    Morphological heterogeneity of HeLa cell mitochondria visualized by a modified diaminobenzidine staining technique

    The diaminobenzidine (DAB) technique for the ultrastructural localization of sites of cytochrome c oxidase activity in animal tissues has been adapted to the visualization of mitochondria in animal cells growing in culture. The modified technique allows the staining of mitochondria in all cells in coverslip preparatins for light microscopy. Electron microscopy of thin sections of material treated by this method has revealed that all mitochondrial profiles within a cell (and only these) are stained and they exhibit a well preserved size and internal structure. Coverslip cultures of synchronized and unsynchronized HeLa (F-315) cells stained with the DAB reaction were examined under oil immersion. In the majority of the cells, mitochondria were recognized as discrete bodies in the thinner peripheral portion of the cytoplasm. This observation indicates that in a large proportion of HeLa F-315 cells, at least under the growth conditions used here, the mitochondrial complement is dividied into distinct organelles. This examination also revealed a considerable morphological heterogeneity of mitochondria, which exhibited an ovoid or short rod-like or, less frequently, long filamentous shape, with some evidence of branching. The variability in mitochondrial morphology appeared to be far more prounced between different cells than within individual cells; this cellular heterogeneity was not related in any obvious way to cell-cycle-dependent changes

    Journal ArticlePetroleum-, coal- and shale-derived liquid fuels were burned in a downfired tunnel furnace to assess the impact of fuel properties on the formation and control of NOx emissions. A nitrogen-free oxidant mixture (Ar, C02 , 02) was used to isolate fuel NOx formation. Under excess air conditions fuel NOx correlated well with total fuel nitrogen content for both the petroleum and alternate fuels. Under staged combustion conditions the influence of fuel nitrogen content was much less pronounced but equally highly correlated except in the case of a coal-derived liquid

    Imaging of canine neoplastic reproductive disorders

    Diagnostic imaging plays an essential role in the diagnosis and management of reproductive neoplasia in dogs and cats. The initial diagnosis, staging, and planning of surgical and ra-diation treatment and the response to therapy all involve imaging to varying degrees. Routine ra-diographs, ultrasound, nuclear medicine, and cross‐sectional imaging in the form of computed tomography (CT) and magnetic resonance imaging (MRI) are routinely used in canine reproductive disorders. The choice of imaging modality depends on many factors, including the level of referral and the pathological information required. The biological behaviour of the tumour also guides the choice of imaging in cancer staging, and imaging may play an important role in guiding serial tumour biopsy during the course of therapy. The sophistication of imaging modalities is in-creasing exponentially. Each modality has advantages and disadvantages in terms of cost, availa-bility, sensitivity, specificity, and qualities of anatomic versus functional imaging

    Quantum optical signal processing in diamond

    Controlling the properties of single photons is essential for a wide array of emerging optical quantum technologies spanning quantum sensing, quantum computing, and quantum communications. Essential components for these technologies include single photon sources, quantum memories, waveguides, and detectors. The ideal spectral operating parameters (wavelength and bandwidth) of these components are rarely similar; thus, frequency conversion and spectral control are key enabling steps for component hybridization. Here we perform signal processing of single photons by coherently manipulating their spectra via a modified quantum memory. We store 723.5 nm photons, with 4.1 nm bandwidth, in a room-temperature diamond crystal; upon retrieval we demonstrate centre frequency tunability over 4.2 times the input bandwidth, and bandwidth modulation between 0.5 to 1.9 times the input bandwidth. Our results demonstrate the potential for diamond, and Raman memories in general, to be an integrated platform for photon storage and spectral conversion.Comment: 6 pages, 4 figure

    Storage and retrieval of ultrafast single photons using a room-temperature diamond quantum memory

    We report the storage and retrieval of single photons, via a quantum memory, in the optical phonons of room-temperature bulk diamond. The THz-bandwidth heralded photons are generated by spontaneous parametric downconversion and mapped to phonons via a Raman transition, stored for a variable delay, and released on demand. The second-order correlation of the memory output is g(2)(0)=0.65±0.07g^{(2)}(0) = 0.65 \pm 0.07, demonstrating preservation of non-classical photon statistics throughout storage and retrieval. The memory is low-noise, high-speed and broadly tunable; it therefore promises to be a versatile light-matter interface for local quantum processing applications.Comment: 6 pages, 4 figure

    Storage of polarization-entangled THz-bandwidth photons in a diamond quantum memory

    Bulk diamond phonons have been shown to be a versatile platform for the generation, storage, and manipulation of high-bandwidth quantum states of light. Here we demonstrate a diamond quantum memory that stores, and releases on demand, an arbitrarily polarized ∌\sim250 fs duration photonic qubit. The single-mode nature of the memory is overcome by mapping the two degrees of polarization of the qubit, via Raman transitions, onto two spatially distinct optical phonon modes located in the same diamond crystal. The two modes are coherently recombined upon retrieval and quantum process tomography confirms that the memory faithfully reproduces the input state with average fidelity 0.784±0.0040.784\pm0.004 with a total memory efficiency of (0.76±0.03)%(0.76\pm0.03)\%. In an additional demonstration, one photon of a polarization-entangled pair is stored in the memory. We report that entanglement persists in the retrieved state for up to 1.3 ps of storage time. These results demonstrate that the diamond phonon platform can be used in concert with polarization qubits, a key requirement for polarization-encoded photonic processing

    Veterinary practitioners’ selection of diagnostic tests for the primary evaluation of colic in the horse

    The aim of this study was to survey veterinary practitioners’ selection of diagnostic tests for horses with clinical signs of abdominal pain. A questionnaire was distributed to veterinary surgeons involved in the primary evaluation of horses with abdominal pain, including the respondent's demographics, selection of diagnostic tests and factors affecting decision-making. Data analysis included descriptive analysis, categorisation of free text and simple univariable correlations to explore the relationships between independent variables and the relative self-estimated frequency that diagnostic tests were performed. A total of 228 responses were analysed. Participants worked in mixed practice (55.7 per cent), first opinion equine (22.8 per cent), first and second opinion equine (17.9 per cent) and referral practice (3.1 per cent). The majority (48.2 per cent, 105/218) were very confident managing a colic case (confidence level 4/5). The most frequently used diagnostic tests were ‘response to analgesia’ (87.2±24.0 per cent cases), rectal examination (75.9±21.2 per cent) and nasogastric intubation (43.8±27.6 per cent). Approach varied between practitioners, and for all diagnostic tests with frequency of use ranging from 0 to 100 per cent of cases. ‘Risk to personal safety’ was the most common reason for not using rectal examination. Practitioner's opinion of their confidence level in managing a colic case was associated with how frequently they used different diagnostic tests. There was marked variation in practitioners’ approaches, highlighting the need for further evidence to support decision-making

    Discrimination of bovine milk from non-dairy milk by lipids fingerprinting using routine matrix-assisted laser desorption ionization mass spectrometry

    An important sustainable development goal for any country is to ensure food security by producing a sufficient and safe food supply. This is the case for bovine milk where addition of non-dairy milks such as vegetables (e.g., soya or coconut) has become a common source of adulteration and fraud. Conventionally, gas chromatography techniques are used to detect key lipids (e.g., triacylglycerols) has an effective read-out of assessing milks origins and to detect foreign milks in bovine milks. However, such approach requires several sample preparation steps and a dedicated laboratory environment, precluding a high throughput process. To cope with this need, here, we aimed to develop a novel and simple method without organic solvent extractions for the detection of bovine and non-dairy milks based on lipids fingerprint by routine MALDI-TOF mass spectrometry (MS). The optimized method relies on the simple dilution of milks in water followed by MALDI-TOF MS analyses in the positive linear ion mode and using a matrix consisting of a 9:1 mixture of 2,5-dihydroxybenzoic acid and 2-hydroxy-5-methoxybenzoic acid (super-DHB) solubilized at 10 mg/mL in 70% ethanol. This sensitive, inexpensive, and rapid method has potential for use in food authenticity applications

    Ultrasonographic imaging is an important diagnostic tool because it allows assessment of the shape, size, position, margination and internal architecture of organs, as well as facilitating the study of vascular supply and vascularisation. Recently, there has been considerable development of B-mode, Doppler and contrast-enhanced ultrasonography for examination of the reproductive tract of dogs, both for studying normal physiology and in the clinical setting. This article describes the practical examination of the canine prostate gland and testes using a variety of ultrasound techniques, and details the normal appearance and blood flow of these organs as well as changes that may be observed with common reproductive disorders

    Thinking beyond the hybrid:“actually-existing” cities “after neoliberalism” in Boyle <i>et al.</i>

    In their article, ‘The spatialities of actually existing neoliberalism in Glasgow, 1977 to present’, Mark Boyle, Christopher McWilliams and Gareth Rice (2008) usefully problematise our current understanding of neoliberal urbanism. Our response is aimed at developing a sympathetic but critical approach to Boyle et al's understanding of neoliberal urbanism as illustrated by the Glasgow example. In particular, the counterposing by Boyle et al of a 'hybrid, mutant' model to a 'pure' model of neoliberalism for us misrepresents existing models of neoliberalism as a perfectly finished object rather than a roughly mottled process. That they do not identify any ‘pure’ model leads them to create a straw construct against which they can claim a more sophisticated, refined approach to the messiness of neoliberal urbanism. In contrast, we view neoliberalism as a contested and unstable response to accumulation crises at various scales of analysis
