    Experimental results for the evolution of the probability distribution function (PDF) of a scalar mixed by a turbulence flow in a channel are presented. The sequence of PDF from an initial skewed distribution to a sharp Gaussian is found to be non universal. The route toward homogeneization depends on the ratio between the cross sections of the dye injector and the channel. In link with this observation, advantages, shortcomings and applicability of models for the PDF evolution based on a self-convolution mechanisms are discussed.Comment: 4 page

    How does the brain estimate time? This old question has led to many biological and psychological models of time perception (R. A. Block, 1989; P. Fraisse, 1963; J. Gibbon, 1977; D. L. I. Zakay, 1989). Because time cannot be directly measured at a given moment, it has been proposed that the brain estimates time based on the number of changes in an event (S. W. Brown, 1995; P. Fraisse, 1963; W. D. Poynter, 1989). Consistent with this idea, dynamic visual stimuli are known to lengthen perceived time (J. F. Brown, 1931; S. Goldstone & W. T. Lhamon, 1974; W. T. Lhamon & S. Goldstone, 1974, C. O. Z. Roelofs & W. P. C. Zeeman, 1951). However, the kind of information that constitutes the basis for time perception remains unresolved. Here, we show that the temporal frequency of a stimulus serves as the “clock” for perceived duration. Other aspects of changes, such as speed or coherence, were found to be inconsequential. Time dilation saturated at a temporal frequency of 4–8 Hz. These results suggest that the clock governing perceived time has its basis at early processing stages. The possible links between models of time perception and neurophysiological functions of early visual areas are discussed

    Measuring Endocytosis During Proliferative Cell Quiescence

    Quiescence (also called "G0") is the state in which cells have exited the cell cycle but are capable to reenter as required. Though poorly understood, it represents one of the most prevalent cell states across all life. Many biologically important cell types reside in quiescence including mature hepatocytes, endothelial cells, and dormant adult stem cells. Furthermore, the quiescence program occurs in both short- and long-term varieties, depending on the physiological environments. A barrier slowing our understanding of quiescence has been a scarcity of available in vitro model systems to allow for the exploration of key regulatory pathways, such as endocytosis. Endocytosis, the internalization of extracellular material into the cell, is a fundamental and highly regulated process that impacts many cell biological functions. Accordingly, we have developed an in vitro model of deep quiescence in hTERT-immortalized RPE1 cells, combining both long-term contact inhibition and mitogen removal, to measure endocytosis. In addition, we present an analytical approach employing automated high-throughput microscopy and image analysis that yields high-content data allowing for meaningful and statistically robust interpretation. Importantly, the methods presented herein provide a suitable platform that can be easily adapted to investigate other regulatory processes across the cell cycle

    Stem cells are maintained in an undifferentiated state by signals from their microenvironment, the stem cell niche. Despite its central role for organogenesis throughout the plant's life, little is known about how niche development is regulated in the Arabidopsis embryo. Here we show that, in the absence of functional ZWILLE (ZLL), which is a member of the ARGONAUTE (AGO) family, stem cell-specific expression of the signal peptide gene CLAVATA3 (CLV3) is not maintained despite increased levels of the homeodomain transcription factor WUSCHEL (WUS), which is expressed in the organising centre (OC) of the niche and normally promotes stem cell identity. Tissue-specific expression indicates that ZLL acts to maintain the stem cells from the neighbouring vascular primordium, providing direct evidence for a non-cell-autonomous mechanism. Furthermore, mutant and marker gene analyses suggest that during shoot meristem formation, ZLL functions in a similar manner but in a sequential order with its close homologue AGO1, which mediates RNA interference. Thus, WUS-dependent OC signalling to the stem cells is promoted by AGO1 and subsequently maintained by a provascular ZLL-dependent signalling pathway.Matthew R. Tucker, Annika Hinze, Elise J.
