    Spider venom neurotoxin based bioinsecticides: A novel bioactive for the control of the Asian citrus psyllid Diaphorina citri (Hemiptera)

    The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Psyllidae), is a key vector of the phloem-limited bacteria Candidatus Liberibacter asiaticus (CLas) associated with huanglongbing (HLB), the most serious and currently incurable disease of citrus worldwide. Here we report the first investigation into the potential use of a spider venom-derived recombinant neurotoxin, ω/κ-HxTx-Hv1h (hereafter HxTx-Hv1h) when delivered alone or when fused to snowdrop lectin (Galanthus nivalis agglutinin; GNA) to control D. citri. Proteins, including GNA alone, were purified from fermented transformed yeast Pichia pastoris cultures. Recombinant HxTx-Hv1h, HxTx-Hv1h/GNA and GNA were all orally toxic to D. citri, with Day 5 median lethal concentrations (LC 50) derived from dose-response artificial diet assays of 27, 20 and 52 μM, respectively. Western analysis of whole insect protein extracts confirmed that psyllid mortality was attributable to protein ingestion and that the fusion protein was stable to cleavage by D. citri proteases. When applied topically (either via droplet or spray) HxTx-Hv1h/GNA was the most effective of the proteins causing >70 % mortality 5 days post treatment, some 2 to 3-fold higher levels of mortality as compared to the toxin alone. By contrast, no significant mortality or phenotypic effects were observed for bumble bees (Bombus terrestris L.) fed on the recombinant proteins in acute toxicity assays. This suggests that HxTx-Hv1h/GNA has potential as a novel bioinsecticide for the management of D. citri offering both enhanced target specificity as compared to chemical pesticides and compatibility with integrated pest management (IPM) strategies

    A New Method to Predict the Epidemiology of Fungal Keratitis by Monitoring the Sales Distribution of Antifungal Eye Drops in Brazil

    Purpose: Fungi are a major cause of keratitis, although few medications are licensed for their treatment. The aim of this study is to observe the variation in commercialisation of antifungal eye drops, and to predict the seasonal distribution of fungal keratitis in Brazil. Methods: Data from a retrospective study of antifungal eye drops sales from the only pharmaceutical ophthalmologic laboratory, authorized to dispense them in Brazil (Opthalmos) were gathered. These data were correlated with geographic and seasonal distribution of fungal keratitis in Brazil between July 2002 and June 2008. Results: A total of 26,087 antifungal eye drop units were sold, with a mean of 2.3 per patient. There was significant variation in antifungal sales during the year (p < 0.01). A linear regression model displayed a significant association between reduced relative humidity and antifungal drug sales (R-2 = 0.17, p < 0.01). Conclusions: Antifungal eye drops sales suggest that there is a seasonal distribution of fungal keratitis. A possible interpretation is that the third quarter of the year (a period when the climate is drier), when agricultural activity is more intense in Brazil, suggests a correlation with a higher incidence of fungal keratitis.     O manejo adequado da irrigação é uma prática agrícola que requer informações para cada cultura explorada, em particular nas fases fenológicas do ciclo produtivo pois muitas espécies possuem períodos críticos durante os quais o estresse hídrico causa sérias reduções na produção. Este trabalho teve, como objetivo, avaliar o efeito de cinco níveis de potencial matricial de água no solo sobre o crescimento e produção do feijão-de-metro. O experimento foi conduzido em casa de vegetação, na área experimental pertencente ao Departamento de Engenharia da Universidade Federal de Lavras, Lavras, MG. O delineamento experimental foi o de blocos casualizados, em esquema fatorial 5 x 2, com quatro repetições. Os tratamentos foram constituídos de cinco potenciais matriciais de água no solo como indicativos do momento de irrigar (-15, -35, -55, -75 e -95 kPa), em duas fases fenológicas (vegetativa e produtiva), monitorados por sensores instalados a 0,15 m de profundidade. Os resultados permitiram concluir que a fase mais sensível ao déficit hídrico foi a produtiva e que a irrigação realizada no potencial matricial em torno de -15 kPa induziu a melhor resposta da cultura quanto ao desenvolvimento, produção e qualidade das vagens.<br>Proper irrigation water management requires knowledge on each exploited crop; particularly regarding vegetative and productive phases since many species have critical periods during which water stress causes serious yield reductions. This study was undertaken so as to assess the effect of five matric potential levels on asparagus bean growth and yield. The experiment was conducted under greenhouse conditions at the experimental area of Department of Engineering, Federal University of Lavras, Lavras, MG. The experimental design was randomized blocks in factorial 5 x 2. The treatments consisted of five matric potentials as indicative of the irrigation scheduling -15 -35 -55 -75 -95 kPa and at two phenological stages (vegetative and reproductive) as monitored by sensors installed at 0.15 m depth. The results allowed to conclude that the most sensitive phase to water deficit was the productive, and that irrigation held at matric potential around -15 kPa led to better crop response in the development, production and quality of pods
