    Introduction: Muslim Pilgrimage through the lens of women's new mobilities

    This book investigates female Muslims pilgrimage practices and how these relate to women’s mobility, social relations, identities, and the power structures that shape women’s lives. Bringing together scholars from different disciplines and regional expertise, it offers in-depth investigation of the gendered dimensions of Muslim pilgrimage and the life-worlds of female pilgrims.     Allogeneic Non-Adherent Bone Marrow Cells Facilitate Hematopoietic Recovery but Do Not Lead to Allogeneic Engraftment

    Background Non adherent bone marrow derived cells (NA-BMCs) have recently been described to give rise to multiple mesenchymal phenotypes and have an impact in tissue regeneration. Therefore, the effects of murine bone marrow derived NA-BMCs were investigated with regard to engraftment capacities in allogeneic and syngeneic stem cell transplantation using transgenic, human CD4+, murine CD4?/?, HLA-DR3+ mice. Methodology/Principal Findings Bone marrow cells were harvested from C57Bl/6 and Balb/c wild-type mice, expanded to NA-BMCs for 4 days and characterized by flow cytometry before transplantation in lethally irradiated recipient mice. Chimerism was detected using flow cytometry for MHC-I (H-2D[b], H-2K[d]), mu/huCD4, and huHLA-DR3). Culturing of bone marrow cells in a dexamethasone containing DMEM medium induced expansion of non adherent cells expressing CD11b, CD45, and CD90. Analysis of the CD45+ showed depletion of CD4+, CD8+, CD19+, and CD117+ cells. Expanded syngeneic and allogeneic NA-BMCs were transplanted into triple transgenic mice. Syngeneic NA-BMCs protected 83% of mice from death (n = 8, CD4+ donor chimerism of 5.8±2.4% [day 40], P<.001). Allogeneic NA-BMCs preserved 62.5% (n = 8) of mice from death without detectable hematopoietic donor chimerism. Transplantation of syngeneic bone marrow cells preserved 100%, transplantation of allogeneic bone marrow cells 33% of mice from death. Conclusions/Significance NA-BMCs triggered endogenous hematopoiesis and induced faster recovery compared to bone marrow controls. These findings may be of relevance in the refinement of strategies in the treatment of hematological malignancies

    Beyond Bukhara: trade, identity and interregional exchange across Asia

    This article explores the nature of inter-Asian trade dynamics through a consideration of the role played by traders from northern Afghanistan’s Central Asian borderlands. It explores the role that traders from this region have played in commercial exchanges involving China, the Arabian Peninsula and a range of settings in West Asia. In addition to documenting the inter-Asian scope of these traders’ activities, the article also addresses the shifting nature of their identity formations in relationship to successive waves of migration. The traders often identify themselves in relationship to ethno-national identity categories (Turkmen, Uzbek and Tajik) that are politically salient in Central Asia and Afghanistan today. At the same time, the traders also emphasise their being from families that migrated from the territories of the Emirate of Bukhara during the early years of communist rule in the 1920s and 1930s. In the context of the Soviet invasion of Afghanistan in 1979, many of these families moved from Afghanistan to Saudi Arabia, often staying for several years in cities and towns in Pakistan.     GATA1s induces hyperproliferation of eosinophil precursors in Down syndrome transient leukemia

    Transient leukemia (TL) is evident in 5–10% of all neonates with Down syndrome (DS) and associated with N-terminal truncating GATA1-mutations (GATA1s). Here we report that TL cell clones generate abundant eosinophils in a substantial fraction of patients. Sorted eosinophils from patients with TL and eosinophilia carried the same GATA1s-mutation as sorted TL-blasts, consistent with their clonal origin. TL-blasts exhibited a genetic program characteristic of eosinophils and differentiated along the eosinophil lineage in vitro. Similarly, ectopic expression of Gata1s, but not Gata1, in wild-type CD34+-hematopoietic stem and progenitor cells induced hyperproliferation of eosinophil promyelocytes in vitro. While GATA1s retained the function of GATA1 to induce eosinophil genes by occupying their promoter regions, GATA1s was impaired in its ability to repress oncogenic MYC and the pro-proliferative E2F transcription network. ChIP-seq indicated reduced GATA1s occupancy at the MYC promoter. Knockdown of MYC, or the obligate E2F-cooperation partner DP1, rescued the GATA1s-induced hyperproliferative phenotype. In agreement, terminal eosinophil maturation was blocked in Gata1Δe2 knockin mice, exclusively expressing Gata1s, leading to accumulation of eosinophil precursors in blood and bone marrow. These data suggest a direct relationship between the N-terminal truncating mutations of GATA1 and clonal eosinophilia in DS patients

    Endogenous tumor suppressor microRNA-193b: Therapeutic and prognostic value in acute myeloid leukemia

    Purpose Dysregulated microRNAs are implicated in the pathogenesis and aggressiveness of acute myeloid leukemia (AML). We describe the effect of the hematopoietic stem-cell self-renewal regulating miR-193b on progression and prognosis of AML. Methods We profiled miR-193b-5p/3p expression in cytogenetically and clinically characterized de novo pediatric AML (n = 161) via quantitative real-time polymerase chain reaction and validated our findings in an independent cohort of 187 adult patients. We investigated the tumor suppressive function of miR-193b in human AML blasts, patient-derived xenografts, and miR-193b knockout mice in vitro and in vivo. Results miR-193b exerted important, endogenous, tumor-suppressive functions on the hematopoietic system. miR-193b-3p was downregulated in several cytogenetically defined subgroups of pediatric and adult AML, and low expression served as an independent indicator for poor prognosis in pediatric AML (risk ratio 6 standard error, 20.56 6 0.23; P = .016). miR-193b-3p expression improved the prognostic value of the European LeukemiaNet risk-group stratification or a 17-gene leukemic stemness score. In knockout mice, loss of miR-193b cooperated with Hoxa9/Meis1 during leukemogenesis, whereas restoring miR-193b expression impaired leukemic engraftment. Similarly, expression of miR-193b in AML blasts from patients diminished leukemic growth in vitro and in mouse xenografts. Mechanistically, miR-193b induced apoptosis and a G1/S-phase block in various human AML subgroups by targeting multiple factors of the KIT-RAS-RAF-MEK-ERK (MAPK) signaling cascade and the downstream cell cycle regulator CCND1. Conclusion The tumor-suppressive function is independent of patient age or genetics; therefore, restoring miR-193b would assure high antileukemic efficacy by blocking the entire MAPK signaling cascade while preventing the emergence of resistance mechanisms

    This collection brings together a variety of anthropological, historical and sociological case studies from Central Asia and the Caucasus to examine the concept of translocality. The chapters scrutinize the capacity of translocality to describe, in new ways, the multiple mobilities, exchange practices and globalizing processes that link places, people and institutions in Central Asia and the Caucasus with others in Russia, China and the United Arab Emirates. Illuminating translocality as a productive concept for studying cross‐regional connectivities and networks, this volume is an important contribution to a lively field of academic discourse. Following new directions in Area Studies, the chapters aim to overcome ‘territorial containers’ such as the nation‐state or local community, and instead emphasize the significance of processes of translation and negotiation for understanding how meaningful localities emerge beyond conventional boundaries. Structured by the four themes ‘crossing boundaries’, ‘travelling ideas’, ‘social and economic movements’ and ‘pious endeavours’, this volume proposes three conceptual approaches to translocality: firstly, to trace how it is embodied, narrated, virtualized or institutionalized within or in reference to physical or imagined localities; secondly, to understand locality as a relational concept rather than a geographically bounded unit; and thirdly, to consider cross‐border traders, travelling students, business people and refugees as examples of non-elite mobilities that provide alternative ways to think about what ‘global’ means today.
