    Cryoultramicrotomy and Immunocytochemistry in the Analysis of Muscle Fine Structure

    Cryoultramicrotomy, which avoids the use of harsh fixation procedures, deleterious dehydration and plastic embedding can be combined with immunocytochemis try to determine the ultra-structural localization of cellular proteins. Our attempts to use the cryosectioning technique in combination with immunolabelling to bridge the gap between light and electron microscopic analysis of muscle morphology have enabled us to obtain new information on fibre typing at the ultrastructural level. Furthermore, we have obtained a marked improvement in the resolution of myofibrillar structures by using semithin cryosections for fluorescence microscopy. Data are also presented on correlated light and electron microscope immunocytochemistry of myocardial intermediate filaments confirming the presence of longitudinally oriented intermediate filaments of desmin in the region of the intercalated discs of mammalian cardiac myocytes, whereas elsewhere in the myocyte the bulk of intermediate filaments of desmin is concentrated in the intermyofibrillar space at the level of the Z disc

    Financial costs of the Zanzibar elimination of schistosomiasis transmission project

    We estimated the financial costs of different interventions against urogenital schistosomiasis, implemented by the Zanzibar Elimination of Schistosomiasis Transmission (ZEST) project, on Pemba and Unguja islands, Tanzania. We used available data on project activities, resources used, and costs reported in the accounting information systems of ZEST partners. The costs were estimated for all the activities related to snail control, behavior change interventions, the impact assessment surveys, and management of the whole program. Costs are presented in USforthefulldurationoftheZESTprojectfrom2011/2012to2017.Thetotalfinancialcostsofimplementingsnailcontrolactivitiesover5years,excludingthecostsfordonatedBayluscide,wereUS for the full duration of the ZEST project from 2011/2012 to 2017. The total financial costs of implementing snail control activities over 5 years, excluding the costs for donated Bayluscide, were US55,796 on Pemba and US73,581onUnguja,mainlydrivenbypersonnelcosts.ThetotalfinancialcostsofimplementingbehaviorchangeactivitieswereUS73,581 on Unguja, mainly driven by personnel costs. The total financial costs of implementing behavior change activities were US109,165 on Pemba and US155,828onUnguja,withcostsforpersonnelaccountingfor47155,828 on Unguja, with costs for personnel accounting for 47% on Pemba and 69% on Unguja. Costs of implementing biannual mass drug administration refer to the estimated 2.4 million treatments provided on Pemba over 4 years (2013-2016), and do not include the costs of donated praziquantel. The total cost per provided treatment was, on average, US0.21. This study showed the value of exploiting administrative data to estimate costs of major global health interventions. It also provides an evidence base for financial costs and main cost drivers of implementing multiple combinations of intervention sets that inform decisions regarding the feasibility and affordability of implementing schistosomiasis control and elimination strategies

    The s Process: Nuclear Physics, Stellar Models, Observations

    Nucleosynthesis in the s process takes place in the He burning layers of low mass AGB stars and during the He and C burning phases of massive stars. The s process contributes about half of the element abundances between Cu and Bi in solar system material. Depending on stellar mass and metallicity the resulting s-abundance patterns exhibit characteristic features, which provide comprehensive information for our understanding of the stellar life cycle and for the chemical evolution of galaxies. The rapidly growing body of detailed abundance observations, in particular for AGB and post-AGB stars, for objects in binary systems, and for the very faint metal-poor population represents exciting challenges and constraints for stellar model calculations. Based on updated and improved nuclear physics data for the s-process reaction network, current models are aiming at ab initio solution for the stellar physics related to convection and mixing processes. Progress in the intimately related areas of observations, nuclear and atomic physics, and stellar modeling is reviewed and the corresponding interplay is illustrated by the general abundance patterns of the elements beyond iron and by the effect of sensitive branching points along the s-process path.     New insights into the protein aggregation pathology in myotilinopathy by combined proteomic and immunolocalization analyses

    Introduction: Myofibrillar myopathies are characterized by progressive muscle weakness and impressive abnormal protein aggregation in muscle fibers. In about 10 % of patients, the disease is caused by mutations in the MYOT gene encoding myotilin. The aim of our study was to decipher the composition of protein deposits in myotilinopathy to get new information about aggregate pathology. Results: Skeletal muscle samples from 15 myotilinopathy patients were included in the study. Aggregate and control samples were collected from muscle sections by laser microdissection and subsequently analyzed by a highly sensitive proteomic approach that enables a relative protein quantification. In total 1002 different proteins were detected. Seventy-six proteins showed a significant over-representation in aggregate samples including 66 newly identified aggregate proteins. Z-disc-associated proteins were the most abundant aggregate components, followed by sarcolemmal and extracellular matrix proteins, proteins involved in protein quality control and degradation, and proteins with a function in actin dynamics or cytoskeletal transport. Forty over-represented proteins were evaluated by immunolocalization studies. These analyses validated our mass spectrometric data and revealed different regions of protein accumulation in abnormal muscle fibers. Comparison of data from our proteomic analysis in myotilinopathy with findings in other myofibrillar myopathy subtypes indicates a characteristic basic pattern of aggregate composition and resulted in identification of a highly sensitive and specific diagnostic marker for myotilinopathy. Conclusions: Our findings i) indicate that main protein components of aggregates belong to a network of interacting proteins, ii) provide new insights into the complex regulation of protein degradation in myotilinopathy that may be relevant for new treatment strategies, iii) imply a combination of a toxic gain-of-function leading to myotilin-positive protein aggregates and a loss-of-function caused by a shift in subcellular distribution with a deficiency of myotilin at Z-discs that impairs the integrity of myofibrils, and iv) demonstrate that proteomic analysis can be helpful in differential diagnosis of protein aggregate myopathies

    This article explores how macroeconomic and environmental policies instituted since the 1990s have altered meanings, imaginaries, and the human relationship to nature in Colombia. The Colombian nation-state is pluri-ethnic, multilingual, and megabiodiverse. In this context, indigenous peoples, Afro-Colombians, and some peasant communities survive hybridization of their cultures. They have developed their own ways of seeing, understanding, and empowering the world over centuries of European rule. However, threats to relatively discrete cultural meanings have increased since major changes in the 1990s, when Colombia experienced the emergence of new and modern interpretations of nature, such as “biodiversity,” and a deepening of globalized neoliberal economic and political management. These policies involve a modern logic of being in the world, the establishment of particular regulatory functions for economies, societies, and the environment, and their spread has been facilitated by webs of political and economic power. We trace their local effects with reference to three indigenous groups

    Xirp Proteins Mark Injured Skeletal Muscle in Zebrafish

    Myocellular regeneration in vertebrates involves the proliferation of activated progenitor or dedifferentiated myogenic cells that have the potential to replenish lost tissue. In comparison little is known about cellular repair mechanisms within myocellular tissue in response to small injuries caused by biomechanical or cellular stress. Using a microarray analysis for genes upregulated upon myocellular injury, we identified zebrafish Xin-actin-binding repeat-containing protein1 (Xirp1) as a marker for wounded skeletal muscle cells. By combining laser-induced micro-injury with proliferation analyses, we found that Xirp1 and Xirp2a localize to nascent myofibrils within wounded skeletal muscle cells and that the repair of injuries does not involve cell proliferation or Pax7+ cells. Through the use of Xirp1 and Xirp2a as markers, myocellular injury can now be detected, even though functional studies indicate that these proteins are not essential in this process. Previous work in chicken has implicated Xirps in cardiac looping morphogenesis. However, we found that zebrafish cardiac morphogenesis is normal in the absence of Xirp expression, and animals deficient for cardiac Xirp expression are adult viable. Although the functional involvement of Xirps in developmental and repair processes currently remains enigmatic, our findings demonstrate that skeletal muscle harbours a rapid, cell-proliferation-independent response to injury which has now become accessible to detailed molecular and cellular characterizations
