    Bioactive proanthocyanidins from the root bark of Cassia abbreviata

    Cassia abbreviata is an important medicinal plant used in the treatment of various infectious diseases. The ethnomedical efficacy of extracts of this plant species is attributed to its phytochemical constituents most of which are phenolics and anthraquinones. The aim of this study was to isolate and elucidate bioactive phenolic compounds from the root bark of this species. Consequently, two novel trimmeric proanthocyanidins; 3,7,4'-trihydroxyflavan-(4β→8)-3,5,7,4'-tetrahydroxyflavan-(3'→6)-3,5,7,2',4'- pentahydroxyflavan (cassinidin A) and 3,7,2',4'-tetrahydroxyflavan-(4α→8)-3,5,7,4'-tetrahydroxyflavan-(4α→6)-3,5,7,2',4'-pentahydroxyflavan (cassinidin B) were isolated from the root bark of Cassia abbreviata. The chemical structures were determined using NMR, MS and HRMS spectroscopic data. The cassinidin A and B showed higher to moderate antimicrobial activity against Escherichia coli, Bacillus subtilis, Staphylococcus aureus and Candida mycoderma.Keywords: Medicinal plant, Cassia abbreviata, Caesalpinioideae, root bark, Cassinidin A, Cassinidin B, antimicrobia

    Antimicrobial and cytotoxic activities of fresh leaf extracts of Warburgia ugandensis

    Abstract: Warburgia ugandensis Sprague (Canellaceae), is widely used by Traditional Health Practitioners in Eastern and Southern Africa for treatment of diseases and conditions associated with HIV/AIDS infections. The most commonly prescribed parts of the plant are usually barks and roots. The aim of this study was to investigate whether leaves of this plant have biological activities comparable to those of barks and roots. Fresh leaves of W. ugandensis were freeze-dried to complete dryness while another portion of the leaves from the same collection were air-dried under shade. Ethanolic extracts from the two differently dried leaves weretested for antimicrobial activity and for brine shrimp toxicity test. Ethanolic extract from the freeze-dried leaves exhibited both antibacterial activity against standard strains of Staphylococcus aureus, Escherichia coli,Vibrio cholerae, Bacillus cereus and antifungal activity against Candida albicans and Cryptococcus neoformans. The extract from the air-dried leaves showed similar antibacterial activity against the mentioned organisms but hadno antifungal activity. However, both extracts exhibited comparable cytotoxic activity (95% CI), against brine shrimp larvae with reference to cyclophosphamide, a standard anticancer drug. Fresh leaves of W. ugandensis have shown to exhibit anticandida activity, a factor which substantiates their application as a traditional herbal remedy to complement treatment of fungal infections in HIV/AIDS patients. Further studies to establish constituents responsible for the exhibited antifungal activity are highly desirable

    An ethnobotanical study of plants used for the treatment of diabetes in the Eastern Cape Province, South Africa

    Diabetes mellitus is one of the commonest diseases affecting the citizens of both developed and poor countries. In South Africa, the number of people suffering from diabetes is believed to be rising steadily. An ethnobotanical study of plants used by the traditional healers, herbalists and rural dwellersfor the treatment of diabetes mellitus was conducted in the Eastern Cape Province. The study revealed 14 plant species belonging to six families namely; Asteraceae, Hypoxidaceae, Apocynaceae, Asphodelaceae, Apiaceae and Buddlejaceae. The use of infusions from plant leaves and roots was thecommonest method of herbal preparation. In all cases, the treatment involved drinking the extracts for a long period of time. There was a general belief on the efficacy of the prepared extracts

    Defining malaria burden from morbidity and mortality records, self treatment practices and serological data in Magugu, Babati District, northern Tanzania

    Malaria morbidity and mortality data from clinical records provide essential information towards defining disease burden in the area and for planning control strategies, but should be augmented with data on transmission intensity and serological data as measures for exposure to malaria. The objective of this study was to estimate the malaria burden based on serological data and prevalence of malaria, and compare it with existing self-treatment practices in Magugu in Babati District of northern Tanzania. Prospectively, 470 individuals were selected for the study. Both microscopy and Rapid Diagnostic Test (RDT) were used for malaria diagnosis. Seroprevalence of antibodies to merozoite surface proteins (MSP-119) and apical membrane antigen (AMA-1) was performed and the entomological inoculation rate (EIR) was estimated. To complement this information, retrospective data on treatment history, prescriptions by physicians and use of bed nets were collected. Malaria prevalence in the area was 6.8% (32/470). Of 130 individuals treated with artemisinin combination therapy (ACT), 22.3 % (29/130) were slide confirmed while 75.3% (98/130) of them were blood smear negative. Three of the slides confirmed individuals were not treated with ACT. Fever was reported in 38.2% of individuals, of whom 48.8 % (88/180) were given ACT.     Antimicrobial, Cytotoxic and Antioxidant Activities and Determination of the Total Tannin Content of Bark Extracts Endopleura uchi

    Endopleura uchi is a typical Amazonian tree and its bark is popularly employed in the preparation of teas against myomas, arthritis, influenza, diarrhea and cancer. In this study, the antioxidant activity, cytotoxicity and antimicrobial activity of five different extracts of the bark, selected by their total tannin content, were assessed. The potential antioxidant activity of the extracts was determined by 2.2-diphenyl-1-picrylhydrazyl radical scavenging assay and the values found were very similar among the extracts and to the standards antioxidants used in the tests. Cytotoxicity analysis in mammalian cells indicated that all the tested extracts exhibited IC50 values higher than the highest concentration used, showing that they do not present a risk when consumed under these conditions. Extract tested against five bacterial strains and one yeast strain did not show satisfactory growth inhibitory activity, and even the extracts that showed some antimicrobial activity were not effective at any dilution to determine the minimum inhibitory concentration. The results may serve as a reference for subsequent works, since such reference values described in the literature for the bark of E. uchi

    The effect of Aloe ferox Mill. in the treatment of loperamide-induced constipation in Wistar rats

    <p>Abstract</p> <p>Background</p> <p>Constipation is the most common gastrointestinal complaint all over the world and it is a risk factor of colorectal cancer. In this study, the efficacy of aqueous leaf extract of <it>Aloe </it><it>ferox </it>Mill. was studied against loperamide-induced constipation in Wistar rats.</p> <p>Methods</p> <p>Constipation was induced by oral administration of loperamide (3 mg/kg body weight) while the control rats received normal saline. The constipated rats were treated with 50, 100 and 200 mg/kg body weight/day of the extract for 7 days during which the feeding characteristics, body weight, fecal properties and gastrointestinal transit ratio were monitored.</p> <p>Results</p> <p>The extract improved intestinal motility, increased fecal volume and normalized body weight in the constipated rats, which are indications of laxative property of the herb with the 200 mg/kg body weight of the extract showing the best efficacy.</p> <p>Conclusion</p> <p>The effect of the extract compares favourably well with senokot, a standard laxative drug. These findings have therefore, lent scientific credence to the folkloric use of the herb as a laxative agent by the people of the Eastern Cape of South Africa.</p

    Drug resistance to sulphadoxine-pyrimethamine in Plasmodium falciparum malaria in Mlimba, Tanzania

    BACKGROUND: Sulphadoxine-pyrimethamine (SP) has been and is currently used for treatment of uncomplicated Plasmodium falciparum malaria in many African countries. Nevertheless, the response of parasites to SP treatment has shown significant variation between individuals. METHODS: The genes for dihydrofolate reductase (dhfr) and dihydropteroate synthase (dhps) were used as markers, to investigate parasite resistance to SP in 141 children aged less than 5 years. Parasite DNA was extracted by Chelex method from blood samples collected and preserved on filter papers. Subsequently, polymerase chain reaction (PCR) and restriction fragment length polymorphism (PCR-RFLP) were applied to detect the SP resistance-associated point mutations on dhfr and dhps. Commonly reported point mutations at codons 51, 59, 108 and 164 in the dhfr and codons 437, 540 and 581 in the dhps domains were examined. RESULTS: Children infected with parasites harbouring a range of single to quintuple dhfr/dhps mutations were erratically cured with SP. However, the quintuple dhfr/dhps mutant genotypes were mostly associated with treatment failures. High proportion of SP resistance-associated point mutations was detected in this study but the adequate clinical response (89.4%) observed clinically at day 14 of follow up reflects the role of semi-immunity protection and parasite clearance in the population. CONCLUSION: In monitoring drug resistance to SP, concurrent studies on possible confounding factors pertaining to development of resistance in falciparum malaria should be considered. The SP resistance potential detected in this study, cautions on its useful therapeutic life as an interim first-line drug against malaria in Tanzania and other malaria-endemic countries

    Protection against Diarrhea Associated with Giardia intestinalis Is Lost with Multi-Nutrient Supplementation: A Study in Tanzanian Children

    Giardia intestinalis is a well-known cause of diarrhea in industrialized countries. In children in developing countries, asymptomatic infections are common and their role as cause of diarrhea has been questioned. In a cohort of rural Tanzanian pre-school children, we assessed the association between the presence of Giardia at baseline and subsequent diarrhea risk. The study was conducted in the context of a randomised trial assessing the effect of supplementation with zinc and other micro-nutrients on malaria, and half of the children daily received a multi-nutrient supplement. Surprisingly, we found that the presence of Giardia at baseline was associated with a substantial reduction in diarrhea risk. Multivariate statistical analysis showed that this protection could not be explained by differences in age or walking distance to the dispensary between children with and without Giardia. Because we cannot exclude that children differed in other (unmeasured) characteristics, we cannot draw firm conclusions about the causality of the observed association, but our findings support the view that the parasite is not an important cause of diarrhea in highly endemic settings. Striking was that the Giardia-associated protection was lost when children received multi-nutrients. Our data do not provide information about the mechanisms involved, but suggest that multi-nutrients may influence the compositionor pathogenicity of intestinal biota
