Development and evaluation of a Gal4-mediated LUC/GFP/GUS enhancer trap system in Arabidopsis
BACKGROUND: Gal4 enhancer trap systems driving expression of LacZ and GFP reporters have been characterized and widely used in Drosophila. However, a Gal4 enhancer trap system in Arabidopsis has not been described in the primary literature. In Drosophila, the reporters possess a Gal4 upstream activation sequence (UAS) as five repeats (5XUAS) and lines that express Gal4 from tissue specific enhancers have also been used for the ectopic expression of any transgene (driven by a 5XUAS). While Gal4 transactivation has been demonstrated in Arabidopsis, wide use of a trap has not emerged in part because of the lack of detailed analysis, which is the purpose of the present study. RESULTS: A key feature of this study is the use of luciferase (LUC) as the primary reporter and rsGFP-GUS as secondary reporters. Reporters driven by a 5XUAS are better suited in Arabidopsis than those containing a 1X or 2X UAS. A 5XUAS-LUC reporter is expressed at high levels in Arabidopsis lines transformed with Gal4 driven by the full, enhanced 35S promoter. In contrast, a minimum 35S (containing the TATA region) upstream of Gal4 acts as an enhancer trap system. Luciferase expression in trap lines of the T1, T2, and T3 generations are generally stable but by the T4 generation approximately 25% of the lines are significantly silenced. This silencing is reversed by growing plants on media containing 5-aza-2'-deoxycytidine. Quantitative multiplex RT-PCR on the Gal4 and LUC mRNA indicate that this silencing can occur at the level of Gal4 or LUC transcription. Production of a 10,000 event library and observations on screening, along with the potential for a Gal4 driver system in other plant species are discussed. CONCLUSION: The Gal4 trap system described here uses the 5XUAS-LUC and 5XUAS rsGFP-GUS as reporters and allows for in planta quantitative screening, including the rapid monitoring for silencing. We conclude that in about 75% of the cases silencing is at the level of transcription of the Gal4 transgene and is at an acceptable frequency to make the Gal4 trap system in Arabidopsis of value. This system will be useful for the isolation and comprehensive characterization of specific reporter and driver lines
Design and feasibility testing of a novel group intervention for young women who binge drink in groups
BackgroundYoung women frequently drink alcohol in groups and binge drinking within these natural drinking groups is common. This study describes the design of a theoretically and empirically based group intervention to reduce binge drinking among young women. It also evaluates their engagement with the intervention and the acceptability of the study methods.MethodsFriendship groups of women aged 18–35 years, who had two or more episodes of binge drinking (>6 UK units on one occasion; 48g of alcohol) in the previous 30 days, were recruited from the community. A face-to-face group intervention, based on the Health Action Process Approach, was delivered over three sessions. Components of the intervention were woven around fun activities, such as making alcohol free cocktails. Women were followed up four months after the intervention was delivered. Results The target of 24 groups (comprising 97 women) was recruited. The common pattern of drinking was infrequent, heavy drinking (mean consumption on the heaviest drinking day was UK 18.1 units). Process evaluation revealed that the intervention was delivered with high fidelity and acceptability of the study methods was high. The women engaged positively with intervention components and made group decisions about cutting down. Twenty two groups set goals to reduce their drinking, and these were translated into action plans. Retention of individuals at follow up was 87%.ConclusionsThis study successfully recruited groups of young women whose patterns of drinking place them at high risk of acute harm. This novel approach to delivering an alcohol intervention has potential to reduce binge drinking among young women. The high levels of engagement with key steps in the behavior change process suggests that the group intervention should be tested in a full randomised controlled trial

Review of Flywheel based Internal Combustion Engine Hybrid Vehicles
Hybrid vehicles of different configurations and utilizing different energy storage systems have existed in development for many decades and more recently in limited production. They can be grouped as parallel, series or complex hybrids. Another classification is micro, mild and full hybrids which makes the distinction on the basis of functionality. The common energy storage systems in hybrid vehicles are batteries, supercapacitors and high speed flywheels. This paper aims to review a specific type of hybrid vehicle which involves the internal combustion engine (ICE) as the prime mover and the high speed flywheel as an energy storage device. Such hybrids are now attracting considerable interest given their potential for low cost. It is hence timely to produce a review of research and development in this subject. The flywheel is coupled to the drive line with a continuous variable transmission (CVT). The CVT can be of various types such as electrical, hydraulic or mechanical but usually in this case it is a non-electrical one. Different configurations are possible and the paper provides a timeline of the development of such powertrains with various examples. These types of hybrid vehicles have existed as prototypes for many decades and the authors believe that their development has reached levels where they can be considered serious contenders for production vehicles
Dark Energy and Gravity
I review the problem of dark energy focusing on the cosmological constant as
the candidate and discuss its implications for the nature of gravity. Part 1
briefly overviews the currently popular `concordance cosmology' and summarises
the evidence for dark energy. It also provides the observational and
theoretical arguments in favour of the cosmological constant as the candidate
and emphasises why no other approach really solves the conceptual problems
usually attributed to the cosmological constant. Part 2 describes some of the
approaches to understand the nature of the cosmological constant and attempts
to extract the key ingredients which must be present in any viable solution. I
argue that (i)the cosmological constant problem cannot be satisfactorily solved
until gravitational action is made invariant under the shift of the matter
lagrangian by a constant and (ii) this cannot happen if the metric is the
dynamical variable. Hence the cosmological constant problem essentially has to
do with our (mis)understanding of the nature of gravity. Part 3 discusses an
alternative perspective on gravity in which the action is explicitly invariant
under the above transformation. Extremizing this action leads to an equation
determining the background geometry which gives Einstein's theory at the lowest

Differential Effects of Aging on Fore– and Hindpaw Maps of Rat Somatosensory Cortex
Getting older is associated with a decline of cognitive and sensorimotor abilities, but it remains elusive whether age-related changes are due to accumulating degenerational processes, rendering them largely irreversible, or whether they reflect plastic, adaptational and presumably compensatory changes. Using aged rats as a model we studied how aging affects neural processing in somatosensory cortex. By multi-unit recordings in the fore- and hindpaw cortical maps we compared the effects of aging on receptive field size and response latencies. While in aged animals response latencies of neurons of both cortical representations were lengthened by approximately the same amount, only RFs of hindpaw neurons showed severe expansion with only little changes of forepaw RFs. To obtain insight into parallel changes of walking behavior, we recorded footprints in young and old animals which revealed a general age-related impairment of walking. In addition we found evidence for a limb-specific deterioration of the hindlimbs that was not observed in the forelimbs. Our results show that age-related changes of somatosensory cortical neurons display a complex pattern of regional specificity and parameter-dependence indicating that aging acts rather selectively on cortical processing of sensory information. The fact that RFs of the fore- and hindpaws do not co-vary in aged animals argues against degenerational processes on a global scale. We therefore conclude that age-related alterations are composed of plastic-adaptive alterations in response to modified use and degenerational changes developing with age. As a consequence, age-related changes need not be irreversible but can be subject to amelioration through training and stimulation
