    Antiretroviral Therapy Reduces Markers of Endothelial and Coagulation Activation in Patients Infected with Human Immunodeficiency Virus Type 1

    We investigated the effect of antiretroviral therapy on vascular activation in 41 human immunodeficiency (HIV)—infected patients receiving a regimen that included either at least 1 protease inhibitor (PI; n = 21) or a nonnucleoside reverse-transcriptase inhibitor (NNRTI; n = 20). Acontrol group of 21 healthy subjectswas included for comparison. Levels of endothelialmarkers (soluble vascular cell adhesion molecule [sVCAM]-1, soluble intercellular adhesionmolecule-1, and vonWillebrand factor) were higher in HIV-infected persons before treatment than in control subjects anddecreasedsignificantlyafter 5-13 months of treatment. Levels of sVCAM-1 and vonWillebrand factor correlated significantly with initial virus load. D-dimer concentrations also decreased significantly after initiation of treatment. PI- and NNRTI-containing regimens had similar effects. Therapy did not reduce levels of the soluble platelet (sP) activation markers sP-selectin and CD40 ligand. The inhibition of markers of vascular activation may counterbalance sequelae of therapy-induced dyslipidemia and potentially prevent development of atherosclerosis in HIVinfected patient

    Homology and Specificity of Natural Sound-Encoding in Human and Monkey Auditory Cortex

    Understanding homologies and differences in auditory cortical processing in human and nonhuman primates is an essential step in elucidating the neurobiology of speech and language. Using fMRI responses to natural sounds, we investigated the representation of multiple acoustic features in auditory cortex of awake macaques and humans. Comparative analyses revealed homologous large-scale topographies not only for frequency but also for temporal and spectral modulations. In both species, posterior regions preferably encoded relatively fast temporal and coarse spectral information, whereas anterior regions encoded slow temporal and fine spectral modulations. Conversely, we observed a striking interspecies difference in cortical sensitivity to temporal modulations: While decoding from macaque auditory cortex was most accurate at fast rates (> 30 Hz), humans had highest sensitivity to ~3 Hz, a relevant rate for speech analysis. These findings suggest that characteristic tuning of human auditory cortex to slow temporal modulations is unique and may have emerged as a critical step in the evolution of speech and language

    Dual Agents : Fungal Macrocidins and Synthetic Analogues with Herbicidal and Antibiofilm Activities

    Eight analogues of the bioherbicides macrocidin A (1) and Z (2) with structural variance in the size of the macrocycle, its para- or meta-cyclophane character, and its functional groups were synthesized on two modular routes and tested for herbicidal, antibiotic, and antibiofilm activities. Apart from the lead compounds 1 and 2, the structurally simplified dihydromacrocidin Z (3) and normacrocidin Z (4) showed high herbicidal activity in either thistles, dandelions or in both. The derivatives 2, 3, and dibromide 9 also inhibited the growth of Staphylococcus aureus biofilms by ca 70% when applied at subtoxic concentrations as low as ca 20 ”M, which are unlikely to induce bacterial resistance. They also led to the dispersion of preformed biofilms of S. aureus, exceeding a similar effect by microporenic acid A, a known biofilm inhibitor. Compounds 3 and 9 showed no noticeable cytotoxicity against human cancer and endothelial cells at concentrations below 50 ”M, making them conceivable candidates for application as anti-biofilm agents in a medicinal context

    Challenges and Opportunities for Improving Eco-Efficiency of Tropical Forage-Based Systems to Mitigate Greenhouse Gas Emissions

    Forage-based livestock production plays a key role in national and regional economies, for food security and poverty alleviation. Livestock production is also considered as a major contributor to agricultural GHG emissions, however. While demand for livestock products is predicted to continue to increase, there is political and societal pressure both to reduce environmental impacts and to convert some of the pasture area to alternative uses such as crop production and environmental conservation. Thus it is essential to develop approaches for sustainable intensification of livestock systems to mitigate GHG emissions, addressing biophysical, socioeconomic and policy challenges. This paper highlights the potential of improved tropical forages in crop-livestock systems, and linked with policy incentives, to enhance livestock production while reducing its environmental footprint. We give examples for sustainable intensification to mitigate GHG emissions based on improved forages in Brazil and Colombia and suggest future perspectives

    Impacts of Scaling up Agroecology on the Sustainability of European Agriculture in 2050

    The European Commission recently embraced the concept of agroecology as a pathway to reduce negative impacts from agri-food systems on the environment. So far, it remains unclear whether agroecology can deliver on these high hopes if implemented on a large scale. We here assess socio-economic and environmental implications of multiple agroecological futures in the European Union in 2050, based on a novel diagnostic scenario approach, i.e. the biomass balancing model BioBaM-GHG 2.0. We find that agroecological measures from the plot to the food systems level can indeed reduce environmental pressures while maintaining domestic food availability within the EU. Such measures are, for example, more hedgerows on croplands or reduced biomass harvest on high natural value – HNV grasslands. However, a key prerequisite is an overall reduction of the food system's size (based on the reduction of animal production, food wastes, and export production) and an optimised crop-livestock integration. Only then does the transformation towards an agroecological agri-food system in the EU not risk overstretching domestic land availability or produce insufficient agricultural commodities. Mitigating the accompanied trade-off of reduced farm income is a central mandate for policy development aimed at re-designing agriculture in Europe to align with the Green Deal goals

    Peri-operative red blood cell transfusion in neonates and infants: NEonate and Children audiT of Anaesthesia pRactice IN Europe: A prospective European multicentre observational study

    BACKGROUND: Little is known about current clinical practice concerning peri-operative red blood cell transfusion in neonates and small infants. Guidelines suggest transfusions based on haemoglobin thresholds ranging from 8.5 to 12 g dl-1, distinguishing between children from birth to day 7 (week 1), from day 8 to day 14 (week 2) or from day 15 (≄week 3) onwards. OBJECTIVE: To observe peri-operative red blood cell transfusion practice according to guidelines in relation to patient outcome. DESIGN: A multicentre observational study. SETTING: The NEonate-Children sTudy of Anaesthesia pRactice IN Europe (NECTARINE) trial recruited patients up to 60 weeks' postmenstrual age undergoing anaesthesia for surgical or diagnostic procedures from 165 centres in 31 European countries between March 2016 and January 2017. PATIENTS: The data included 5609 patients undergoing 6542 procedures. Inclusion criteria was a peri-operative red blood cell transfusion. MAIN OUTCOME MEASURES: The primary endpoint was the haemoglobin level triggering a transfusion for neonates in week 1, week 2 and week 3. Secondary endpoints were transfusion volumes, 'delta haemoglobin' (preprocedure - transfusion-triggering) and 30-day and 90-day morbidity and mortality. RESULTS: Peri-operative red blood cell transfusions were recorded during 447 procedures (6.9%). The median haemoglobin levels triggering a transfusion were 9.6 [IQR 8.7 to 10.9] g dl-1 for neonates in week 1, 9.6 [7.7 to 10.4] g dl-1 in week 2 and 8.0 [7.3 to 9.0] g dl-1 in week 3. The median transfusion volume was 17.1 [11.1 to 26.4] ml kg-1 with a median delta haemoglobin of 1.8 [0.0 to 3.6] g dl-1. Thirty-day morbidity was 47.8% with an overall mortality of 11.3%. CONCLUSIONS: Results indicate lower transfusion-triggering haemoglobin thresholds in clinical practice than suggested by current guidelines. The high morbidity and mortality of this NECTARINE sub-cohort calls for investigative action and evidence-based guidelines addressing peri-operative red blood cell transfusions strategies.
