mediated endothelium-dependent relaxations to acetylcholine (ACh, 10−10−10−5 mol/L) in untreated hypertensive rats (p<0.0001 vs DR-placebo). Relaxation to ACh improved after celecoxib (p<0.005 vs DS-placebo and DS-rofecoxib), but remained unchanged after rofecoxib and diclofenac treatment. Vasoconstriction after NOS inhibition with Nω-Nitro-L-Arginine Methyl Ester (10-4 mol/L) was blunted in DS rats (p<0.05 vs DR-placebo), normalized by celecoxib, but not affected by rofecoxib or diclofenac. Protein expression of eNOS was decreased in DS rats with a trend for increased eNOS levels in the DS-celecoxib group (97.8±25.6 vs 54.8±2.8 %, p=0.088 vs DS-placebo). Indicators of oxidative stress, 8-isoprostane levels, were elevated in untreated DS rats on 4% NaCl (6.55±0.58 vs 3.65±1.05 ng/ml, p<0.05) and normalized by celecoxib only (4.29±0.58 ng/ml), while SOD protein expression was decreased in DS rats and not affected by any treatment. Plasma levels of prostaglandines did not change during high sodium diet or any treatment. These data show that celecoxib, but not rofecoxib or diclofenac, improves endothelial dysfunction and reduces oxidative stress, thus pointing to differential effects of coxibs in salt-sensitive hypertension. Identification of a Proliferation Gene Cluster Associated with HPV E6/E7 Expression Level and Viral DNA Load in Invasive Cervical Carcinoma
Specific HPV DNA sequences are associated with more than 90% of invasive
carcinomas of the uterine cervix. Viral E6 and E7 oncogenes are key mediators
in cell transformation by disrupting TP53 and RB pathways. To investigate
molecular mechanisms involved in the progression of invasive cervical
carcinoma, we performed a gene expression study on cases selected according to
viral and clinical parameters. Using Coupled Two-Way Clustering and Sorting
Points Into Neighbourhoods methods, we identified a Cervical Cancer
Proliferation Cluster composed of 163 highly correlated transcripts, many of
which corresponded to E2F pathway genes controlling cell proliferation, whereas
no primary TP53 targets were present in this cluster. The average expression
level of the genes of this cluster was higher in tumours with an early relapse
than in tumours with a favourable course (P=0.026). Moreover, we found that
E6/E7 mRNA expression level was positively correlated with the expression level
of the cluster genes and with viral DNA load. These findings suggest that HPV
E6/E7 expression level plays a key role in the progression of invasive
carcinoma of the uterine cervix via the deregulation of cellular genes
controlling tumour cell proliferation. HPV expression level may thus correspond
to a biological marker useful for prognosis assessment and specific therapy of
the disease
Keep it SMPL: Automatic Estimation of 3D Human Pose and Shape from a Single Image
We describe the first method to automatically estimate the 3D pose of the
human body as well as its 3D shape from a single unconstrained image. We
estimate a full 3D mesh and show that 2D joints alone carry a surprising amount
of information about body shape. The problem is challenging because of the
complexity of the human body, articulation, occlusion, clothing, lighting, and
the inherent ambiguity in inferring 3D from 2D. To solve this, we first use a
recently published CNN-based method, DeepCut, to predict (bottom-up) the 2D
body joint locations. We then fit (top-down) a recently published statistical
body shape model, called SMPL, to the 2D joints. We do so by minimizing an
objective function that penalizes the error between the projected 3D model
joints and detected 2D joints. Because SMPL captures correlations in human
shape across the population, we are able to robustly fit it to very little
data. We further leverage the 3D model to prevent solutions that cause
interpenetration. We evaluate our method, SMPLify, on the Leeds Sports,
HumanEva, and Human3.6M datasets, showing superior pose accuracy with respect

Alteration of Multiple Leukocyte Gene Expression Networks is Linked with Magnetic Resonance Markers of Prognosis After Acute ST-Elevation Myocardial Infarction
Prognostic relevant pathways of leukocyte involvement in human myocardial ischemic-reperfusion injury are largely unknown. We enrolled 136 patients with ST-elevation myocardial infarction (STEMI) after primary angioplasty within 12 h after onset of symptoms. Following reperfusion, whole blood was collected within a median time interval of 20 h (interquartile range: 15-25 h) for genome-wide gene expression analysis. Subsequent CMR scans were performed using a standard protocol to determine infarct size (IS), area at risk (AAR), myocardial salvage index (MSI) and the extent of late microvascular obstruction (lateMO). We found 398 genes associated with lateMO and two genes with IS. Neither AAR, nor MSI showed significant correlations with gene expression. Genes correlating with lateMO were strongly related to several canonical pathways, including positive regulation of T-cell activation (p = 3.44 x 10(-5)), and regulation of inflammatory response (p = 1.86 x 10(-3)). Network analysis of multiple gene expression alterations associated with larger lateMO identified the following functional consequences: facilitated utilisation and decreased concentration of free fatty acid, repressed cell differentiation, enhanced phagocyte movement, increased cell death, vascular disease and compensatory vasculogenesis. In conclusion, the extent of lateMO after acute, reperfused STEMI correlated with altered activation of multiple genes related to fatty acid utilisation, lymphocyte differentiation, phagocyte mobilisation, cell survival, and vascular dysfunction
Overcoming global inequality is critical for land-based mitigation in line with the Paris Agreement
Transformation pathways for the land sector in line with the Paris Agreement depend on the assumption of globally implemented greenhouse gas (GHG) emission pricing, and in some cases also on inclusive socio-economic development and sustainable land-use practices. In such pathways, the majority of GHG emission reductions in the land system is expected to come from low- and middle-income countries, which currently account for a large share of emissions from agriculture, forestry and other land use (AFOLU). However, in low- and middle-income countries the economic, financial and institutional barriers for such transformative changes are high. Here, we show that if sustainable development in the land sector remained highly unequal and limited to high-income countries only, global AFOLU emissions would remain substantial throughout the 21st century. Our model-based projections highlight that overcoming global inequality is critical for land-based mitigation in line with the Paris Agreement. While also a scenario purely based on either global GHG emission pricing or on inclusive socio-economic development would achieve the stringent emissions reductions required, only the latter ensures major co-benefits for other Sustainable Development Goals, especially in low- and middle-income regions. © 2022, The Author(s)
A semi-schematic model for the center of mass dynamics in supercooled molecular liquids
We introduce a semi-schematic mode-coupling model to describe the slow
dynamics in molecular liquids, retaining explicitly only the description of the
center of mass degrees of freedom. Angular degrees of freedom are condensed in
a q-vector independent coupling parameter. We compare the time and q-dependence
of the density fluctuation correlators with numerical data from a 250 ns long
molecular dynamics simulation. Notwithstanding the choice of a network-forming
liquid as a model for comparing theory and simulation, the model describes the
main static and dynamic features of the relaxation in a broad q-vector range.Comment: Revtex, 2 figure
