    Expression of ornithine decarboxylase enzyme in central nervous system tissue in experimental allergic encephalomyelitis (EAE) in the Lewis rat

    Ornithine decarboxylase (ODC) is the primary and rate-limiting enzyme in the natural synthesis of polyamines putrescine, spermidine and spermine, which have been implicated in the breakdown of the blood-brain barrier (BBB), the initial event in the pathogenesis of experimental allergic encephalomyelitis (EAE) and multiple sclerosis (MS). This thesis INOUid like to suggest that there is increased expression of ODC during heightened disease expression in EAE, and therefore that ODC is involved in the development of EAE and MS through the polyamine synthetic pathway. To assess the levels of ODC enzyme in central nervous system (CNS) tissue, which included cerebellum, cervical spinal cord and medulla, the tissue was first homogenised and the protein concentration of the resulting sample determined. This required the establishment of an appropriate protein estimation mP.thod i.e. a suitable protein assay. Much time and experimentation was spent on achieving this because of the effect of interfering substances on the assays used. Assessment of the levels of ODC in a model of EAE using normal tissue, CFA treated tissue (control), day-13 post inoculation (PI) (height of disease) and day-21 PI (early recovery stage) was carried out by Western blotting. The studies show that there is increased expression of ODC at the height of disease i.e. day-13 PI in cerebellum and medulla pons tissue but not cervical spinal cord tissue

    A Psychoanalytic Qualitative Study of Subjective Life Experiences of Women With Breast Cancer

    This article exemplifies research on the subjective life experiences of women with breast cancer, designed from a psychoanalytic perspective. Such research aims to reveal the subjective intrapsychic processes of women suffering from breast cancer, which can provide researchers and health care professionals with useful insight. Using Biographic narrative interpretative method, the study reveals some common denominators in the subjective life experiences of women with breast cancer. The study revealed that the subjects consider the diagnosis of breast cancer as one of the four main events in their lives. Even though these events are very different from each other, in terms of type and severity, the results indicate a common process in the way these life events are experienced by the participants. These results are relevant for health care practitioners in structuring their psychosocial support programs so as to better accommodate their patients' psychological needs

    Early antiretroviral therapy not associated with higher cryptococcal meningitis mortality in people with human immunodeficiency virus in high-income countries: An international collaborative cohort study

    BACKGROUND: Randomized controlled trials (RCTs) from low- and middle-income settings suggested that early initiation of antiretroviral therapy (ART) leads to higher mortality rates among people with HIV (PWH) who present with cryptococcal meningitis (CM). There is limited information about the impact of ART timing on mortality rates in similar people in high-income settings. METHODS: Data on ART-naive PWH with CM diagnosed from 1994 to 2012 from Europe/North America were pooled from the COHERE, NA-ACCORD, and CNICS HIV cohort collaborations. Follow-up was considered to span from the date of CM diagnosis to earliest of the following: death, last follow-up, or 6 months. We used marginal structural models to mimic an RCT comparing the effects of early (within 14 days of CM) and late (14-56 days after CM) ART on all-cause mortality, adjusting for potential confounders. RESULTS: Of 190 participants identified, 33 (17%) died within 6 months. At CM diagnosis, their median age (interquartile range) was 38 (33-44) years; the median CD4+ T-cell count, 19/μL (10-56/μL); and median HIV viral load, 5.3 (4.9-5.6) log10 copies/mL. Most participants (n = 157 [83%]) were male, and 145 (76%) started ART. Mimicking an RCT, with 190 people in each group, there were 13 deaths among participants with an early ART regimen and 20 deaths among those with a late ART regimen. The crude and adjusted hazard ratios comparing late with early ART were 1.28 (95% confidence interval, .64-2.56) and 1.40 (.66-2.95), respectively. CONCLUSIONS: We found little evidence that early ART was associated with higher mortality rates among PWH presenting with CM in high-income settings, although confidence intervals were wide

    Evaluation of Diffusion Coefficient and Surface Emission Coefficient by an Optimization Technique

    The rate at which wood dries can be expressed by two factors:(1) the diffusion coefficient, as an indication of internal resistance to moisture removal, and (2) the surface emission coefficient, as an indication of external resistance to moisture removal. An optimization technique was applied to calculate these two coefficients. Unlike existing methods where these coefficients are evaluated at only one point, usually at one-half of the evaporable water remaining in wood, this optimization technique used the entire drying data. The technique searches for the optimum pair of coefficients based on the least squares principle. The performance of this method was demonstrated by reanalyzing published data using a digital computer. The results indicate that a more accurate prediction of the drying process can be achieved by the optimization method than by existing methods.
