Intoxicação alimentar por queijo Minas contaminado com Staphylococcus aureus
Relata-se surto de intoxicação alimentar ocorrido em julho de 1987, na cidade de Ouro Preto, MG (Brasil). O alimento causador foi um queijo Minas, contaminado por Staphylococcus aureus ao nível de 9,3 x 10(7) UFG/g. Detectaram-se cepas produtoras de enterotoxinas do tipo A,B,D e E. A amostra analisada revelou contaminação por coliformes fecais acima de 1,1 x 10(5)/g(NMP), mas não continha Salmonella.Devido aos sintomas característicos e à elevada contaminação, concluiu-se que o Staphylococcus aureus foi o patogênico responsável pelo surto.An outbreak of food poisoning which occurred in July, 1987, in the city of Ouro Preto, State of Minas Gerais, Brazil, is reported. The food involved was a Minas-type cheese, contamined by Staphylococcus aureus to the level of 9.3 x 10(7) CFU/g. Enterotoxin producing strains of types A,B,D and E were detected. The sample analysed also showed contamination by fecal coliforms above 1.1 x 10(5)/g (MPN), but Salmonella were not present. Due to characteristic symptoms and high contamination it was concluded that Staphylococcus aureus was the pathogene responsible for the outbreak
Application of infrared spectral techniques on quality and compositional attributes of coffee: An overview
AbstractDuring the last two decades, near and mid-infrared spectral analyses have emerged as a reliable and promising analytical tool for objective assessment of coffee quality attributes. The literature presented in this review clearly reveals that near and mid-infrared approaches have a huge potential for gaining rapid information about the chemical composition and related properties of coffee. In addition to its ability for effectively quantifying and characterising quality attributes of some important features of coffee such as moisture, lipids and caffeine content, classification into quality grades and determination of sensory attributes, it is able to measure multiple chemical constituents simultaneously avoiding extensive sample preparation. Developing a quality evaluation system based on infrared spectral information to assess the coffee quality parameters and to ensure its authentication would bring economical benefits to the coffee industry by increasing consumer confidence in the quality of products. This paper provides an overview of the recently developed approaches and latest research carried out in near and mid-infrared spectral technology for evaluating the quality and composition of coffee and the possibility of its widespread deployment
Milho orgânico: impacto da adubação com lixiviado de aterro sanitário na composição química, produtividade e concentração de metais em grãos
PRODUÇÃO E CARACTERIZAÇÃO DE AMILASE DE Fusarium moniliforme E Aspergillus flavus
Considerando o interesse econômico da cultura do
milho, alvo de ataque de fungos micotoxigênicos,
esta investigação teve por objetivo a produção e
caracterização bioquímica parcial de amilase das
principais espécies problemáticas (Fusarium
moniliforme e Aspergillus flavus). Empregandose
cultivo em caldo de milho verde, adicionado de milho
triturado, a maximização da produção de amilase de
F. moniliforme e A. flavus ocorreu no 15o dia de
fermentação. Submetendose
o sobrenadante de
cultura à precipitação fracionada com sulfato de
amônio, entre 60 a 90% de saturação, obtevese
a
amilase fúngica, que foi caracterizada perante pH,
temperatura e estabilidade térmica por metodologia
de superfície de resposta. Em ambos os grupos
fúngicos, a atividade enzimática foi maximizada em
temperatura próxima a 40 °C, com pH em torno de
6,7 para amilase de F. moniliforme e 5,5 para A.
flavus. A máxima estabilidade térmica da enzima de
F. moniliforme ocorreu próximo a 21,0 °C.
Abstract
Regarding the economic relevance of corn culture and the fact that it can be a target to
micotoxigenic fungi under improper storage conditions, this research aims the production
of amylase and its partial biochemical characterization of the main problematic species
Fusarium moniliforme and Aspergillus flavus. The fungi were cultivated in prepared
medium containing both unripe and ground corn kernels. Maximum amylase production
occurred on the 15th day of fermentation for both strains. The supernatant phase was
fractionally precipitated with ammonium sulfate between 60 and 90% of saturation and the
separated amylase had its parameters (pH, temperature and thermal stability)
characterized by response surface methodology. Maximum activity was obtained at 40 °C
and pH of 6.7 for the amylase of F. moniliforme and pH of 5.5 for the amylase of A. flavus.
The optimal thermal stability for the enzyme of F. moniliforme occurred at 21 °C
Sinergismo entre ácidos orgânicos e sorbato de potássio no controle de Aspergillus flavus
Devido aos riscos de contaminação por micotoxinas, o controle fúngico em alimentos deve ser adotar medidas desde o início do desenvolvimento microbiano. A substituição de fungicidas químicos por ácidos orgânicos (geralmente reconhecidos como seguro) desponta como uma perspectiva interessante. A pesquisa teve como objetivo determinar concentração inibitória mínima (CIM) de ácidos orgânicos, sorbato de potássio e respectivas combinações no controle de Aspergillus flavus NRRL 3251 produtor de aflatoxina B1. O inóculo de 104 esporos.mL-1 foi calculado baseado na contaminação real em milho e ração animal. As melhores CIM in vitro foram: combinação de ácido acético (AA 83,26 mM) + propiônico (AP 6,74 mM); AA (8,32 mM) + sorbato de potássio (SP 13,31mM) e AP (6,74 mM) + SP (33,28 mM). Considerando a dose comercialmente utilizada de ácido propiônico 0,5%, a combinação do ácido propiônico com sorbato de potássio e ácido acético poderia reduzir respectivamente cerca de 24,63 e 70,19% o custo em conservante
CROMATOGRAFIA DE AFINIDADE COM ÍON CU2+: ELIMINAÇÃO DE FALSO-POSITIVOS NA DETECÇÃO DE ENTEROTOXINA ESTAFILOCÓCICA
Avaliou-se a aplicação da cromatografia de afinidade com
íon Cu2+ para minimizar a interferência de componentes
alimentares na detecção de enterotoxinas estafilocócicas
mediante imunoensaio, baseado em aglutinação de látex
(RPLA). Os ensaios preliminares com chantilly contaminado
com 25 ng de enterotoxina estafilocócica A (EEA) indicaram
que a cromatografia de afinidade com íon Cu2+ eliminou
interferentes que causavam reação falso-positiva em RPLA.
A eficiência do procedimento de limpeza foi testada em 15
extratos alimentares (de surtos e/ou com alta contagem
estafilocócica), compreendendo vários tipos de alimentos
comumente consumidos no Brasil. Onze extratos alimentares
apresentaram um pico único de eluição com perfis
semelhantes e as frações testadas por RPLA demonstraram
EEA (4 alimentos), EEB (1), EEA + EEB (3), EEA + EED (2) e
EEA + EEB + EED (1). Os problemas de reações inespecíficas
com kit RPLA em 2 extratos de bolos recheados foram
eliminados, indicando que a adoção desta operação evitaria
a reação falso-positiva.
CU2+ AFFINITY CHROMATOGRAPHY: ELIMINATION OF FALSE-POSITIVE IN THE
DETECTION OF STAPHYLOCOCCAL ENTEROTOXIN
Abstract
The application of Cu2+ chelate Sepharose affinity chromatography was evaluated to
reduce the interference of food constituents in the detection of staphylococcal enterotoxin
with reversed passive latex agglutination immunoassay (RPLA). The preliminary assay
using chantilly contaminated with 25 nanograms of staphylococcal enterotoxin A (SEA)
indicated that Cu2+ ion affinity chromatography eliminated the food matrix constituents
responsible for false-positive reaction of RPLA. The efficiency of this clean-up procedure
was tested in 15 food extracts (from outbreaks and/or with high staphylococcal count), comprising various types of food commonly consumed in Brazil. Eleven food extracts
showed only one elution peak with similar profile and the fractions assayed by RPLA
demonstrated SEA (4 food), SEB (1), SEA + SEB (3), SEA + SED (2) e SEA + SEB +
SED (1). The problems of nonspecific reactions with the RPLA kit in 2 cake extracts
were eliminated, indicating that the adoption of this operation avoids the falsepositive
reaction
