    Calculating risk change with management actions using Bayesian networks for the South River, Virginia, USA

    Ecological managers often implement one or more management options to manage risk without the direct integration of a quantitative risk assessment and evaluation of management alternatives. Throughout the decision making process a manager should consider multiple stressors as well as stressor interactions and the resulting effects. In my study, I used Bayesian networks in a relative risk assessment model framework (BN-RRM) to integrate two management options into existing risk assessment models for biotic endpoints and water quality endpoints in the mercury contaminated site, South River, VA. The two management options assessed were agricultural best management practices (BMPs) and bank stabilization. The primary management goal expressed by managers is no regrets. In other words, managers do not want to make the site worse in any way, such as reducing mercury levels at the detriment of habitat, loss of other species, degradation of water quality, or other environmental parameters. The Bayesian networks represent the expected effects of a management option and the potential for unintended consequences. Agricultural BMPs did not change the skew of the risk distributions, but aligns with the no regrets management focus because risk did not increase. Bank stabilization management shifted the risk distribution for smallmouth bass so that there was greater likelihood of zero risk. The risk distribution for the water quality-fishing endpoint changed because likelihood of medium and high risk increased. If bank stabilization is implemented without consideration of Belted Kingfisher nests, there was 100% likelihood of high risk to the Kingfisher. My research provides South River managers with a tool that describes how management options are expected to change risk to biotic and water quality endpoints. Adaptive management for the South River is a long-term process. The BN-RRM models can be updated with new monitoring data to inform future management decisions

    The Incidence and Clinical Relevance of Graft Hypertrophy After Matrix-Based Autologous Chondrocyte Implantation

    Background: Graft hypertrophy is the most common complication of periosteal autologous chondrocyte implantation (p-ACI). Purpose: The aim of this prospective study was to analyze the development, the incidence rate, and the persistence of graft hypertrophy after matrix-based autologous chondrocyte implantation (mb-ACI) in the knee joint within a 2-year postoperative course. Study Design: Case series; Level of evidence, 4. Methods: Between 2004 and 2007, a total of 41 patients with 44 isolated cartilage defects of the knee were treated with the mb-ACI technique. The mean age of the patients was 35.8 years (standard deviation [SD], 11.3 years), and the mean body mass index was 25.9 (SD, 4.2; range, 19-35.3). The cartilage defects were arthroscopically classified as Outerbridge grades III and IV. The mean area of the cartilage defect measured 6.14 cm2 (SD, 2.3 cm2). Postoperative clinical and magnetic resonance imaging (MRI) examinations were conducted at 3, 6, 12, and 24 months to analyze the incidence and course of the graft. Results: Graft hypertrophy developed in 25% of the patients treated with mb-ACI within a postoperative course of 1 year; 16% of the patients developed hypertrophy grade 2, and 9% developed hypertrophy grade 1. Graft hypertrophy occurred primarily in the first 12 months and regressed in most cases within 2 years. The International Knee Documentation Committee (IKDC) and visual analog scale (VAS) scores improved during the postoperative follow-up time of 2 years. There was no difference between the clinical results regarding the IKDC and VAS pain scores and the presence of graft hypertrophy. Conclusion: The mb-ACI technique does not lead to graft hypertrophy requiring treatment as opposed to classic p-ACI. The frequency of occurrence of graft hypertrophy after p-ACI and mb-ACI is comparable. Graft hypertrophy can be considered as a temporary excessive growth of regenerative cartilage tissue rather than a true graft hypertrophy. It is therefore usually not a persistent or systematic complication in the treatment of circumscribed cartilage defects with mb-ACI

    <p>Abstract</p> <p>Background</p> <p>Chitin synthase 3a (CHS3a) from <it>Botrytis cinerea </it>(Bc) catalyses the multiple transfer of <it>N</it>-acetylglucosamine (GlcNAc) residues to the growing chitin chain. Chitin, a β-1,4 linked GlcNAc homopolymer, is an essential cell wall component of filamentous fungi. Chitin synthase, processive membranous protein, has been recognized as a promising target for new antifungicides. Enzymatic characterizations of chitin synthases have been limited, mainly because purity and amounts of integral enzyme obtained after purification procedures have not been sufficient.</p> <p>Findings</p> <p>We undertook the preparation of two BcCHS3a fragment proteins, containing only the central domain and devoid of the N-terminal and transmembrane C-terminal regions. The central domain of CHS3a, named SGC (Spsa GntI Core), is conserved in all UDP-glycosyltransferases and it is believed to contain the active site of the enzyme. CHS3a-SGC protein was totally expressed as inclusion bodies in <it>Escherichia coli</it>. We performed recombinant CHS3a-SGC purification in denaturing conditions, followed by a refolding step. Although circular dichroism spectra clearly exhibited secondary structures of renatured CHS3a-SGC, no chitin synthase activity was detected. Nevertheless CHS3a-SGC proteins show specific binding for the substrate UDP-GlcNAc with a dissociation constant similar to the Michaelis constant and a major contribution of the uracil moiety for recognition was confirmed.</p> <p>Conclusions</p> <p>Milligram-scale quantities of CHS3a-SGC protein with native-like properties such as specific substrate UDP-GlcNAc binding could be easily obtained. These results are encouraging for subsequent heterologous expression of full-length CHS3a.</p

    Validation of the Vaccination Confidence Scale: A Brief Measure to Identify Parents at Risk for Refusing Adolescent Vaccines

    Objective To validate a brief measure of vaccination confidence using a large, nationally representative sample of parents. Methods We analyzed weighted data from 9018 parents who completed the 2010 National Immunization Survey–Teen, an annual, population-based telephone survey. Parents reported on the immunization history of a 13- to 17-year-old child in their households for vaccines including tetanus, diphtheria, and acellular pertussis (Tdap), meningococcal, and human papillomavirus vaccines. For each vaccine, separate logistic regression models assessed associations between parents\u27 mean scores on the 8-item Vaccination Confidence Scale and vaccine refusal, vaccine delay, and vaccination status. We repeated analyses for the scale\u27s 4-item short form. Results One quarter of parents (24%) reported refusal of any vaccine, with refusal of specific vaccines ranging from 21% for human papillomavirus to 2% for Tdap. Using the full 8-item scale, vaccination confidence was negatively associated with measures of vaccine refusal and positively associated with measures of vaccination status. For example, refusal of any vaccine was more common among parents whose scale scores were medium (odds ratio, 2.08; 95% confidence interval, 1.75–2.47) or low (odds ratio, 4.61; 95% confidence interval, 3.51–6.05) versus high. For the 4-item short form, scores were also consistently associated with vaccine refusal and vaccination status. Vaccination confidence was inconsistently associated with vaccine delay. Conclusions The Vaccination Confidence Scale shows promise as a tool for identifying parents at risk for refusing adolescent vaccines. The scale\u27s short form appears to offer comparable performance

    Spatio-temporal dynamics of wormlike micelles under shear

    Velocity profiles in a wormlike micelle solution (CTAB in D2O) are recorded using ultrasound every 2 s after a step-like shear rate into the shear-banding regime. The stress relaxation occurs over more than six hours and corresponds to the very slow nucleation and growth of the high-shear band. Moreover, oscillations of the interface position with a period of about 50 s are observed during the growth process. Strong wall slip, metastable states and transient nucleation of three-band flows are also reported and discussed in light of previous experiments and theoretical models.Comment: 4 pages, 5 figures, submitted to Phys.Rev.Let

    Virtual environment to quantify the influence of colour stimuli on the performance of tasks requiring attention

    <p>Abstract</p> <p>Background</p> <p>Recent studies indicate that the blue-yellow colour discrimination is impaired in ADHD individuals. However, the relationship between colour and performance has not been investigated. This paper describes the development and the testing of a virtual environment that is capable to quantify the influence of red-green versus blue-yellow colour stimuli on the performance of people in a fun and interactive way, being appropriate for the target audience.</p> <p>Methods</p> <p>An interactive computer game based on virtual reality was developed to evaluate the performance of the players.</p> <p>The game's storyline was based on the story of an old pirate who runs across islands and dangerous seas in search of a lost treasure. Within the game, the player must find and interpret the hints scattered in different scenarios. Two versions of this game were implemented. In the first, hints and information boards were painted using red and green colours. In the second version, these objects were painted using blue and yellow colours. For modelling, texturing, and animating virtual characters and objects the three-dimensional computer graphics tool Blender 3D was used. The textures were created with the GIMP editor to provide visual effects increasing the realism and immersion of the players. The games were tested on 20 non-ADHD volunteers who were divided into two subgroups (A1 and A2) and 20 volunteers with ADHD who were divided into subgroups B1 and B2. Subgroups A1 and B1 used the first version of the game with the hints painted in green-red colors, and subgroups A2 and B2 the second version using the same hints now painted in blue-yellow. The time spent to complete each task of the game was measured.</p> <p>Results</p> <p>Data analyzed with ANOVA two-way and posthoc TUKEY LSD showed that the use of blue/yellow instead of green/red colors decreased the game performance of all participants. However, a greater decrease in performance could be observed with ADHD participants where tasks, that require attention, were most affected.</p> <p>Conclusions</p> <p>The game proved to be a user-friendly tool capable to detect and quantify the influence of color on the performance of people executing tasks that require attention and showed to be attractive for people with ADHD.</p

    Funding Shortfall for Housing Vouchers Could Have Serious Health Consequences for Children

    In the fourth brief in its Policy Action series, Children's HealthWatch finds that unaffordable housing endangers the health and development of young children. Due to a federal funding shortfall, state and local housing agencies will be forced to reduce or eliminate rental assitance to thousands of families starting this month. Voucher cuts will push more families into the ranks of the "hidden homeless" -- families that move frequently, crowd into apartments that are too small, or live doubled up with other households when they cannot find affordable housing. Children in hidden homeless families are at increased risk for poor health, nutrition, and growth, as well as developmental delays. Timely Congressional action to protect the Housing Choice Voucher Program will ensure that families have stable, affordable housing essential to children's health
