    The Planetary Nebula System and Dynamics in the Outer Halo of NGC 5128

    The halos of elliptical galaxies are faint and difficult to explore, but they contain vital clues to both structure and formation. We present the results of an imaging and spectroscopic survey for planetary nebulae (PNe) in the nearby elliptical NGC 5128. We extend the work of Hui et al.(1995) well into the halo of the galaxy--out to distances of 100 and 50 kpc along the major and minor axes. We now know of 1141 PNe in NGC 5128, 780 of which are confirmed. Of these 780 PNe, 349 are new from this survey, and 148 are at radii beyond 20 kpc. PNe exist at distances up to 80 kpc (~15 r_e), showing that the stellar halo extends to the limit of our data. This study represents by far the largest kinematic study of an elliptical galaxy to date, both in the number of velocity tracers and in radial extent. We confirm the large rotation of the PNe along the major axis, and show that it extends in a disk-like feature into the halo. The rotation curve of the stars flattens at ~100 km/s with V/sigma between 1 and 1.5, and with the velocity dispersion of the PNe falling gradually at larger radii. The two-dimensional velocity field exhibits a zero-velocity contour with a pronounced twist, showing that the galaxy potential is likely triaxial in shape, tending toward prolate. The total dynamical mass of the galaxy within 80 kpc is ~5 x 10^{11} M_sun, with M/L_B ~ 13.     The Surface Brightness Fluctuations and Globular Cluster Populations of M87 and its Companions

    Using the surface brightness fluctuations in HST WFPC-2 images, we determine that M87, NGC 4486B, and NGC 4478 are all at a distance of ~16 Mpc, while NGC 4476 lies in the background at ~21 Mpc. We also examine the globular clusters of M87 using archived HST fields. We detect the bimodal color distribution, and find that the amplitude of the red peak relative to the blue peak is greatest near the center. This feature is in good agreement with the merger model of elliptical galaxy formation, where some of the clusters originated in progenitor galaxies while other formed during mergers.Comment: 5 pages, 2 figure

    The Globular Cluster System of NGC 5128 II. Ages, Metallicities, Kinematics, and Formation

    We present a study of the nearby post-merger giant elliptical galaxy, NGC 5128 (Centaurus A), in which we use the properties of its globular cluster (GC) and planetary nebula (PN) systems to constrain its evolution. Using photometric and spectroscopic data for 215 GCs presented in Paper I, we study trends in age, metallicity, and kinematics for the GC system. We confirm that the GC metallicity distribution is bimodal, and show that these two sub-populations have different properties. Using spectral line index measurements of the brightest clusters, the metal-poor GCs have old ages like the Milky Way globular clusters, while the metal-rich GCs have H-beta line-strengths that could be interpreted as a mean age of ~5 (+3/-2) Gyr. Both populations appear to have [Mg/Fe] ratios consistent with that of the Galactic GC system, although this quantity is not very well-constrained. The kinematics of the metal-rich GCs are similar to those of the planetary nebulae, exhibiting significant rotation about a misaligned axis, while the metal-poor GCs have a higher velocity dispersion and show a weaker kinematic correlation with the field stars. The total gravitating mass of NGC 5128 derived from the GCs is in excellent agreement with the value derived from stellar (PN) kinematics. We suggest that these and other data support a picture in which the main body of NGC 5128 was formed 3-8 Gyr ago by the dissipational merger of two unequal-mass disk galaxies supplemented by the continual accretion of both gas-rich and gas-poor satellites.Comment: 15 pages, 21 figures (figures 14-20 best viewed in color), accepted for publication in the Astrophysical Journa

    Astrocyte-Specific Expression Patterns Associated with the PDGF-Induced Glioma Microenvironment

    The tumor microenvironment contains normal, non-neoplastic cells that may contribute to tumor growth and maintenance. Within PDGF-driven murine gliomas, tumor-associated astrocytes (TAAs) are a large component of the tumor microenvironment. The function of non-neoplastic astrocytes in the glioma microenvironment has not been fully elucidated; moreover, the differences between these astrocytes and normal astrocytes are unknown. We therefore sought to identify genes and pathways that are increased in TAAs relative to normal astrocytes and also to determine whether expression of these genes correlates with glioma behavior.We compared the gene expression profiles of TAAs to normal astrocytes and found the Antigen Presentation Pathway to be significantly increased in TAAs. We then identified a gene signature for glioblastoma (GBM) TAAs and validated the expression of some of those genes within the tumor. We also show that TAAs are derived from the non-tumor, stromal environment, in contrast to the Olig2+ tumor cells that constitute the neoplastic elements in our model. Finally, we validate this GBM TAA signature in patients and show that a TAA-derived gene signature predicts survival specifically in the human proneural subtype of glioma.Our data identifies unique gene expression patterns between populations of TAAs and suggests potential roles for stromal astrocytes within the glioma microenvironment. We show that certain stromal astrocytes in the tumor microenvironment express a GBM-specific gene signature and that the majority of these stromal astrocyte genes can predict survival in the human disease

    SummaryMaintenance of genomic integrity is essential for adult tissue homeostasis and defects in the DNA-damage response (DDR) machinery are linked to numerous pathologies including cancer. Here, we present evidence that the DDR exerts tumor suppressor activity in gliomas. We show that genes encoding components of the DDR pathway are frequently altered in human gliomas and that loss of elements of the ATM/Chk2/p53 cascade accelerates tumor formation in a glioma mouse model. We demonstrate that Chk2 is required for glioma response to ionizing radiation in vivo and is necessary for DNA-damage checkpoints in the neuronal stem cell compartment. Finally, we observed that the DDR is constitutively activated in a subset of human GBMs, and such activation correlates with regions of hypoxia

    Wind-driven upwelling around grounded tabular icebergs

    Funding was provided by NSF Polar Programs - Grant Number: ARC-1304137.Temperature and salinity data collected around grounded tabular icebergs in Baffin Bay in 2011, 2012 and 2013 indicate wind-induced upwelling at certain locations around the icebergs. These data suggest that along one side of the iceberg, wind forcing leads to Ekman transport away from the iceberg, which causes upwelling of the cool saline water from below. The upwelling water mixes with the water in the thermocline, causing the mixed layer to become cooler and more saline. Along the opposite side of the iceberg, the surface Ekman transport moves towards the iceberg, which causes a sharpening of the thermocline as warm fresh water is trapped near the surface. This results in higher mixed layer temperatures and lower mixed layer salinities on this side of the iceberg. Based on these in situ measurements, we hypothesize that the asymmetries in water properties around the iceberg, caused by the opposing effects of upwelling and sharpening of the thermocline, lead to differential deterioration around the iceberg. Analysis of satellite imagery around iceberg PII-B-1 over a six month monitoring period reveals differential decay around the iceberg, in agreement with this mechanism.Publisher PDFPeer reviewe

    Perivascular Nitric Oxide Activates Notch Signaling and Promotes Stem-like Character in PDGF-Induced Glioma Cells

    SummaryeNOS expression is elevated in human glioblastomas and correlated with increased tumor growth and aggressive character. We investigated the potential role of nitric oxide (NO) activity in the perivascular niche (PVN) using a genetic engineered mouse model of PDGF-induced gliomas. eNOS expression is highly elevated in tumor vascular endothelium adjacent to perivascular glioma cells expressing Nestin, Notch, and the NO receptor, sGC. In addition, the NO/cGMP/PKG pathway drives Notch signaling in PDGF-induced gliomas in vitro, and induces the side population phenotype in primary glioma cell cultures. NO also increases neurosphere forming capacity of PDGF-driven glioma primary cultures, and enhances their tumorigenic capacity in vivo. Loss of NO activity in these tumors suppresses Notch signaling in vivo and prolongs survival of mice. This mechanism is conserved in human PDGFR amplified gliomas. The NO/cGMP/PKG pathway's promotion of stem cell-like character in the tumor PVN may identify therapeutic targets for this subset of gliomas

    Reconstruction of the joint state of a two-mode Bose-Einstein condensate

    We propose a scheme to reconstruct the state of a two-mode Bose-Einstein condensate, with a given total number of atoms, using an atom interferometer that requires beam splitter, phase shift and non-ideal atom counting operations. The density matrix in the number-state basis can be computed directly from the probabilities of different counts for various phase shifts between the original modes, unless the beamsplitter is exactly balanced. Simulated noisy data from a two-mode coherent state is produced and the state is reconstructed, for 49 atoms.
