Commentary: Selecting synaptic partners: GRASPing the role of UNC- 6/netrin
Forming synaptic connections of the appropriate strength between specific neurons is crucial for constructing neural circuits to control behavior. A recent paper in Neural Development describes the use of a synapse-specific label in Caenorhabditis elegans to implicate local UNC-6/netrin signaling in this developmental process. Thus, as well as their well known roles in cell migration and axon guidance, UNC-6/netrin signals distinguish an appropriate synaptic partner from other potential targets
Prostate cancer risk related to foods, food groups, macronutrients and micronutrients derived from the UK Dietary Cohort Consortium food diaries.
BACKGROUND/OBJECTIVES: The influence of dietary factors remains controversial for screen-detected prostate cancer and inconclusive for clinically detected disease. We aimed to examine these associations using prospectively collected food diaries. SUBJECTS/METHODS: A total of 1,717 prostate cancer cases in middle-aged and older UK men were pooled from four prospective cohorts with clinically detected disease (n=663), with routine data follow-up (means 6.6-13.3 years) and a case-control study with screen-detected disease (n=1054), nested in a randomised trial of prostate cancer treatments (ISCTRN 20141297). Multiple-day food diaries (records) completed by men prior to diagnosis were used to estimate intakes of 37 selected nutrients, food groups and items, including carbohydrate, fat, protein, dairy products, fish, meat, fruit and vegetables, energy, fibre, alcohol, lycopene and selenium. Cases were matched on age and diary date to at least one control within study (n=3528). Prostate cancer risk was calculated, using conditional logistic regression (adjusted for baseline covariates) and expressed as odds ratios in each quintile of intake (±95% confidence intervals). Prostate cancer risk was also investigated by localised or advanced stage and by cancer detection method. RESULTS: There were no strong associations between prostate cancer risk and 37 dietary factors. The Mothers and Childrenâs Environmental Health (MOCEH) study
The MOCEH study is a prospective hospital- and community-based cohort study designed to collect information related to environmental exposures (chemical, biological, nutritional, physical, and psychosocial) during pregnancy and childhood and to examine how exposure to environmental pollutants affects growth, development, and disease. The MOCEH network includes one coordinating center, four local centers responsible for recruiting pregnant women, and four evaluation centers (a nutrition center, bio-repository center, neurocognitive development center, and environment assessment center). At the local centers, trained nurses interview the participants to gather information regarding their demographic and socioeconomic characteristics, complications related to the current gestation period, health behaviors and environmental factors. These centers also collect samples of blood, placenta, urine, and breast milk. Environmental hygienists measure each participantâs level of exposure to indoor and outdoor pollutants during the pre- and postnatal periods. The participants are followed up through delivery and until the child is 5Â years of age. The MOCEH study plans to recruit 1,500 pregnant women between 2006 and 2010 and to perform follow-up studies on their children. We expect this study to provide evidence to support the hypothesis that the gestational environment has an effect on the development of diseases during adulthood. We also expect the study results to enable evaluation of latency and age-specific susceptibility to exposure to hazardous environmental pollutants, evaluation of growth retardation focused on environmental and genetic risk factors, selection of target environmental diseases in children, development of an environmental health index, and establishment of a national policy for improving the health of pregnant women and their children
The Genetic Association Between ADHD Symptoms and Reading Difficulties: The Role of Inattentiveness and IQ
Previous studies have documented the primarily genetic aetiology for the stronger phenotypic covariance between reading disability and ADHD inattention symptoms, compared to hyperactivity-impulsivity symptoms. In this study, we examined to what extent this covariation could be attributed to âgeneralist genesâ shared with general cognitive ability or to âspecialistâ genes which may specifically underlie processes linking inattention symptoms and reading difficulties. We used multivariate structural equation modeling on IQ, parent and teacher ADHD ratings and parent ratings on reading difficulties from a general population sample of 1312 twins aged 7.9â10.9Â years. The covariance between reading difficulties and ADHD inattention symptoms was largely driven by genetic (45%) and child-specific environment (21%) factors not shared with IQ and hyperactivity-impulsivity; only 11% of the covariance was due to genetic effects common with IQ. Aetiological influences shared among all phenotypes explained 47% of the variance in reading difficulties. The current study, using a general population sample, extends previous findings by showing, first, that the shared genetic variability between reading difficulties and ADHD inattention symptoms is largely independent from genes contributing to general cognitive ability and, second, that child-specific environment factors, independent from IQ, also contribute to the covariation between reading difficulties and inattention symptoms
Kinetochore fiber formation in animal somatic cells : dueling mechanisms come to a draw

mediated by the formation of a bundle of microtubules (MTs) that tethers the
kinetochore on the chromosome to a spindle pole. The origin of these âkinetochore
fibersâ (K-fibers) has been investigated for over 125 years. As noted in 1944 by
Schrader, there are only three possible ways to form a K-fiber: either it a) grows from
the pole until it contacts the kinetochore; b) grows directly from the kinetochore; or c)
it forms as a result of an interaction between the pole and the chromosome. Since
Schraderâs time it has been firmly established that K-fibers in centrosome-containing
animal somatic cells form as kinetochores capture MTs growing from the spindle pole
(route a). It is now similarly clear that in cells lacking centrosomes, including plants
and many animal oocytes, K-fibers âself-assembleâ from MTs generated by the
chromosomes (route b). Can animal somatic cells form K-fibers in the absence of
centrosomes by the âself-assemblyâ pathway? In 2000 the answer to this question
was shown to be a resounding âyesâ. With this result, the next question became
whether the presence of a centrosome normally suppresses K-fiber self-assembly, or
if this route works concurrently with centrosome-mediated K-fiber formation. This
question, too, has recently been answered: observations on untreated live animal cells
expressing GFP-tagged tubulin clearly show that kinetochores can nucleate the
formation of their associated MTs in the presence of functional centrosomes. The
concurrent operation of these two âduelingâ routes for forming K-fibers in animals
helps explain why the attachment of kinetochores and the maturation of K-fibers
occur as quickly as it does on all chromosomes within a cell.The work is sponsored by

Textural and geochemical constraints on andesitic plug emplacement prior to the 2004â2010 vulcanian explosions at Galeras volcano, Colombia
Hazardous sequences of vulcanian explosions are thought to result from the repeated emplacement and destruction of degassed, highly crystalline magma plugs in the shallow conduit of arc volcanoes. The processes governing the timing and magnitude of these explosions are thought to be related to magma ascent rate and efficiency of degassing and crystallisation. We study a rare suite of time-constrained ballistic bombs from the 2004â2010 period of activity of Galeras volcano to reconstruct magma plug architecture prior to six individual vulcanian explosions. We find that each plug was vertically stratified with respect to crystallinity, vesicularity and melt volatile content, melt composition and viscosity. We interpret this structure as resulting from multiple bubble nucleation events and degassing-driven crystallisation during multi-step ascent of the magma forming the plug, followed by spatially variable crystallisation within the plug under contrasting conditions of effective undercooling created by degassing. We propose that the shallow conduit evolved from more open degassing conditions during 2004â2008 to more closed conditions during 2009â2010. This resulted in explosions becoming smaller and less frequent over time during 2004â2008, then larger and more frequent over time during 2009â2010. This evolution was controlled by changing average ascent rates and is recorded by systematic changes in plagioclase microlite textures. Our results suggest that small volume vulcanian explosions (~â105 m3) should generally be associated with longer repose times (hundreds of days) and produce ballistics characterised by small numbers of large, prismatic plagioclase microlites. Larger volume vulcanian explosions (~â106 m3) should be associated with shorter repose times (tens of days) and produce ballistics characterised by high numbers of small, more tabular plagioclase microlites
