    IDEF3 formalization report

    The Process Description Capture Method (IDEF3) is one of several Integrated Computer-Aided Manufacturing (ICAM) DEFinition methods developed by the Air Force to support systems engineering activities, and in particular, to support information systems development. These methods have evolved as a distillation of 'good practice' experience by information system developers and are designed to raise the performance level of the novice practitioner to one comparable with that of an expert. IDEF3 is meant to serve as a knowledge acquisition and requirements definition tool that structures the user's understanding of how a given process, event, or system works around process descriptions. A special purpose graphical language accompanying the method serves to highlight temporal precedence and causality relationships relative to the process or event being described

    Cellular interactions with hydrogels

    An efficient means of evaluating potential biomaterials is to use the in vitro fibroblast cell culture model. However, the chemistry which influences cell adhesion on polymer substrates is poorly understood. The work in this thesis aims to rationalise several theories of current opinion and introduce new chemical techniques that may predict cellular behaviour. The keratoprosthesis is a typical example of the need to be able to manipulate cell adhesion of materials since both adhesive and non adhesive sections are needed for proper integration and optical function. Calcein AM/ethidium homodimer-1 and DAPI assays were carried out using 3T3 and EKl.BR cells. Poly(HEMA) was found to be the most cell adhesive hydrogel tested. The reactivity of monomers and the resulting sequence distribution were found to affect surface properties and this may explain the poor levels of cell adhesion seen on NVP/MMA copolymers. Surface free energy is shown to be dependent on the polar and non polar groups present along the backbone chain of the polymers. Dehydrated and hydrated contact angle measurements show the effect of rotation of surface groups around the backbone chain. This effect is most apparent on hydrogels containing methacrylic acid. Dynamic contact angle measurements confirm sequence distribution irregularities and demonstrate the mobility of surface groups. Incorporation of NVI or DEAEMA into the hydrogels does not affect the mobility of the surface groups despite their bulkiness. Foetal calf serum was used for the first time as a test solution in an attempt to mimic a biological environment during surface experiments. A Vroman effect may be present, and may involve different surface proteins for each material tested. This interdisciplinary study combines surface characterisation and biological testing to further the knowledge of the biomaterial/host interface. Surface chemistry techniques appear to be insufficiently sensitive to predict cellular behaviour. The degree of ionisation of hydrogels containing ionic groups depends on the nature of the functional groups as well as the concentration and this is an important parameter to consider when comparing charged materials

    The genomic framework that enables corals to adjust to unfavourable conditions is crucial for coral reef survival in a rapidly changing climate. We have explored the striking intraspecific variability in the expression of coral pigments from the green fluorescent protein (GFP) family to elucidate the genomic basis for the plasticity of stress responses among reef corals. We show that multicopy genes can greatly increase the dynamic range over which corals can modulate transcript levels in response to the light environment. Using the red fluorescent protein amilFP597 in the coral Acropora millepora as a model, we demonstrate that its expression increases with light intensity, but both the minimal and maximal gene transcript levels vary markedly among colour morphs. The pigment concentration in the tissue of different morphs is strongly correlated with the number of gene copies with a particular promoter type. These findings indicate that colour polymorphism in reef corals can be caused by the environmentally regulated expression of multicopy genes. High-level expression of amilFP597 is correlated with reduced photodamage of zooxanthellae under acute light stress, supporting a photoprotective function of this pigment. The cluster of light-regulated pigment genes can enable corals to invest either in expensive high-level pigmentation, offering benefits under light stress, or to rely on low tissue pigment concentrations and use the conserved resources for other purposes, which is preferable in less light-exposed environments. The genomic framework described here allows corals to pursue different strategies to succeed in habitats with highly variable light stress levels. In summary, our results suggest that the intraspecific plasticity of reef corals’ stress responses is larger than previously thought

    Are Home Offices Feasible in a University?: Faculty Perceptions of a Home Office Experiment

    This article examines faculty perceptions of how the new technologies of E-mail and voice mail that were widely adopted as a result of the home office experiment changed the ways of working and ways of thinking about what is important. The authors discussed both first- and second-level effects of communication technologies in three areas: (1) on faculty interaction both within the department and in the larger campus community, (2) on student/faculty interaction, and (3) on work styles and the ways faculty members think about their work. When the home office project was envisioned, the goal was to use communication technology to create a work environment in which faculty without an office on campus could come to campus simply to teach classes and perhaps occasionally attend meetings, while performing most other duties off campus. The project seemed more palatable to most faculty in the department and to administrators because it was temporary, but it did require all faculty members to move twice within a three year period

    Caught in the act: Implications for the increasing abundance of mafic enclaves during the eruption of the Soufriere Hills Volcano, Montserrat

    An exceptional opportunity to sample several large blocks sourced from the same region of the growing Soufrière Hills lava dome has documented a significant increase in the presence of mafic enclaves in the host andesite during the course of a long-lived eruptive episode with several phases. In 1997 (Phase I) mafic inclusions comprised ~1 volume percent of erupted material; in 2007 (Phase III) deposits their volumetric abundance increased to 5–7 percent. A broader range of geochemically distinctive types occurs amongst the 2007 enclaves. Crystal-poor enclaves generally have the least evolved (basaltic) compositions; porphyritic enclaves represent compositions intermediate between basaltic and andesitic compositions. The absence of porphyritic enclaves prior to Phase III magmatism at Soufrière Hills Volcano suggests that a mixing event occurred during the course of the current eruptive episode, providing direct evidence consistent with geophysical observations that the system is continuously re-invigorated from depth

    Three-dimensional Structure of the Central Mitotic Spindle of Diatoma vulgare

    Central mitotic spindles in Diatoma vulgare have been investigated using serial sections and electron microscopy. Spindles at both early stages (before metaphase) and later stages of mitosis (metaphase to telophase) have been analyzed. We have used computer graphics technology to facilitate the analysis and to produce stereo images of the central spindle reconstructed in three dimensions. We find that at prometaphase, when the nuclear envelope is dissassembling, the spindle is constructed from two sets of polar microtubules (MTs) that interdigitate to form a zone of overlap. As the chromosomes become organized into the metaphase configuration, the polar MTs, the spindle, and the zone of overlap all elongate, while the number of MTs in the central spindle decreases from greater than 700 to approximately 250. Most of the tubules lost are short ones that reside near the spindle poles. The previously described decrease in the length of the zone of overlap during anaphase central spindle elongation is clearly demonstrated in stereo images. In addition, we have used our three-dimensional data to determine the lengths of the spindle MTs at various times during mitotis. The distribution of lengths is bimodal during prometaphase, but the short tubules disappear and the long tubules elongate as mitosis proceeds. The distributions of MT lengths are compared to the length distributions of MTs polymerized in vitro, and a model is presented to account for our findings about both MT length changes and microtubule movements
