    A broadband study of the 13 August 1978 Santa Barbara earthquake

    The Santa Barbara earthquake of 13 August 1978, provides an opportunity to perform a broadband investigation of body waves for a well-recorded, moderate size (M_L = 5.1) event. The long- and short-period teleseismic body waves are modeled in the time domain to construct a source time function which is consistent in the period range of 1 to 20 sec. The long-period records indicate an overall duration of 6 sec while the short-period records reveal the fine-scale character of the slip history consisting of two sharp pulses separated by about 1 sec. The source mechanism determined from this analysis is a moderately dipping (30Β°NE) thrust with significant left-lateral slip. The moment was determined to be 1.1 Γ— 10^(25) dyne-cm. The earthquake was also reasonably well recorded on accelerographs in the near-field. The modeling of the strong motion displacements was a two step procedure: (1) the displacements were modeled alone, and (2) in an attempt to achieve consistency between the local and far-field time functions, the qualitative features of the teleseismic short-period time function were used to predict the displacements. If the two sources in the short-period time function are allowed to have different mechanisms, the displacements can be modeled quite well. This suggests that the overall faulting process was rough, and the multiple source character suggested at high frequencies is due to high-stress drop asperities. The two sources are modeled as asperities separated by 1.5 km; the first source has a mechanism consistent with the teleseismic solution while the second source is more steeply dipping. The total moment determined from the strong motion data is 3.5 Γ— 10^(24) dyne-cm or one-third the long-period moment. This is consistent with other recent studies which suggest that the high-frequency strong ground motion is controlled by the distribution of asperities even though the sum of their moments may be small compared to the overall moment.     The source mechanism of the August 7, 1966 El Golfo earthquake

    The El Golfo earthquake of August 7, 1966 (m_b = 6.3, M_S = 6.3) occurred near the mouth of the Colorado River at the northern end of the Gulf of California. Synthetic seismograms for this event were computed for both the body waves and the surface waves to determine the source parameters of the earthquake. The body-wave model indicated the source was a right lateral, strike-slip source with a depth of 10 km and a far-field time function 4 sec in duration. The body-wave moment was computed to be 5.0 Γ— 10^(25) dyne-cm. The surface-wave radiation pattern was found to be consistent with that of the body waves with a surface-wave moment of 6.5 Γ— 10^(25) dyne-cm. The agreement of the two different moments indicates that the earthquake had a simple source about 4 sec long. A comparison of this earthquake source with the Borrego Mountain and Truckee events demonstrates that all three of these earthquakes behaved as high stress-drop events. El Golfo was shown to be different from the low stress-drop, plate-boundary events which were located on the Gibbs fracture zone in 1967 and 1974

    Results from a synthetic seismogram analysis of the short-period P waves from the Borrego Mountain earthquake of 9 April 1968 (M_L = 6.4) are used to model the strong-motion recording at El Centro. A short-period-long-period deconvolution analysis of the teleseismic P waves suggested that a two-source model would fit the data much better than the single-source model presented by Burdick and Mellman (1976). Forward and inverse modeling of the data demonstrated that two sources, each of less than 2-sec duration, the second occurring 2.2 sec after the first and both being at about 8-km depth, best fit the short-period waveforms. From this model, long-period synthetics were generated which were found to be quite compatible with the data. This source model was also used to synthesize the strong-motion SH displacement, velocity, and acceleration records from El Centro, California. The close match of synthetics and data is used to argue that short-period waveforms contain much information about asperities which play a crucial role in the near-source strong motions from an earthquake. The Borrego Mountain event probably began with the failure of a fault asperity. The evidence for this is the several-hundred-bars stress drops of the two short-period sources and the probable location of these sources in a place where there was almost no aftershock activity or postseismic creep on the fault

    Isolation of a novel flavanonol and an alkylresorcinol with highly potent anti-trypanosomal activity from Libyan propolis

    Twelve propolis samples from different parts of Libya were investigated for their phytochemical constituents. Ethanol extracts of the samples and some purified compounds were tested against Trypanosoma brucei, Plasmodium falciparum and against two helminth species, Trichinella spiralis and Caenorhabditis elegans, showing various degrees of activity. Fourteen compounds were isolated from the propolis samples, including a novel compound Taxifolin-3-acetyl-4’-methyl ether (4), a flavanonol derivative. The crude extracts showed moderate activity against T. spiralis and C. elegans, while the purified compounds had low activity against P. falciparum. Anti-trypanosomal activity (EC50 = 0.7 Β΅g/mL) was exhibited by a fraction containing a cardol identified as bilobol (10) and this fraction had no effect on Human Foreskin Fibroblasts (HFF), even at 2.0 mg/mL, thus demonstrating excellent selectivity. A metabolomics study was used to explore the mechanism of action of the fraction and it revealed significant disturbances in trypanosomal phospholipid metabolism, especially the formation of choline phospholipids. We conclude that a potent and highly selective new trypanocide may be present in the fraction

    The EHEC Type III Effector NleL Is an E3 Ubiquitin Ligase That Modulates Pedestal Formation

    Enterohemorrhagic Escherichia coli (EHEC) O157:H7 causes hemorrhagic colitis and may result in potentially fatal hemolytic uremia syndrome in humans. EHEC colonize the intestinal mucosa and promote the formation of actin-rich pedestals via translocated type III effectors. Two EHEC type III secreted effectors, Tir and EspFu/TccP, are key players for pedestal formation. We discovered that an EHEC effector protein called Non-LEE-encoded Ligase (NleL) is an E3 ubiquitin ligase. In vitro, we showed that the NleL C753 residue is critical for its E3 ligase activity. Functionally, we demonstrated that NleL E3 ubiquitin ligase activity is involved in modulating Tir-mediated pedestal formation. Surprisingly, EHEC mutant strain deficient in the E3 ligase activity induced more pedestals than the wild-type strain. The canonical EPEC strain E2348/69 normally lacks the nleL gene, and the ectopic expression of the wild-type EHEC nleL, but not the catalytically-deficient nleL(C753A) mutant, in this strain resulted in fewer actin-rich pedestals. Furthermore, we showed that the C. rodentium NleL homolog is a E3 ubiquitin ligase and is required for efficient infection of murine colonic epithelial cells in vivo.     Urinary Incontinence Before and After Bariatric Surgery

    IMPORTANCE: Among women and men with severe obesity, evidence for improvement in urinary incontinence beyond the first year after bariatric surgery–induced weight loss is lacking. OBJECTIVES: To examine change in urinary incontinence before and after bariatric surgery and to identify factors associated with improvement and remission among women and men in the first 3 years after bariatric surgery. DESIGN, SETTING, AND PARTICIPANTS: The Longitudinal Assessment of Bariatric Surgery 2 is an observational cohort study at 10 US hospitals in 6 geographically diverse clinical centers. Participants were recruited between February 21, 2005, and February 17, 2009. Adults undergoing first-time bariatric surgical procedures as part of clinical care by participating surgeons between March 14, 2006, and April 24, 2009, were followed up for 3 years (through October 24, 2012). INTERVENTION: Participants undergoing bariatric surgery completed research assessments before the procedure and annually thereafter. MAIN OUTCOMES AND MEASURES: The frequency and type of urinary incontinence episodes in the past 3 months were assessed using a validated questionnaire. Prevalent urinary incontinence was defined as at least weekly urinary incontinence episodes, and remission was defined as change from prevalent urinary incontinence at baseline to less than weekly urinary incontinence episodes at follow-up. RESULTS: Of 2458 participants, 1987 (80.8%) completed baseline and follow-up assessments. At baseline, the median age was 47 years (age range, 18-78 years), the median body mass index was 46 kg/m(2) (range, 34-94 kg/m(2)), and 1565 of 1987 (78.8%) were women. Urinary incontinence was more prevalent among women (49.3%; 95% CI, 46.9%-51.9%) than men (21.8%; 95% CI, 18.2%-26.1%) (P < .001). After a mean 1-year weight loss of 29.5% (95% CI, 29.0%-30.1%) in women and 27.0% (95% CI, 25.9%-28.6%) in men, year 1 urinary incontinence prevalence was significantly lower among women (18.3%; 95% CI, 16.4%-20.4%) and men (9.8%; 95% CI, 7.2%-13.4%) (P < .001 for all). The 3-year prevalence was higher than the 1-year prevalence for both sexes (24.8%; 95% CI, 21.8%-26.5% among women and 12.2%; 95% CI, 9.0%-16.4% among men) but was substantially lower than baseline (P < .001 for all). Weight loss was independently related to urinary incontinence remission (relative risk, 1.08; 95% CI, 1.06-1.10 in women and 1.07; 95% CI, 1.02-1.13 in men) per 5% weight loss, as were younger age and the absence of a severe walking limitation. CONCLUSIONS AND RELEVANCE: Among women and men with severe obesity, bariatric surgery was associated with substantially reduced urinary incontinence over 3 years. Improvement in urinary incontinence may be an important benefit of bariatric surgery

    Genes Differentially Expressed in Conidia and Hyphae of Aspergillus fumigatus upon Exposure to Human Neutrophils

    Aspergillus fumigatus is the most common etiologic agent of invasive aspergillosis in immunocompromised patients. Several studies have addressed the mechanism involved in host defense but only few have investigated the pathogen's response to attack by the host cells. To our knowledge, this is the first study that investigates the genes differentially expressed in conidia vs hyphae of A. fumigatus in response to neutrophils from healthy donors as well as from those with chronic granulomatous disease (CGD) which are defective in the production of reactive oxygen species.Transcriptional profiles of conidia and hyphae exposed to neutrophils, either from normal donors or from CGD patients, were obtained by using the genome-wide microarray. Upon exposure to either normal or CGD neutrophils, 244 genes were up-regulated in conidia but not in hyphae. Several of these genes are involved in the degradation of fatty acids, peroxisome function and the glyoxylate cycle which suggests that conidia exposed to neutrophils reprogram their metabolism to adjust to the host environment. In addition, the mRNA levels of four genes encoding proteins putatively involved in iron/copper assimilation were found to be higher in conidia and hyphae exposed to normal neutrophils compared to those exposed to CGD neutrophils. Deletants in several of the differentially expressed genes showed phenotypes related to the proposed functions, i.e. deletants of genes involved in fatty acid catabolism showed defective growth on fatty acids and the deletants of iron/copper assimilation showed higher sensitivity to the oxidative agent menadione. None of these deletants, however, showed reduced resistance to neutrophil attack.This work reveals the complex response of the fungus to leukocytes, one of the major host factors involved in antifungal defense, and identifies fungal genes that may be involved in establishing or prolonging infections in humans

    A Multilaboratory Comparison of Calibration Accuracy and the Performance of External References in Analytical Ultracentrifugation

    Analytical ultracentrifugation (AUC) is a first principles based method to determine absolute sedimentation coefficients and buoyant molar masses of macromolecules and their complexes, reporting on their size and shape in free solution. The purpose of this multi-laboratory study was to establish the precision and accuracy of basic data dimensions in AUC and validate previously proposed calibration techniques. Three kits of AUC cell assemblies containing radial and temperature calibration tools and a bovine serum albumin (BSA) reference sample were shared among 67 laboratories, generating 129 comprehensive data sets. These allowed for an assessment of many parameters of instrument performance, including accuracy of the reported scan time after the start of centrifugation, the accuracy of the temperature calibration, and the accuracy of the radial magnification. The range of sedimentation coefficients obtained for BSA monomer in different instruments and using different optical systems was from 3.655 S to 4.949 S, with a mean and standard deviation of (4.304 Β± 0.188) S (4.4%). After the combined application of correction factors derived from the external calibration references for elapsed time, scan velocity, temperature, and radial magnification, the range of s-values was reduced 7-fold with a mean of 4.325 S and a 6-fold reduced standard deviation of Β± 0.030 S (0.7%). In addition, the large data set provided an opportunity to determine the instrument-to-instrument variation of the absolute radial positions reported in the scan files, the precision of photometric or refractometric signal magnitudes, and the precision of the calculated apparent molar mass of BSA monomer and the fraction of BSA dimers. These results highlight the necessity and effectiveness of independent calibration of basic AUC data dimensions for reliable quantitative studies
