    Characterization of Methicillin-Resistant Staphylococcus aureus Isolated from Healthy Turkeys and Broilers Using DNA Microarrays

    Methicillin-resistant Staphylococcus aureus (MRSA) is a major human health problem and recently, domestic animals are described as carriers and possible reservoirs. Twenty seven S. aureus isolates from five turkey farms (n = 18) and two broiler farms (n = 9) were obtained by culturing of choana and skin swabs from apparently healthy birds, identified by Taqman-based real-time duplex nuc-mecA-PCR and characterized by spa typing as well as by a DNA microarray based assay which covered, amongst others, a considerable number of antibiotic resistance genes, species controls, and virulence markers. The antimicrobial susceptibility profiles were tested by agar diffusion assays and genotypically confirmed by the microarray. Five different spa types (3 in turkeys and 2 in broilers) were detected. The majority of MRSA isolates (24/27) belonged to clonal complex 398-MRSA-V. The most frequently occurring spa types were accordingly t011, t034, and t899. A single CC5-MRSA-III isolated from turkey and CC398-MRSA with an unidentified/truncated SCCmec element in turkey and broiler were additionally detected. The phenotypic antimicrobial resistance profiles of S. aureus isolated from both turkeys and broilers against 14 different antimicrobials showed that all isolates were resistant to ampicillin, cefoxitin, oxacillin, doxycycline, and tetracycline. Moreover, all S. aureus isolated from broilers were resistant to erythromycin and azithromycin. All isolates were susceptible to gentamicin, chloramphenicol, sulphonamides, and fusidic acid. The resistance rate against ciprofloxacin was 55.6% in broiler isolates and 42.1% in turkey isolates. All tetracycline resistant isolates possessed genes tetK/M. All erythromycin- resistant broiler isolates carried ermA. Only one broiler isolate (11.1%) carried genes ermA, ermB, and ermC, while 55.6% of turkey isolates possessed ermA and ermB genes. Neither PVL genes (lukF/S-PV), animal-associated leukocidin (lukM and luk-P83) nor the gene encoding the toxic shock syndrome toxin (tst1) were found in turkey and broiler isolates. In conclusion, the detection of MRSA in healthy turkeys and broilers with even additional antibiotic resistance markers is of major public health concern.     Serological and Molecular Identification of Brucella spp. in Pigs from Cairo and Giza Governorates, Egypt

    Brucellosis is considered as endemic disease of animals and humans since thousands of years in Egypt. However, brucellosis in pigs has never been reported in Egypt. Thus, serological and molecular assays were applied to detect anti-Brucella antibodies and DNA in serum samples collected from pigs. In total 331 blood samples collected from male and female pigs at slaughterhouses of Cairo and Giza governorates were investigated using Brucella c- and i-ELISA and Brucella real-time PCR. Anti-Brucella antibodies were detected in 16 (4.83%) and 36 (10.8%) sera by i-ELISA and c-ELISA, respectively. Brucella DNA was detected in 10 (3.02%) seropositive samples and identified as Brucella melitensis (7/10) and Brucella suis (3/10). A higher prevelance was found in boars. This is the first study investigating pig brucellosis in Egypt. The results of this study will raise awareness for brucellosis in these farm animals and will help to develop effective control strategies

    Effects of exogenous enzymes, Lactobacillus acidophilus or their combination on feed performance response and carcass characteristics of rabbits fed sugarcane bagasse

    The aim of this study was to evaluate the effect of Lactobacillus acidophilus (LAC), exogenous enzymes of ZAD® (exogenous enzyme preparation) or their combination on feed conversion, and dressing of rabbits fed different treatments of sugarcane bagasse (SCB). Five rations were allotted randomly to five groups of New-Zealand White (NZW) rabbits (n=10) with initial live body weight of (838±42.4) g and 5 weeks of age. Rabbits were fed on diets with different sources of fiber as follows, (i) a control diet composed of 100% berseem hay and 0% SCB, (ii) 50% berseem hay and 50% untreated SCB (USCB), (iii) 50% berseem hay and 50% SCB treated with L. acidophilus (LAC), (iv) 50% berseem hay and 50% SCB treated with ZAD® (ZAD), and (v) 50% berseem hay and 50% SCB treated with a combination of LAC+ZAD® (LZ). Treatments of SCB with L. acidophilus, ZAD® and LAC+ZAD® had the highest feed conversion ratio than both USCB and control. The dressing percentage of rabbits that fed the LAC and LZ diets was higher (P<0.05) compared with that in the other groups. Performance index (PI) for LAC group was improved (P<0.05) compared to that for the other groups; however, PI for USCB group was the lowest (P<0.05). It could be concluded that treating SCB with L. acidophilus, exogenous enzymes of ZAD® or their combination improved feed conversion and performance with more positive effects with L. acidophilus than the other treatments

    Prevalence, genotyping and risk factors of thermophilic Campylobacter spreading in organic turkey farms in Germany

    Background The need for organic food of animal origin has increased rapidly in recent years. However, effects of organic animal husbandry on food safety have not been rigorously tested especially in meat turkey flocks. This study provides for the first time an overview on the prevalence and genetic diversity of Campylobacter species (spp.) in five organic meat turkey farms located in different regions in Germany, as well as on potential risk factors of bacterial spreading. Thirty cloacal swabs as well as water samples and darkling beetles were collected from each flock and examined for the presence of Campylobacter by conventional and molecular biological methods. The isolates were genotyped by flaA-RFLP. Results Campylobacter spp. were detected in cloacal swabs in all 5 turkey flocks with prevalence ranged from 90.0 to 100 %. 13 cloacal swabs collected from birds in farm III and IV were harboured mixed population of thermophilic campylobacters. In total, from 158 Campylobacter isolated from turkeys 89 (56.33 %) were identified as C. coli and 69 (43.76 %) as C. jejuni. Three Campylobacter (2 C. jejuni and 1 C. coli) were detected in drinkers of two farms and 3 C. coli were isolated from darkling beetles of one farm. No Campylobacter were isolated from main water tanks. flaA-RFLP assay showed that turkey farms can harbour more than one genotype. In a single turkey two different genotypes could be detected. The genotypes of campylobacters isolated from water samples or beetles were identical with those isolated from turkeys. No effect was found of some environmental parameters [ammonia concentration (NH3), carbon dioxide concentration (CO2), relative humidity (RH) and air temperature)] on Campylobacter prevalence in organic turkey farms. Additionally, drinking water and darkling beetles might be considered as risk factors for the spreading of Campylobacter in turkey flocks. Conclusions This study highlights the high prevalence and genotypic diversity of Campylobacter spp. isolated from organic turkey flocks. Further research is needed to assess other potential risk factors responsible for bacteria spreading in order to mitigate the spread of Campylobacter in organic turkey flocks by improving biosecurity control measures

    The aim of this study was to evaluate the effect of Lactobacillus acidophilus (LAC), exogenous enzymes ZAD® or their combination on feed intake, nutrient digestibility and performance of rabbits fed increasing levels of sugarcane bagasse (SCB). Five rations were allotted randomly to five groups of New Zealand white (NZW) rabbits (838±42.4g average BW at 5 weeks of age. Treatment of SCB with Lactobacillus acidophilus, ZAD® and LAC+ ZAD® led to higher CP content and lower contents of ADF, NDF and DM. Total DM intake was not affected by treatments. Digestibility coefficient of CP for LAC and LZ were higher (P<0.05) compared to the other groups. The BW for LAC and LZ rabbits was higher (P<0.05) than for the other groups, while the BW for LAC rabbit was the highest (P<0.05) and for USCB was the lowest. It can be concluded that treating sugarcane bagasse with Lactobacillus acidophilus, exogenous enzymes of ZAD® or their combination improved feeding values; however, the treatment with Lactobacillus acidophilus was found to be the best

    A Finite Element Model for Recognizing Breast Cancer

    Abstract Breast cancer recognition is an important issue in elastography diagnostic imaging. Breast tumor biopsy has been for many years the reference procedure to assess histological definition for breast diseases. But biopsy measurement is an invasive method besides it takes larger time. So, fast and improved methods are needed. Using elastography technology, a digital image correlation technique can be used to calculate the displacement of breast tissue after it has suffered a compression force. This displacement is related to tissue stiffness, and breast cancer can be classified into benign or malignant according to that displacement. The value of compression force affects the displacement of tissue, and then affects the results of the breast cancer recognition. Finite element method was being used to simulate a model for the breast cancer as a phantom to be used in measurements and study of breast cancer diagnosis. The breast cancer using this phantom can be recognized within a short time. The proposed work succeeded in recognizing breast tumor phantom by an average correct recognition ratio CRR of about 94.25% on a simulation environment. The strain ratio SR for benign and malignant models is also computed. The result of the simulated breast tumor model is compared with real data of 10 lesion cases (6 benign and 4 malignant). The coefficient of variation CV between the simulated SR and the SR using real data reaches to about 5% for benign lesions and 4.78% for malignant lesions. The results of CRR and CV in this proposed work assure that the proposed breast cancer model using finite element modeling is a robust technique for breast tumor simulation where the behavior of real data of breast cancer can be predicted

    Effect of Mediterranean saltbush (Atriplex halimus) ensilaging with two developed enzyme cocktails on feed intake, nutrient digestibility and ruminal fermentation in sheep

    The aim of this study was to assess the effects of feeding Atriplex halimus (AH) silage treated with two developed enzyme cocktails to sheep on feed intake, nutrient digestibility and ruminal fermentation. The AH silage was treated without or with 2 L of ZAD1® or ZAD2®/1000 kg with 5% molasses and ensiled for 30 days. Barley grain (300 g/head/day) was fed as an energy supplement once daily at 10.00 hours and AH silage with or without enzyme treatment was offered ad libitum to animals twice daily at 09.00 and 16.00 hours. Sheep were fed on four experimental forage diets comprised of AH silage and barley (D1), AH silage treated with ZAD1® and barley (D2), AH silage treated with ZAD2® and barley (D3) and AH silage treated with a combination of ZAD1® and ZAD2® (1:1) and barley (D4). Ensiling AH with enzymes reduced its contents of neutral detergent fiber and acid detergent fiber. The dry matter intake of AH of D2, D3 and D4 decreased (P < 0.001) as compared to D1. However, enzyme-treated diets had greater total digestible nutrients intake (P < 0.001) as compared to D1. The nutrients digestibility for D2, D3 and D4 were higher than those for D1 (P < 0.001), and were higher for D3 as compared to both D2 and D4. Sheep fed on D3 had highest (P < 0.001) ruminal total volatile fatty acids concentration, ammonia nitrogen concentration and microbial protein yield. It could be concluded that AH silage treated with ZAD1® or ZAD2® improved digestibility and rumen fermentation in sheep

    The effects of feeding Atriplex halimus treated with three developed enzyme cocktails to Barki sheep on feed intake, nutrient digestibility, N utilization, and ruminal fermentation were assessed. A. halimus was ensiled with two developed enzyme cocktails of ZAD1® (Z1) and/or ZAD2® (Z2) as liquid enzyme preparations (2 l/t) with 5% molasses and ensiled for 30 days. Three Barki rams (45 ± 3.2 kg) were used per treatment in five consecutive digestibility trials, while three ewes fitted with a permanent rumen fistula were used as source of inoculum for in vitro rumen fermentation trials. Barley grain (300 g/animal/day) was fed as energy supplement during the experimental trial for all diets. Five diets were composed as follows: A. halimus (leaves and stems) (D1); untreated A. halimus plus 4 g/animal/day ZADO® (Z) (enzyme preparation in powder form) (D2); A. halimus ensiled with Z1 and barley plus 4 g/animal/day Z (D3); A. halimus ensiled with Z2 and barley (D3) plus 4 g/animal/day Z (D4); A. halimus ensiled with a combination of Z1 and Z2 (1 :1) and barley plus 4 g/head/day Z (D5). For all trials, ad libitum A. halimus was offered twice a day at 9:00 and 16:00 h while barley grain was given once a day at 10:00 h. Both D1 and D2 diets increased (P <0.001) dry matter intake of A. halimus and total dry matter intake. Addition of 4 g/day of Z to Z1 and/or Z2 ensiled diets improved (P < 0.0001) organic matter, crude protein, crude fibre, and neutral detergent fibre digestibilities. Diets D1 and D2 increased (P < 0.001) N intake, whereas the direct addition of Z to D3, D4, and D5 decreased (P < 0.001) N balance and N balance/N absorption ratio. Sheep fed on Z in addition to Z2 ensiled A. halimus showed higher improvements for total volatile fatty acids (P < 0.001), ammonia N (P = 0.007), and microbial protein production (P = 0.003). It can be concluded that feeding sheep on A. halimus ensiled with Z1 and Z2 with direct feeding of Z enzyme preparation improved intake, digestibility, nitrogen balance and utilization, as well as rumen fermentation
