    Structural Basis for the Antiviral Activity of BST-2/Tetherin and Its Viral Antagonism

    The interferon-inducible host restriction factor bone marrow stromal antigen 2 (BST-2/tetherin) blocks the release of HIV-1 and other enveloped viruses. In turn, these viruses have evolved specific antagonists to counteract this host antiviral molecule, such as the HIV-1 protein Vpu. BST-2 is a type II transmembrane protein with an unusual topology consisting of an N-terminal cytoplasmic tail (CT) followed by a single transmembrane (TM) domain, a coiled-coil extracellular (EC) domain, and a glycosylphosphatidylinositol (GPI) anchor at the C terminus. We and others showed that BST-2 restricts enveloped virus release by bridging the host and virion membranes with its two opposing membrane anchors and that deletion of either one completely abrogates antiviral activity. The EC domain also shows conserved structural properties that are required for antiviral function. It contains several destabilizing amino acids that confer the molecule with conformational flexibility to sustain the protein’s function as a virion tether, and three conserved cysteine residues that mediate homodimerization of BST-2, as well as acting as a molecular ruler that separates the membrane anchors. Conversely, the efficient release of virions is promoted by the HIV-1 Vpu protein and other viral antagonists. Our group and others provided evidence from mutational analyses indicating that Vpu antagonism of BST-2-mediated viral restriction requires a highly specific interaction of their mutual TM domains. This interpretation is further supported and expanded by the findings of the latest structural modeling studies showing that critical amino acids in a conserved helical face of these TM domains are required for Vpu–BST-2 interaction and antagonism.     Discovery of a new pulsating X-ray source with a 1549.1-s period, AX J183220-0840

    A new pulsating X-ray source, AX J183220-0840, with a 1549.1-s period was discovered at R.A.= 18h32m20s and Dec.=-8d40'30'' (J2000, uncertainty=0.6degree) during an ASCA observation on the Galactic plane. The source was observed two times, in 1997 and in 1999. A phase-averaged X-ray flux of 1.1E-11 ergs cm-2 s-1 and pulsation period of 1549.1+/-0.4 s were consistently obtained from these two observations. The X-ray spectrum was represented by a flat absorbed power-law with a photon-index of =~0.8 and an absorption column density of =~1.3E22 cm-2. Also, a signature of iron K-shell line emission with a centroid of 6.7 keV and an equivalent width of approximately 450 eV was detected. From the pulsation period and the iron-line feature, AX J183220-0840 is likely to be a magnetic white dwarf binary with a complexly absorbed thermal spectrum with a temperature of about 10 keV.Comment: 13 pages, 4 figures, accepted for publication in ApJ Letter

    Differences in serum IL-6 response after 1°C rise in core body temperature in individuals with spinal cord injury and cervical spinal cord injury during local heat stress

    Objectives: Passive rise in core body temperature achieved by head-out hot water immersion (HHWI) results in acute increases in serum interleukin (IL)-6 but no change in plasma adrenaline in patients with cervical spinal cord injury (CSCI). The purpose of the present study was to determine the mechanism of heat stress-induced increase in serum IL-6. Setting: A cross-sectional study. Methods: The study subjects were 9 with CSCI, 10 with thoracic and lumbar spinal cord injury (TLSCI) and 8 able-bodied (AB) subjects. Time since injury was 16.4±4.1 years in TLSCI and 16.1±3.4 years in CSCI. Subjects were subjected to lower-body heat stress (LBH) by wearing a hot water-perfused suit until 1°C increase in core temperature. The levels of serum IL-6, plasma adrenaline, tumor necrosis factor (TNF)-α, C-reactive protein (CRP), and counts of blood cells were measured at normothermia and after LBH. Results: Serum IL-6 concentrations increased significantly immediately after LBH in all the three groups. ΔIL-6% was lower in CSCI subjects compared with AB subjects. Plasma adrenaline concentrations significantly increased after LBH in AB and TLSCI subjects, but did not change throughout the study in CSCI subjects. Cardiac output and heart rate increased at the end of LBH in all three groups. Conclusion: Under a similar increase in core temperature, ΔIL-6% was lower in the CSCI group compared with the AB group. These findings suggest that the observed rise in IL-6 during hyperthermia is mediated, at least in part, by plasma adrenaline

    Weltrohstoffversorgung: Konflikt oder Kooperation?

    Unter dem Eindruck der jüngsten Preishausse auf den internationalen Rohstoffmärkten wird nunmehr in vielen Ländern befürchtet, daß die Welt an der Schwelle zunehmender Schwierigkeiten bei der Versorgung mit mineralischen und landwirtschaftlichen Rohstoffen einschließlich Nahrungsmitteln steht. Der kräftige Preisanstieg bei diesen Rohstoffen war jedoch überwiegend konjunkturell bedingt. Bei Getreide verursachten Mißernten unerwartete Produktionsausfälle größeren Ausmaßes. Wirksame Preisabsprachen zwischen Produzentenländern hat es, anders als bei Erdöl, entgegen einer verbreiteten Meinung nicht gegeben. Längerfristig wird sich das Preisniveau auf den Rohstoffmärkten, wenn auch nicht drastisch, so doch kontinuierlich erhöhen. Preisfluktuationen um den Trend werden weiterhin diese Märkte kennzeichnen. Ernsthafte Rohstoffverknappungen brauchen für den Rest dieses Jahrhunderts nicht erwartet zu werden. So übersteigen z. B. im Falle zahlreicher nicht-regenerierbarer NE-Metalle die bekannten Weltreserven deutlich den Weltbedarf; bei tropischen Ernährungsgütern und bei Getreide ist der Spielraum für eine nachhaltige Erhöhung der Flächenerträge noch sehr groß, vor allem in Entwicklungsländern. Die Wahrscheinlichkeit für die Bildung von Produzentenkartellen in der Zukunft ist gering. Käme es dennoch zu Kartellabsprachen, so wären diese bestenfalls kurzfristig erfolgreich. Das gleiche gilt für Exportbeschränkungen, die in Zeiten knappen Angebots sensibler Produkte (z. B. Getreide) den internen Preisanstieg dämpfen sollen. Angesichts der verbreiteten Knappheitspsychose kann nicht ausgeschlossen werden, daß es im Falle von temporären Angebotsdefiziten zu Überreaktionen kommt, die erhebliche Gefahren für den freien und multilateralen Welthandel in sich bergen. Solchen Gefahren kann nur erfolgreich durch partnerschaftliche Zusammenarbeit, nicht jedoch durch nationale Alleingänge, begegnet werden. Erforderlich sind ein international vereinbartes System von Verhaltensregeln zur Sicherung des undiskriminierten Zugangs zu Rohstoffmärkten bei angemessenen Preisen, die koordinierte Bildung von Rohstoffvorräten, internationale Rohstoffabkommen zwischen Produzenten- und Verbraucherländern, Regelungen zur Verbesserung des allgemeinen Investitionsklimas im Rohstoffbereich und der Abbau der Handelsschranken der Industrieländer gegenüber Exporten landwirtschaftlicher Produkte sowie rohstoff- und/oder arbeitsintensiver Halb- und Fertigwaren aus Entwicklungsländern. --

    Welthandel und Strukturanpassung in den Industrieländern

    Ziel der sechsten GATT-Runde ist es, die Schranken im Welthandel weiter abzubauen. Anders als bei den fünf früheren GATT-Runden herrscht dieses Mal in vielen Ländern Skepsis gegenüber einer weiteren Liberalisierung, weil man die Risiken, die mit zunehmendem Importwettbewerb verbunden sein können, höher bewertet als die möglichen Vorteile der Liberalisierung. Das Aufrechterhalten von Handelsschranken kostet Ressourcen. Die Protektion wenig effizienter Industrien in den wohlhabenden Ländern hält deren Einkommen unter dem Niveau, das sie sonst erreichen könnten und verzögert das Tempo der Industrialisierung in den Entwicklungsländern. Die ökonomischen Argumente zugunsten eines freieren Handels sind stark und weitgehend unanfechtbar. Handelsbeschränkungen sind zudem politisch belastend. Es gibt eine Reihe staatlicher Maßnahmen, die geeignet sind, die Schocks einer Anpassung an wirtschaftliche Veränderungen zu mindern. Interne Anpassungmaßnahmen spielen daher eine bedeutende Rolle in der künftigen Handelspolitik. Sehr zweifelhaft ist, ob alle Möglichkeiten für wirksame Anpassungsmaßnahmen bereits ausgeschöpft sind. In der Textilindustrie, der Stahlindustrie und im Kohlenbergbau existieren nach allgemeiner Meinung hartnäckige Anpassungsprobleme, die besondere und permanente Schutzmaßnahmen gegen den Importwettbewerb verlangen. Obwohl es viele Anstrengungen gegeben hat, den Status quo aufrechtzuerhalten, hat in allen drei Fällen die Dynamik des Strukturwandels die Lage grundlegend verändert. Diese Entwicklungstendenzen erlauben den Schluß, daß weitere Schritte zur Liberalisierung auch in den schwierigsten Fällen nicht nur wünschenswert wären, sondern wahrscheinlich durch den immanenten Strukturwandel in der Wirtschaft erleichtert werden. Eine Veränderung ist selten bequem, selbst dann nicht, wenn sie zu Verbesserungen führt. Für die Politik darf die relevante Frage nicht sein, wie man eine Anpassung an veränderte Umstände verlangsamen oder verhindern könnte. Sie lautet vielmehr, wie man die Chancen, die ein freierer Handel bietet, dadurch nutzen kann, daß man die Anpassung erleichtert und ihre Kosten minimiert. --

    TRY plant trait database - enhanced coverage and open access

    Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

    The ASTRO-H X-ray Observatory

    The joint JAXA/NASA ASTRO-H mission is the sixth in a series of highly successful X-ray missions initiated by the Institute of Space and Astronautical Science (ISAS). ASTRO-H will investigate the physics of the high-energy universe via a suite of four instruments, covering a very wide energy range, from 0.3 keV to 600 keV. These instruments include a high-resolution, high-throughput spectrometer sensitive over 0.3-2 keV with high spectral resolution of Delta E < 7 eV, enabled by a micro-calorimeter array located in the focal plane of thin-foil X-ray optics; hard X-ray imaging spectrometers covering 5-80 keV, located in the focal plane of multilayer-coated, focusing hard X-ray mirrors; a wide-field imaging spectrometer sensitive over 0.4-12 keV, with an X-ray CCD camera in the focal plane of a soft X-ray telescope; and a non-focusing Compton-camera type soft gamma-ray detector, sensitive in the 40-600 keV band.     Convergence in phosphorus constraints to photosynthesis in forests around the world

The source code for ORCHIDEE is at https://doi.org/10.14768/20200407002.1.Tropical forests take up more carbon (C) from the atmosphere per annum by photosynthesis than any other type of vegetation. Phosphorus (P) limitations to C uptake are paramount for tropical and subtropical forests around the globe. Yet the generality of photosynthesis-P relationships underlying these limitations are in question, and hence are not represented well in terrestrial biosphere models. Here we demonstrate the dependence of photosynthesis and underlying processes on both leaf N and P concentrations. The regulation of photosynthetic capacity by P was similar across four continents. Implementing P constraints in the ORCHIDEE-CNP model, gross photosynthesis was reduced by 36% across the tropics and subtropics relative to traditional N constraints and unlimiting leaf P. Our results provide a quantitative relationship for the P dependence for photosynthesis for the front-end of global terrestrial C models that is consistent with canopy leaf measurements
