    Aprendizagem e fatores motivacionais relacionados

    Este texto resulta de pesquisa bibliográfica, exploratória, de caráter qualitativo, parte do trabalho de conclusão de pós-graduação, para a qual se utilizou como fontes básicas obras e artigos de autores reconhecidos. O objetivo proposto é demonstrar a importância da motivação no espaço escolar, como ela contribui, interfere nos processos educativos, no ambiente escolar, na aquisição do conhecimento, em síntese, no trabalho do professor, no ensino e na aprendizagem. A arquitetura do texto está estruturada de modo a conceituar motivação e aprendizagem e, posteriormente, relacioná-los com os fatores motivacionais interferentes na aprendizagem e na melhoria da qualidade do ensino. Conclui-se, demonstrando que a motivação é discurso recorrente nos espaços escolares, no entanto, longo caminho a percorrer no que diz respeito a constituir-se em ações objetivas para sua incorporação na prática pedagógica. O que se evidencia são atitudes reveladoras de ausência de motivação para o aprendizado e para o trabalho escolar. Os educadores têm noções elementares a respeito de sua importância no processo educativo, mas falta-lhes conhecimento aprofundado para inseri-las na prática cotidiana e em como fazer uso dos fatores motivacionais no sentido de melhorar as relações com os estudantes e sua aquisição de conhecimento

    A satellite DNA array barcodes chromosome 7 and regulates totipotency via ZFP819.

    Mammalian genomes are a battleground for genetic conflict between repetitive elements and KRAB-zinc finger proteins (KZFPs). We asked whether KZFPs can regulate cell fate by using ZFP819, which targets a satellite DNA array, ZP3AR. ZP3AR coats megabase regions of chromosome 7 encompassing genes encoding ZSCAN4, a master transcription factor of totipotency. Depleting ZFP819 in mouse embryonic stem cells (mESCs) causes them to transition to a 2-cell (2C)-like state, whereby the ZP3AR array switches from a poised to an active enhancer state. This is accompanied by a global erosion of heterochromatin roadblocks, which we link to decreased SETDB1 stability. These events result in transcription of active LINE-1 elements and impaired differentiation. In summary, ZFP819 and TRIM28 partner up to close chromatin across Zscan4, to promote exit from totipotency. We propose that satellite DNAs may control developmental fate transitions by barcoding and switching off master transcription factor genes

    Experimental infection parameters in Galea spixii (Rodentia: Caviidae) with Leishmania infantum chagasi

    In order to better understand the epidemiological transmission network of leishmaniasis, an endemic disease in Northeast Brazil, we investigated the susceptibility of Spix yellow-toothed cavies (Galea spixii) to the Leishmania infantum chagasi parasite. Nine cavies were experimentally infected, separated into three groups and monitored at 30, 90 and 180 days, respectively. Amastigotes were identified in the spleen slides of two cavies killed 180 days after infection. Antibodies against the L. i. chagasi were identified in one of the cavies. This demonstrates that G. spixii is in fact capable of maintaining a stable infection by L. i. chagasi without alterations in biochemical and hematological parameters of the host and without perceivable micro and macroscopic lesions

    The Clacton Spear: the last one hundred years

    In 1911 an eminent amateur prehistorian pulled the broken end of a pointed wooden shaft from Palaeolithic-age sediments at a seaside town in Essex. This artefact, still the earliest worked wood to be discovered in the world, became known as the Clacton Spear. Over the past 100 years it has variously been interpreted as a projectile weapon, a stave, a digging stick, a snow probe, a lance, a game stake and a prod to ward off rival scavengers. These perspectives have followed academic fashions, as the popular views of early hominins have altered. Since discovery the Clacton spear has also been replicated twice, has undergone physical transformations due to preservation treatments, and has featured in two public exhibitions. Within this article the changing context of the spear, its parallels, and all previous conservation treatments and their impacts are assessed.© 2015 Royal Archaeological Institute.     LINE-1 Evasion of Epigenetic Repression in Humans

    Epigenetic silencing defends against LINE-1 (L1) retrotransposition in mammalian cells. However, the mechanisms that repress young L1 families and how L1 escapes to cause somatic genome mosaicism in the brain remain unclear. Here we report that a conserved Yin Yang 1 (YY1) transcription factor binding site mediates L1 promoter DNA methylation in pluripotent and differentiated cells. By analyzing 24 hippocampal neurons with three distinct single-cell genomic approaches, we characterized and validated a somatic L1 insertion bearing a 3' transduction. The source (donor) L1 for this insertion was slightly 5' truncated, lacked the YY1 binding site, and was highly mobile when tested in\ua0vitro. Locus-specific bisulfite sequencing revealed that the donor L1 and other young L1s with mutated YY1 binding sites were hypomethylated in embryonic stem cells, during neurodifferentiation, and in liver and brain tissue. These results explain how L1 can evade repression and retrotranspose in the human body
