    ECONOMIC ANALYSIS OF TEMIK ON CITRUS IN THE INDIAN RIVER AREA IN SOUTHEASTERN FLORIDA

    Temik [R](aldicarb) is a pesticide labeled for use on several citrus crops to control rust mite, whitefly, nematode and brown citrus aphid pests. Analysis of previous research experiments indicates that this pesticide is beneficial to both orange and grapefruit production and that both cost savings and higher yields can be experienced in many types of groves. Actual grove data shows that net returns for mature grapefruit that receive Temik [R] can be $500 per acre greater than net returns for identical acreage that uses other pest control options. Also, based on grove reset data it is shown that with an application of Temik [R] the resulting increased yields for three-year-old trees more than cover the additional cost of applying the Temik [R].mature citrus, resets, revenue-cost, net return, grapefruit, Crop Production/Industries,

    The state of SQL-on-Hadoop in the cloud

    Managed Hadoop in the cloud, especially SQL-on-Hadoop, has been gaining attention recently. On Platform-as-a-Service (PaaS), analytical services like Hive and Spark come preconfigured for general-purpose and ready to use. Thus, giving companies a quick entry and on-demand deployment of ready SQL-like solutions for their big data needs. This study evaluates cloud services from an end-user perspective, comparing providers including: Microsoft Azure, Amazon Web Services, Google Cloud, and Rackspace. The study focuses on performance, readiness, scalability, and cost-effectiveness of the different solutions at entry/test level clusters sizes. Results are based on over 15,000 Hive queries derived from the industry standard TPC-H benchmark. The study is framed within the ALOJA research project, which features an open source benchmarking and analysis platform that has been recently extended to support SQL-on-Hadoop engines. The ALOJA Project aims to lower the total cost of ownership (TCO) of big data deployments and study their performance characteristics for optimization. The study benchmarks cloud providers across a diverse range instance types, and uses input data scales from 1GB to 1TB, in order to survey the popular entry-level PaaS SQL-on-Hadoop solutions, thereby establishing a common results-base upon which subsequent research can be carried out by the project. Initial results already show the main performance trends to both hardware and software configuration, pricing, similarities and architectural differences of the evaluated PaaS solutions. Whereas some providers focus on decoupling storage and computing resources while offering network-based elastic storage, others choose to keep the local processing model from Hadoop for high performance, but reducing flexibility.     High-resolution neutron and X-ray diffraction room-temperature studies of an H-FABP-oleic acid complex: study of the internal water cluster and ligand binding by a transferred multipolar electron-density distribution

    Crystal diffraction data of heart fatty acid binding protein (H-FABP) in complex with oleic acid were measured at room temperature with high-resolution X-ray and neutron protein crystallography (0.98 and 1.90 Å resolution, respectively). These data provided very detailed information about the cluster of water molecules and the bound oleic acid in the H-FABP large internal cavity. The jointly refined X-ray/neutron structure of H-FABP was complemented by a transferred multipolar electron-density distribution using the parameters of the ELMAMII library. The resulting electron density allowed a precise determination of the electrostatic potential in the fatty acid (FA) binding pocket. Bader's quantum theory of atoms in molecules was then used to study interactions involving the internal water molecules, the FA and the protein. This approach showed H···H contacts of the FA with highly conserved hydrophobic residues known to play a role in the stabilization of long-chain FAs in the binding cavity. The determination of water hydrogen (deuterium) positions allowed the analysis of the orientation and electrostatic properties of the water molecules in the very ordered cluster. As a result, a significant alignment of the permanent dipoles of the water molecules with the protein electrostatic field was observed. This can be related to the dielectric properties of hydration layers around proteins, where the shielding of electrostatic interactions depends directly on the rotational degrees of freedom of the water molecules in the interface.Facultad de Ciencias ExactasInstituto de Física de Líquidos y Sistemas Biológico

    Multiscale spatial mapping of cell populations across anatomical sites in healthy human skin and basal cell carcinoma

    \ua9 2024 National Academy of Sciences. All rights reserved.Our understanding of how human skin cells differ according to anatomical site and tumour formation is limited. To address this, we have created a multiscale spatial atlas of healthy skin and basal cell carcinoma (BCC), incorporating in vivo optical coherence tomography, single-cell RNA sequencing, spatial global transcriptional profiling, and in situ sequencing. Computational spatial deconvolution and projection revealed the localisation of distinct cell populations to specific tissue contexts. Although cell populations were conserved between healthy anatomical sites and in BCC, mesenchymal cell populations including fibroblasts and pericytes retained signatures of developmental origin. Spatial profiling and in silico lineage tracing support a hair follicle origin for BCC and demonstrate that cancer-associated fibroblasts are an expansion of a POSTN+ subpopulation associated with hair follicles in healthy skin. RGS5+ pericytes are also expanded in BCC suggesting a role in vascular remodelling. We propose that the identity of mesenchymal cell populations is regulated by signals emanating from adjacent structures and that these signals are repurposed to promote the expansion of skin cancer stroma.     Coding potential of the products of alternative splicing in human

    Background: Analysis of the human genome has revealed that as much as an order of magnitude more of the genomic sequence is transcribed than accounted for by the predicted and characterized genes. A number of these transcripts are alternatively spliced forms of known protein coding genes; however, it is becoming clear that many of them do not necessarily correspond to a functional protein. Results: In this study we analyze alternative splicing isoforms of human gene products that are unambiguously identified by mass spectrometry and compare their properties with those of isoforms of the same genes for which no peptide was found in publicly available mass spectrometry datasets. We analyze them in detail for the presence of uninterrupted functional domains, active sites as well as the plausibility of their predicted structure. We report how well each of these strategies and their combination can correctly identify translated isoforms and derive a lower limit for their specificity, that is, their ability to correctly identify non-translated products. Conclusions: The most effective strategy for correctly identifying translated products relies on the conservation of active sites, but it can only be applied to a small fraction of isoforms, while a reasonably high coverage, sensitivity and specificity can be achieved by analyzing the presence of non-truncated functional domains. Combining the latter with an assessment of the plausibility of the modeled structure of the isoform increases both coverage and specificity with a moderate cost in terms of sensitivity

    Longitudinal evaluation of quality of life in 288 patients with Neurofibromatosis 2

    Advances in molecular biology have resulted in novel therapy for neurofibromatosis 2-related (NF2) tumours, highlighting the need for robust outcome measures. The disease-focused NF2 impact on quality of life (NFTI-QOL) patient questionnaire was assessed as an outcome measure for treatment in a multi-centre study. NFTI-QOL was related to clinician-rated severity (ClinSev) and genetic severity (GenSev) over repeated visits. Data were evaluated for 288 NF2 patients (n = 464 visits) attending the English national NF2 clinics from 2010 to 2012. The male-to-female ratio was equal and the mean age was 42.2 (SD 17.8) years. The analysis included NFTI-QOL eight-item score, ClinSev graded as mild, moderate, or severe, and GenSev as a rank order of the number of NF2 mutations (graded as mild, moderate, severe). The mean (SD) 8.7 (5.4) score for NFTI-QOL for either a first visit or all visits 9.2 (5.4) was similar to the published norm of 9.4 (5.5), with no significant relationships with age or gender. NFTI-QOL internal reliability was good, with a Cronbach’s alpha score of 0.85 and test re-test reliability r = 0.84. NFTI related to ClinSev (r = 0.41, p < 0.001; r = 0.46 for all visits), but weakly to GenSev (r = 0.16, p < 0.05; r = 0.15 for all visits). ClinSev related to GenSev (r = 0.41, p < 0.001; r = 0.42 for all visits). NFTI-QOL showed a good reliability and ability to detect significant longitudinal changes in the QOL of individuals. The moderate relationships of NFTI-QOL with clinician- and genetic-rated severity suggest that NFTI-QOL taps into NF2 patient experiences that are not encompassed by ClinSev rating or genotype
