    PRRSV RNA detection in different matrices under typical storage conditions in the UK

    In the UK, approximately 40 per cent of the pig breeding herds are outdoors. To monitor their porcine reproductive and respiratory syndrome virus (PRRSV) status, blood is collected commonly from piglets around weaning. Sample collection in British outdoor pigs often occurs during the early morning hours when the piglets tend to accumulate inside sheltered areas. For practical reasons, dry cotton swabs are occasionally used for blood collection and stored at room temperature until arrival in the laboratory. Detection of PRRSV RNA is a function of viral concentration, sample type and storage condition. To evaluate a possible impact of the sampling protocol on PRRSV1 detection, experimentally spiked blood samples using three dilutions of a representative PRRSV1 strain were prepared. In addition, blood samples from pigs naturally infected with PRRSV were obtained from a PRRSV-positive British herd. Spiked blood and blood from infected pigs were used to obtain sera, dry or wet (immersed in saline) polyester or cotton swabs and FTA cards. The different samples were stored for 24 hours, 48 hours or 7 days at 4°C or 20°C and tested by a real-time reverse transcriptase PRRSV PCR assay. Under the study conditions, the best matrix was serum (96.7 per cent), followed by wet swabs (78 per cent), dry swabs (61.3 per cent) and FTA cards (51 per cent). Polyester swabs (76 per cent) showed a better performance than cotton swabs (63.3 per cent). The reduction in sensitivity obtained for swabs and FTA cards was particularly high at low viral concentrations. The results indicate that wet polyester swabs should be used whenever possible

    The Effect of Acoustic Forcing on Instabilities and Breakdown in Swept-Wing Flow over a Backward-Facing Step

    Instability interaction and breakdown were experimentally investigated in the flow over a swept backward-facing step. Acoustic forcing was used to excite the Tollmien-Schlichting (TS) instability and to acquire phase-locked results. The phase-averaged results illustrate the complex nature of the interaction between the TS and stationary cross flow instabilities. The weak stationary cross flow disturbance causes a distortion of the TS wavefront. The breakdown process is characterized by large positive and negative spikes in velocity. The positive spikes occur near the same time and location as the positive part of the TS wave. Higher-order spectral analysis was used to further investigate the nonlinear interactions between the TS instability and the traveling cross flow disturbances. The results reveal that a likely cause for the generation of the spikes corresponds to nonlinear interactions between the TS, traveling cross flow, and stationary cross flow disturbances. The spikes begin at low amplitudes of the unsteady and steady disturbances (2-4% U (sub e) (i.e. boundary layer edge velocity)) but can achieve very large amplitudes (20-30 percent U (sub e) (i.e. boundary layer edge velocity)) that initiate an early, though highly intermittent, breakdown to turbulence

    Functional analysis of two-amino acid substitutions in gp91 phox in a patient with X-linked flavocytochrome b558-positive chronic granulomatous disease by means of transgenic PLB-985 cells

    Chronic granulomatous disease (CGD) is a rare inherited disorder in which phagocytes lack NADPH oxidase activity. The most common form is caused by mutations in the CYBB gene encoding gp91phox protein, the heavy chain of cytochrome b558, which is the redox element of NADPH oxidase. In some rare cases, the mutated gp91phox is normally expressed but no NADPH oxidase can be detected. This type of CGD is called X91+ CGD. We have previously reported an X+ CGD case with a double-missense mutation in gp91phox. Transgenic PLB-985 cells have now been made to study the impact of each single mutation on oxidase activity and assembly to rule out a possible new polymorphism in the CYBB gene. The His303Asn/Pro304Arg gp91phox transgenic PLB-985 cells exactly mimic the phenotype of the neutrophils of the X+ CGD patient. The His303Asn mutation is sufficient to inhibit oxidase activity in intact cells and in a broken cell system, whereas in the Pro304Arg mutant, residual activity suggests that the Pro304Arg substitution is less devastating to oxidase activity than the His303Asn mutation. The study of NADPH oxidase assembly following the in vitro and in vivo translocation of cytosolic factors p47phox and p67phox has demonstrated that, in the double mutant and in the His303Asn mutant, NADPH oxidase assembly is abolished, although the translocation is only attenuated in Pro304Arg mutant cells. Thus, even though the His303Asn mutation has a more severe inhibitory effect on NADPH oxidase activity and assembly than the Pro304Arg mutation, neither mutation can be considered as a polymorphism

    Economic values for ecosystem services:A global synthesis and way forward

    This paper presents a global synthesis of economic values for ecosystem services provided by 15 terrestrial and marine biomes. Information from over 1,300 studies, yielding over 9,400 value estimates in monetary units, has been collected and organised in the Ecosystem Services Valuation Database (ESVD). This is a substantial expansion of data since the de Groot et al. (2012) description of the ESVD and provides an important juncture to explore developments in the use of valuation methods and the contexts in which valuations are conducted. In this paper we provide summary values for 23 ecosystem services from 15 biomes to represent the magnitude, variation and gaps in economic values. To enable the comparison and synthesis of values, estimates in the ESVD are standardised to a common set of units (Int$/ha/year at 2020 price levels). This data provides a basis for value transfers to inform decision-making in current policy contexts but requires due consideration and adjustment for context specific determinants of value. Although the coverage of the ESVD is global, the geographic distribution of data is not even. There is a particularly high representation of European ecosystems and relatively little information for Russia, Central Asia and North Africa. Therefore, the data are not globally representative of biophysical and socio-economic contexts. The distribution of data across ecosystem services is also far from even, with some services very well represented (e.g. recreation, wild fish and wild animals, ecosystem and species appreciation, air filtration and global climate regulation) and others with almost no value estimates (e.g. disease control, water baseflow maintenance, rainfall pattern regulation). In the past decade, there has been a notable increase in demand for information on the economic value of ecosystem services from both public and private institutions to improve the conservation and management of natural capital. The literature is developing to meet this demand but there is a need for targeted and refined valuation research to ensure sufficient certainty, comparability, and representativeness of the data, and to enable transferability and fill knowledge gaps. This paper concludes by identifying avenues for future development to further increase the amount, quality, representativeness and application of data on economic values for ecosystem services.</p

    Slow Molecules Produced by Photodissociation

    A simple method to control molecular translation with a chemical reaction is demonstrated. Slow NO molecules have been produced by partially canceling the molecular beam velocity of NO2_2 with the recoil velocity of the NO photofragment. The NO2_2 molecules were photodissociated using a UV laser pulse polarized parallel to the molecular beam. The spatial profiles of NO molecules showed two peaks corresponding to decelerated and accelerated molecules, in agreement with theoretical prediction. A significant portion of the decelerated NO molecules stayed around the initial dissociation positions even several hundred nanoseconds after their production.Comment: 17 pages, 4 figure
