    Structure of the magnetoionic medium around the FR Class I radio galaxy 3C 449

    The goal of this work is to constrain the strength and structure of the magnetic field associated with the environment of the radio source 3C 449, using observations of Faraday rotation, which we model with a structure function technique and by comparison with numerical simulations. We assume that the magnetic field is a Gaussian, isotropic random variable and that it is embedded in the hot intra-group plasma surrounding the radio source. For this purpose, we present detailed rotation measure images for the polarized radio source 3C 449, previously observed with the Very Large Array at seven frequencies between 1.365 and 8.385 GHz. We quantify the statistics of the magnetic-field fluctuations by deriving rotation measure structure functions, which we fit using models derived from theoretical power spectra. We quantify the errors due to sampling by making multiple two-dimensional realizations of the best-fitting power spectrum.We also use depolarization measurements to estimate the minimum scale of the field variations. We then make three-dimensional models with a gas density distribution derived from X-ray observations and a random magnetic field with this power spectrum. Under these assumptions we find that both rotation measure and depolarization data are consistent with a broken power-law magnetic-field power spectrum, with a break at about 11 kpc and slopes of 2.98 and 2.07 at smaller and larger scales respectively. The maximum and minimum scales of the fluctuations are around 65 and 0.2 kpc, respectively. The average magnetic field strength at the cluster centre is 3.5 +/-1.2 micro-G, decreasing linearly with the gas density within about 16 kpc of the nucleus.Comment: 19 pages; 14 figures; accepted for publication on A&A.     Vitamin e-loaded membrane dialyzers reduce hemodialysis inflammaging

    Background Inflammaging is a persistent, low-grade, sterile, nonresolving inflammatory state, associated with the senescence of the immune system. Such condition downregulates both innate and adaptive immune responses during chronic disorders as type II diabetes, cancer and hemodialysis, accounting for their susceptibility to infections, malignancy and resistance to vaccination. Aim of this study was to investigate hemodialysis inflammaging, by evaluating changes of several hemodialysis treatments on indoleamine 2,3-dioxygenase-1 activity and nitric oxide formation. Methods We conducted a randomized controlled observational crossover trial. Eighteen hemodialysis patients were treated with 3 different hemodialysis procedures respectively: 1) Low-flux bicarbonate hemodialysis, 2) Low-flux bicarbonate hemodialysis with vitamin E - loaded dialyzers, and 3) Hemodialfitration. The control group consisted of 14 hospital staff healthy volunteers. Blood samples were collected from all 18 hemodialysis patients just after the long interdialytic interval, at the end of each hemodialysis treatment period. Results Hemodialysis kynurenine and kynurenine/L - tryptophan blood ratio levels were significantly higher, when compared to the control group, indicating an increased indoleamine 2,3-dioxygenase-1 activity in hemodialysis patients. At the end of the low-flux bicarbonate hemodialysis with vitamin E - loaded dialyzers period, L - tryptophan serum levels remained unchanged vs both low-flux bicarbonate hemodialysis and hemodialfitration. Kynurenine levels instead decreased, resulting in a significant reduction of kynurenine/L - tryptophan blood ratio and indoleamine 2,3-dioxygenase-1 activity, when matched to both low-flux bicarbonate hemodialysis and HDF respectively. Serum nitric oxide control group levels, were significantly lower when compared to all hemodialysis patient groups. Interestingly, low-flux bicarbonate hemodialysis with vitamin E - loaded dialyzers nitric oxide serum levels from venous line blood samples taken 60 min after starting the hemodialysis session were significantly lower vs serum taken simultaneously from the arterial blood line. Conclusions The treatment with more biocompatible hemodialysis procedure as low-flux bicarbonate hemodialysis with vitamin E - loaded dialyzers, reduced indoleamine 2,3-dioxygenase-1 activity and nitric oxide formation when compared to both low-flux bicarbonate hemodialysis and hemodialfitration. These data suggest that low-flux bicarbonate hemodialysis with vitamin E - loaded dialyzers lowering hemodialysis inflammaging, could be associated to changes of proinflammatory signalling a regulated molecular level

    Humanity’s main societal and epistemic transitions also mirror changes in its approach to the food system. This particularly holds true for human–animal interactions and the consumption of animal source foods (red meat especially, and to a lesser degree dairy, eggs, poultry, and fish). Hunter-gathering has been by far the longest prevailing form of human sustenance, followed by a diffuse transition to crop agriculture and animal husbandry. This transition eventually stabilized as a state-controlled model based on the domestication of plants, animals, and humans. A shift to a post-domestic paradigm was initiated during the 19th century in the urbanizing populations of the Anglosphere, which was characterized by the rise of agri-food corporations, an increased meat supply, and a disconnect of most of its population from the food chain. While this has improved undernutrition, various global threats have been emerging in parallel. The latter include, among others, a public health crisis, climate change, pandemics, and societal class anxieties. This state of affairs is an unstable one, setting the conditions of possibility for a new episteme that may evolve beyond mere adjustments within the business-as-usual model. At least two disruptive scenarios have been described in current food discourses, both by scientists and mass media. Brought to its extreme, the first scenario relates to the radical abolishment of livestock, rewilding, a ‘plants-only’ diet, and vegan ideology. A second option consists of a holistic approach to animal husbandry, involving more harmonic and richer types of human–animal–land interactions. We argue that – instead of reactive pleas for less or none – future thoughtscapes should emphasize ‘more of the better.

    Infertility and hypergonadotropic hypogonadism as first evidence of hereditary apolipoprotein A-I amyloidosis

    Purpose: We report that primary infertility and hypergonadotropic hypogonadism in young patients may be caused by testicular amyloidosis and it is associated with the presence of a mutation in the apoA-I gene, resulting in the replacement of proline for leucine at residue 75 of the protein. Materials and Methods: Ten patients presenting with infertility, gynecomastia, decreased libido, erectile dysfunction or a family history of amyloidosis underwent clinical evaluation, hormone assays, semen analysis, ultrasonographic investigation of the testicles, testicular biopsy and DNA sequencing of the apoA-I gene. Results: All patients showed azoospermia and 9 had increased testicular volume. Massive amyloid deposition was observed in all testicular biopsies and the apoA-I mutation of replacement of proline for leucine at residue 75 of the protein was noted. Five patients showed hypergonadotropic hypogonadism and 5 had normal testosterone values with high gonadotropin levels. Conclusions: Nonobstructive azoospermia and macro-orchidism with or without hypogonadism may be caused by hereditary apoA-I amyloidosis in young patients. Testicular amyloidosis can be the first manifestation of this systemic disease. Specific staining for amyloid deposits and genetic analysis of apoA-I mutations are recommended in young, infertile patients with macro-orchidism. Finally, surveillance in asymptomatic mutation carriers is suggested to evaluate the opportunity to implement sperm retrieval and start androgen replacement therapy when necessary

    Shape of the Galactic Orbits in Clusters

    A kinematical analysis applied to a sample of galaxy clusters indicates that the differences between the velocity distribution of elliptical and spiral galaxies are associated with the shape of their orbit families. The orbital anisotropies present on each morphological population could be measured with the use of a parameter which is the ratio of the radial and tangential velocity dispersions, and can be recovered through the observed line-of-sight velocity distribution. When a Gaussian velocity distribution is assumed, having different dispersions along the radial and tangential directions, we conclude that the orbits of elliptical galaxies in clusters are close to radial, while spirals have more circular shaped or isotropic orbits.     Costimulatory Pathways in Kidney Transplantation: Pathogenetic Role, Clinical Significance and New Therapeutic Opportunities.

    Costimulatory pathways play a key role in immunity, providing the second signal required for a full activation of adaptive immune response. Different costimulatory families (CD28, TNF-related, adhesion and TIM molecules), characterized by structural and functional analogies, have been described. Costimulatory molecules modulate T cell activation, B cell function, Ig production, cytokine release and many other processes, including atherosclerosis. Patients suffering from renal diseases present significant alterations of the costimulatory pathways, which might make them particularly liable to infections. These alterations are further pronounced in patients undergoing kidney transplantation. In these patients, different costimulatory patterns have been related to distinct clinical features. The importance that costimulation has gained during the last years has led to development of several pharmacological approaches to modulate this critical step in the immune activation. Different drugs, mainly monoclonal antibodies targeting various costimulatory molecules (i.e. anti-CD80, CTLA-4 fusion proteins, anti-CD154, anti-CD40, etc.) were designed and tested in both experimental and clinical studies. The results of these studies highlighted some criticisms, but also some promising findings and now costimulatory blockade is considered a suitable strategy, with belatacept (a CTLA-4 fusion protein) being approved as the first costimulatory blocker for use in renal transplantation. In this review, we summarize the current knowledge on costimulatory pathways in the setting of kidney transplantation. We describe the principal costimulatory molecule families, their role and clinical significance in patients undergoing renal transplantation and the new therapeutic approaches that have been developed to modulate the costimulatory pathways

    Which criteria should we use to end isolation in hemodialysis patients with COVID-19?

    Safe and timely discontinuation of quarantine of in-center hemodialysis (HD) patients with a previous severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is a challenging issue for the nephrological community because current guidelines for ending isolation do not mention dialysis patients. To prevent potentially fatal outbreaks of coronavirus disease 2019 (COVID-19), a cautionary approach has been adopted by most dialysis units. The criteria for ending the isolation in the HD population generally coincide with those recommended for immunocompromised people. Thus, a test-based strategy relying on two consecutive negative reverse transcriptase-polymerase chain reaction (RT-PCR) nasopharyngeal swabs has been adopted to terminate quarantine. This strategy has the disadvantage of prolonging isolation as RT-PCR positivity does not equate to SARS-CoV-2 infectivity. Consequentially, prolonged positivity of SARS-CoV-2 results in excessive workload for the HD staff who must face an increasing number of COVID-19 patients requiring isolation. This condition leads also to serious implications for the patients and their households including work productivity loss, postponement of health-care appointments and an increased risk of COVID-19 reinfection. To counteract this problem, other diagnostic tests should be used to provide the best care to HD patients. Recent results seem to encourage the use of RT-PCR cycle threshold (Ct) values and rapid antigen tests given their better correlation with cell culture for SARS-CoV-2 than RT-PCR testing. Here, we provide an overview of the current scientific evidence on the tests used to verify the infectiousness of the virus in order to stimulate the nephrological community to adopt a streamlined and pragmatic procedure to end isolation in COVID-19 patients on HD

    Italian Science Case for ALMA Band 2+3

    The Premiale Project "Science and Technology in Italy for the upgraded ALMA Observatory - iALMA" has the goal of strengthening the scientific, technological and industrial Italian contribution to the Atacama Large Millimeter/submillimeter Array (ALMA), the largest ground based international infrastructure for the study of the Universe in the microwave. One of the main objectives of the Science Working Group (SWG) inside iALMA, the Work Package 1, is to develop the Italian contribution to the Science Case for the ALMA Band 2 or Band 2+3 receiver. ALMA Band 2 receiver spans from ~67 GHz (bounded by an opaque line complex of ozone lines) up to 90 GHz which overlaps with the lower frequency end of ALMA Band 3. Receiver technology has advanced since the original definition of the ALMA frequency bands. It is now feasible to produce a single receiver which could cover the whole frequency range from 67 GHz to 116 GHz, encompassing Band 2 and Band 3 in a single receiver cartridge, a so called Band 2+3 system. In addition, upgrades of the ALMA system are now foreseen that should double the bandwidth to 16 GHz. The science drivers discussed below therefore also discuss the advantages of these two enhancements over the originally foreseen Band 2 system.Comment: 43 pages, 21 figure

    Management of mineral metabolism in hemodialysis patients: discrepancy between interventions and perceived causes of failure

    none8BACKGROUND: Mineral and bone disorders (MBD) in patients undergoing hemodialysis (HD) are a major clinical complication. Current therapeutic strategies do not attain the expected results. The Italian audit on mineral metabolism was implemented to investigate MBD management through a "patient-oriented" approach. METHODS: Clinical and laboratory data pertinent to MBD from 509 prevalent adult patients on chronic HD were recorded and examined (audit), after which individual strategies were elaborated to improve MBD control. Their effectiveness was evaluated 6 months after the audit (Post-6). RESULTS: The audit disclosed poor MBD control in a high percentage of patients (56 %). Low compliance to treatment was the major determinant of failure (in 43.5 % of cases). Logistic regression showed a direct correlation between high degree of compliance and the achievement of therapeutic targets, e.g. parathyroid hormone: odds ratio (OR) 2.48, p = 0.015. In contrast, a minority of the proposed interventions (14.7 %) included strategies to improve patient compliance. At Post-6, despite a significant increase in drug prescription (p < 0.05 vs. audit), the rate of successful MBD control was unchanged.
